In this study,we provide evidence that oil sands process-affected waters(OSPW)contain factors that activate the antimicrobial and proinflammatory responses of immune cells.Specifically,using the murine macrophage RAW 264.7 cell line,we establish the bioactivity of two different OSPW samples and their isolated fractions.Here,we directly compared the bioactivity of two pilot scale demonstration pit lake(DPL)water samples,which included expressed water from treated tailings(termed the before water capping sample;BWC)as well as an after water capping(AWC)sample consisting of a mixture of expressed water,precipitation,upland runoff,coagulated OSPW and added freshwater.Significant inflammatory(i.***.macrophage activating)bioactivity was associated with the AWC sample and its organic fraction(OF),whereas the BWC sample had reduced bioactivity that was primarily associated with its inorganic fraction(IF).Overall,these results indicate that at non-toxic exposure doses,the RAW 264.7 cell line serves as an acute,sensitive and reliable biosensor for the screening of inflammatory constituents within and among discrete OSPW samples.

many species exhibit color polymorphisms which have distinct physiological and behavioral characteristics.However,the consistency of morph trait covariation patterns across species,time,and ecological contexts remains unclear.This trait covariation is especially relevant in the context of invasion biology and urban adaptation.Specifically,physiological traits pertaining to energy maintenance are crucial to fitness,given their immediate ties to individual reproduction,growth,and population establishment.We investigated the physiological traits of Podarcis muralis,a versatile color polymorphic species that thrives in urban environments(including invasive populations in Ohio,USA).We measured five physiological traits(plasma corticosterone and triglycerides,hematocrit,body condition,and field body temperature),which compose an integrated multivariate phenotype.We then tested variation among co-occurring color morphs in the context of establishment in an urban environment.We found that the traits describing physiological status and strategy shifted across the active season in a morph-dependent manner—the white and yellow morphs exhibited clearly different multivariate physiological phenotypes,characterized primarily by differences in plasma corticosterone.This suggests that morphs have different strategies in physiological regulation,the flexibility of which is crucial to urban adaptation.The white-yellow morph exhibited an intermediate phenotype,suggesting an intermediary energy maintenance strategy.Orange morphs also exhibited distinct phenotypes,but the low prevalence of this morph in our study populations precludes clear interpretation.Our work provides insight into how differences among stable polymorphisms exist across axes of the phenotype and how this variation may aid in establishment within novel environments.

The synthesis of a promising brain imaging agent 4-[F-18]fluoro-4-deoxy-N-acetyl-1,3,6-tri-O-acetylglucosamine, 2, was successfully accomplished from commercially available N-acetyl glucosamine in 5 steps. The non-decay corrected radiochemical yield and purity were found to be 31% ± 4% (n = 3) and >98% respectively. The total reaction time for radio labelling step was 50 min.

males and females have conflicting interests on the frequency and outcomes of mating interactions.males maximize their fitness by mating with as many females as possible,whereas choosy females often reduce receptivity following copulation.Alternative male mating tactics can be adaptive in their expression to a variety of mating contexts,including interactions with a relatively unreceptive mated female.male Rabidosa punctulata wolf spiders can adopt distinctive mating tactics when interacting with a female,a complex courtship display,and/or a more coercive direct mount tactic that often involves grappling with females for copulation.In this study,we set up female mating treatments with initial trials and then paired mated and unmated females with males to observe both female remating frequencies and the male mating tactics used during the interactions.males adopted different mating tactics depending on the mating status of the female they were paired with.males were more likely to adopt a direct mount tactic with already-mated females and courtship with unmated females.Already-mated females were considerably less receptive to males during experimental trials,although they did remate 34%of the time,the majority of which were with males using a direct mount tactic.Whereas males adjusting to these contextual cues were able to gain more copulations,the observation of multiple mating in female R.punctulata introduces the potential for sperm competition.We discuss this sexual conflict in terms of the fitness consequences of these mating outcomes for both males and females.

The current study utilizes a range of diagenetic fingerprints to differentiate between sandstone facies deposited in the Nile Delta before and during the messinian salinity crisis(mSC), which is normally a challenging task considering the complex bio-and lithostratigraphic subdivisions of messinian rock units. Subaerial exposure of the pre-mSC(Qawasim deltaic sandstone), during drawdown of the mediterranean Sea at the time of the mSC, triggered pervasive dissolution of unstable rock fragments, kaolinization of feldspar, and meteoric dolomitization of carbonate. This was followed by mesogenetic calcite cementation and kaolinite transformation into dickite in deeply buried Qawasim sandstone. Comparatively, the Abu madi estuarine facies, deposited during transgression after drawdown related to the mSC,is characterized by eogenetic iron(Fe)-calcite, glauconite, and pyrite(averages of 14.5%, 6%, and 2%,respectively). This facies transition is marked by abundance of mature glauconite(with potassium oxide(K2O) at about 8%) whose content abates upward from the transgression surface. moreover, the compositional variability of the Abu madi sandstone gave rise to multiple diagenetic trajectories that resulted in chlorite formation presumably following smectite and kaolinite. Listed diagenetic variations in the studied messinian sandstone resulted from a complex interplay between rocks’ compositional,depositional, and burial attributes, ultimately serving as a basis for high-resolution stratigraphic correlation in continental and marginal marine settings with poor biostratigraphic controls.

Hepatocyte nuclear factor 4-alpha(HNF4α)is a highly conserved member of nuclear receptor superfamily of ligand-dependent transcription factors that is expressed in liver and gastrointestinal organs(pancreas,stomach,and intestine).In liver,HNF4αis best known for its role as a master regulator of liver-specific gene expression and essential for adult and fetal liver function.Dysregulation of HNF4αexpression has been associated with many human diseases such as ulcerative colitis,colon cancer,maturity-onset diabetes of the young,liver cirrhosis,and hepatocellular carcinoma.However,the precise role of HNF4αin the etiology of these human pathogenesis is not well understood.Limited information is known about the role of HNF4αisoforms in liver and gastrointestinal disease progression.There is,therefore,a critical need to know how disruption of the expression of these isoforms may impact on disease progression and phenotypes.In this review,we will update our current understanding on the role of HNF4αin human liver and gastrointestinal diseases.We further provide additional information on possible use of HNF4αas a target for potential therapeutic approaches.

Acutely,pain serves to protect us from potentially harmful stimuli,however damage to the somatosensory system can cause maladaptive changes in neurons leading to chronic pain.Although acute pain is fairly well controlled,chronic pain remains difficult to treat.Chronic pain is primarily a neuropathic condition,but studies examining the mechanisms underlying chronic pain are now looking beyond afferent nerve lesions and exploring new receptor targets,immune cells,and the role of the autonomic nervous system in contributing chronic pain conditions.The studies outlined in this review reveal how chronic pain is not only confined to alterations in the nervous system and presents findings on new treatment targets and for this debilitating disease.

Aim Portable devices that accurately detect the composition of expired gases and changes in VO_(2)open new possibilities in research methodology and accessibility.Purpose The purpose of this study was to compare the oxygen consumption(VO_(2))measurements of the VO_(2)master Pro(Vm)to the Cosmed K5(K5)during walking,jogging,and running in field and lab conditions.methods Twelve proficient runners,with a current 10 k pace of≥11.29 km/h,performed 3 matched intervals at 3 different speeds(4.82,8.05,11.29 km/h)on a treadmill and on an outdoor track.An airflow test was also performed on both devices by pumping air through the devices using a 3 L syringe timed to a metronome at 15,25,and 35 beats/min.Results The Vm did not report walking data for most participants.During treadmill running,there were significant differences in VO_(2)(47.86±3.94 vs.29.56±4.15 mL/kg/min),ventilation(V_(e))(71 vs.57 mL/min),and tidal volume(TV)(1.89 vs.1.56 L)between the K5 and Vm respectively(Ped significant differences between devices in VO_(2),V_(e),and TV(Pe airflow test showed significant differences between the devices in V_(e)and TV(Pese results suggest that there are significant discrepancies between the K5 and the Vm,likely due to differ-ences in TV measurement.

BACKGROUND In the Spring of 2020,residency programs across the country experienced rapid and drastic changes to their application process as a result of the coronavirus disease 2019(COVID-19)pandemic.In response,residency programs shifted to virtual events and began harnessing social media to communicate with applicants.AIm To analyze the changes in social media usage by orthopaedic surgery programs in response to the COVID-19 pandemic.meTHODS Based on the 2019 residency and fellowship electronic database,accredited US orthopaedic surgery programs were reviewed for social media presence on Instagram and Twitter.Approximately 47000 tweets from 2011-2021 were extracted through the Twitter application programming interface.We extracted:Total number of followers,accounts following,tweets,likes,date of account creation,hashtags,and mentions.Natural language processing was utilized for tweet sentiment analysis and classified as positive,neutral,or negative.Instagram data was collected and deemed current as of August 11,2021.The account foundation date analysis was based on the date recognized as the start of the COVID-19 outbreak in the United States,before or after march 1,2020.ReSULTS A total of 85(42.3%)orthopaedic surgery residency program Twitter handles were identified.Thirty-five(41.2%)programs joined Twitter in the nine months after the 2020 covid outbreak.In 2020,there was a 126.6%increase in volume of tweets by orthopaedic surgery residency accounts as compared to 2019.The median number of followers was 474.5(interquartile range 205.0-796.5).The account with the highest number of tweets was Hospital for Special Surgery(@HSpecialSurgery)with 13776 tweets followed by University of Virginia(@UVA_Ortho)with 5063 and Yale(@OrthoAtYale)with 899.Sentiment analysis before 2020 revealed 30.4%positive,60.8%neutral,and 8.8%negative sentiments across tweets.Interestingly,the positive sentiment percentage increased in 2020 from 30.4%to 34.5%.Of the 201 ACGme-accredited orthopaedic residency programs on Fellowship and Resi
