Isolation and oral immunogenicity assessment of porcine epidemic diarrhea virus NH-TA2020 strain:One of the predominant strains circulating in China from 2017 to 2021

Porcine epidemic diarrhea(PED)caused by porcine epidemic diarrhea virus(PEDV)is one of the most devastating diseases in the global pig industry due to its high mortality rate in piglets.Maternal vaccines can effectively enhance the gut-mammary gland-secretory IgA axis to boost lactogenic immunity and passive protection of nursing piglets against PEDV challenge.From 2017 to 2021,we collected 882 diarrhea samples from 303 farms in China to investigate the epidemiology of PEDV.The result showed that about 52.15%(158/303)of the farms were positive for PEDV with an overall detection rate of 63.95%(564/882)of the samples.The S1 fragments of S gene from 104 strains were sequenced for the phylogenetic analysis.A total of 71 PEDV strains(68.27%)sequenced in this study were clustered into the predominant G2c subgroup,while the newly-defined G2d strains(9.62%)were identified in three provinces of China.The NH-TA2020 strain of G2c subgroup was isolated and cultured,and its infection to piglets caused watery diarrhea within 24 h,indicating its strong pathogenicity.Oral administration of NH-TA2020 strain to pregnant gilts stimulated high levels of IgA antibody in colostrum.The piglets fed by the gilts above were challenged with NH-TA2020 strain or CH-HeB-RY-2020 strain from G2d subgroup,and the clinical symptoms and virus shedding were significantly reduced compared to the mock group.Our findings suggest that G2c subgroup is the predominant branch circulating in China from 2017 to 2021.Oral administration of NH-TA2020 enhances maternal IgA and lactogenic immune responses,which confer protection against the homologous and emerging G2d PEDV strains challenges in neonates.

Analyzing acetylene adsorption of metal-organic frameworks based on machine learning

Metal-organic frameworks(MOFs) containing open metal sites are important materials for acetylene(C_(2)H_(2)) adsorption.However,it is inefficient or even impossible to search suitable MOFs by molecular simulation method in nearly infinite MOFs space.Therefore,machine learning(ML) methods are adopted in the material screening and prediction of high-performance MOFs.In this paper,architecture,chemical and structural features are used to analyze the C_(2)H_(2) adsorption performance of the MOFs.Different ML algorithms are applied to perform classification and regression analysis to the factors affecting material adsorption.By decision tree(DT) algorithm,it is found that only PV,GSA,and Cu-OMS are sufficient to determine the high adsorption of the MOFs.Furthermore,the influence of topology on the performance of MOFs is obtained.Gradient Boosting Decision Tree(GBDT),Support Vector Machine(SVM),and Back Propagation Neural Network(BPNN),are introduced to analyze the quantitative structure-property relationship(QSPR) between C_(2)H_(2) adsorption and the features of MOFs.The prediction of the GBDT model is found to have the highest accuracy,with R~2 as 0.93 and RMSE as 11.58.In addition,the GBDT model is used for feature analysis,and the contribution of each feature to the performance is obtained,which is of great significance for the design and analysis of MOFs.The successful application of ML to MOFs screening greatly reduce the calculation time and provides important reference for the design and synthesis of new MOFs.

Designing energetic covalent organic frameworks for stabilizing high-energy compounds

As an emerging high-energy compound,3-nitro-1,2,4-triazol-5-one(NTO)is used in military explosives and rocket propellants.However,the strong acidic corrosion of NTO,and the high sensitivity and poor thermostability of its salts,severely restrict their practical applications.Therefore,a novel strategy to design and construct energetic covalent organic frameworks(COFs)is proposed in this study.We have successfully prepared a two-dimensional crystalline energetic COF(named ECOF-1)assembled from triaminoguanidine salt,in which NTO anions are trapped in the porous framework via the ionic interaction and hydrogen bonds.The results show that ECOF-1 exhibits superior thermal stability than energetic salt of NTO.It also exhibits insensitivity and excellent heat of detonation of 7,971.71 kJ·kg−1.ECOF-1 greatly inhibits the corrosiveness of NTO.In prospect,energetic COFs are promising as a functional platform to design high-energy and insensitive energetic materials.

High energy and insensitive explosives based on energetic porous aromatic frameworks

The design and synthesis of energetic materials with a compatibility of high energy and insensitivity have always been the research fronts in military and civilian fields.Considering excellent performances of porous organic frameworks and the lack of research in the field of energetic materials,in this study,a new concept named energetic porous aromatic frameworks(EPAFs)is proposed.The strategy of coating high energy explosives such as 2,4,6,8,10,12-hexanitro-2,4,6,8,10,12-hexaazaisowurtzitane(CL-20)and 1,3,5,7-tetranitro-1,3,5,7-tetrazocane(HMX)in the EPAFs by wet-infiltration method has successfully realized the assembly of target energetic composite materials.The results show that the 75 wt.%CL-20@EPAF-1 possesses the safer impact sensitivity of 31.4 J than that of CL-20(4.0 J).Notably,for 75 wt.%CL-20@EPAF-1,in addition to the superior detonation performances of the detonation velocity(8,761 m·s^(-1))and detonation pressure(31.27 GPa),the synergistic effect of the nitrogen-rich EPAFs and the nitramines high energy explosives results in a higher heat of detonation that surpasses the most of pristine high explosives and reported novel energetic materials.In prospect,energetic porous aromatic frameworks could be a promising and inspiring strategy to build high energy insensitive energetic materials.

【目的】利用2株猪轮状病毒(Porcine rotavirus,PoRV)单克隆抗体,研制胶体金检测试纸条,用于PoRV的临床快速检测。【方法】制备40 nm粒径的胶体金溶液标记单克隆抗体4G5,将单克隆抗体5G4和羊抗鼠IgG分别包被在硝酸纤维素膜上,作为检测线和质控线。通过优化胶体金溶液最适pH、标记抗体和检测抗体的最佳使用量,制备PoRV胶体金检测试纸条;对试纸条的灵敏度、敏感性、特异性、重复性和稳定性进行评价。通过检测50份猪粪便样品,计算试纸条与RT-PCR方法的符合率,最后采用试纸条进行1249份临床样品的测定。【结果】经优化后试纸条的最佳参数如下:胶体金溶液最佳pH为8.0,标记抗体4G5和检测抗体5G4最佳使用量分别为14.4μg/mL和1 mg/mL。试纸条能检测出1∶2000稀释的PoRV OSU株(G5型)病毒液,灵敏度为104.5TCID50/mL,且检测PoRV G9、G3和G4型毒株均为阳性。检测猪传染性胃肠炎病毒(Transmissible gastroenteritis virus,TGEV)、猪流行性腹泻病毒(Porcine epidemic diarrhea virus,PEDV)、猪δ冠状病毒(Porcine deltacoronavirus,PDCoV)、猪伪狂犬病病毒(Porcine pseudorabies virus,PRV)和猪圆环病毒2型(Porcine circovirus type 2,PCV2)均为阴性。批内和批间重复性、稳定性良好。检测50份猪粪便样品,与RT-PCR方法的阳性符合率为97.5%(39/40),阴性符合率为100%(10/10),总体符合率为98%(49/50)。检测2022年在河南、山西省猪场收集的临床样品1249份,阳性检出率为5.2%。【结论】本研究制备的PoRV胶体金检测试纸条具有良好的敏感性、特异性和重复性等优点,与RT-PCR方法的符合率较高,可用于规模化猪场PoRV的快速检测。

Prediction of methane storage in covalent organic frameworks using big-data-mining approach

A combination of computational materials screening and machine learning(ML)technique is being adopted as a popular approach to study various materials toward application of interest.In this work,we began with high-throughput molecular simulations to calculate the methane storage(6.5 MPa)and deliverable(6.5-0.58 MPa)capacities of 404,460 covalent organic frameworks(COFs)at 298 K.Then,the full data sets with 23 features were randomly split into training and test sets in a ratio of 20:80,which were applied to evaluate the prediction abilities of several ML algorithms,including gradient boosting decision tree(GBDT),neural network(NN),support vector machine(SVM),random forest(RF)and decision tree(DT).The results indicate that the RF model has the highest prediction accuracy,which was further employed to reduce the dimension of features space and quantitatively analyze the relative importance of each feature value.The binary classification predictors built using the features with the highest influence weight can give a successful identification of top-performing candidates from the test set containing 323,168 COFs with an accuracy exceeding 96%.The deliverable capacities of the identified COFs were found to outperform those reported so far for various adsorbents.The findings may provide a useful guidance for the design and synthesis of new high-performance materials for methane storage application.

Background Accurate knowledge of the spinal structural functions is critical to understand the biomechanical factors that affect spinal pathology. Many studies have investigated the human vertebral motion both in vitro and in vivo. However, determination of in vivo motion of the vertebrae under physiologic loading conditions remains a challenge in biomedical engineering because of the limitations of current technology and the complicated anatomy of the spine. Methods For in vitro validation, a human lumbar specimen was imbedded with steel beads and moved to a known distance by an universal testing machine （UTM）.

Disturbance Observer Design with a Bode’s Ideal Filter for Sigma-Delta Modulators

It is usually difficult to design a high performance Sigma⁃Delta(ΣΔ)modulator due to system noises.In this paper,a disturbance observer(DOB)is utilized to estimate the system noises and eliminate their effects on ΣΔ modulators.The applied DOB is introduced with a Bode's ideal cut⁃off(BICO)filter used for the Q⁃filter.The proposed DOB with the BICO filter used in ΣΔ modulators can achieve better noise⁃shaping ability,resulting from the less phase loss of the BICO filter.Finally,the simulation results show that the proposed BICO filter scheme is a useful additional tool for improving the performance of ΣΔ modulators.

Viability of Two Freeze-dried Strains of Bifidobacterium Preparations at Various Temperatures during Prolonged Storage

Viability of bifidobacteria in freeze-dried probiotic products at various temperatures during prolonged storage was assessed. Bifidobacterium longum and Bifidobacterium infantis were freeze-dried. The freeze-dried preparations were stored at -18 , 4 , and 20 ℃. Cell counts were enumerated using BS agar at 37 ℃ for 48 h under anaerobic conditions at 0, 45 and 120 days. Storage at 20 ℃ showed the greatest decline in the viability of bifidobacteria, whereas that at -18 ℃ showed the least decrease.

近年来,季节性冻土区越冬基坑工程因水平冻胀引起的事故频发,但相关水平冻胀机理的研究成果较少。开展了北京市昌平区的越冬基坑现场监测,分析桩锚支护结构在不同补水和刚度条件下的地温变化、土体水分迁移、支护结构受力状态及支护结构水平位移的演化规律。研究结果表明:温度梯度引起土壤土水势变化,导致土体内部水分的竖向和侧向迁移,从而引起水平冻胀作用,诱发基坑支护桩的水平位移,增大支护结构中的锚杆拉力。此外,开放补水条件显著增强了水平冻胀作用,导致更大的冻胀力和冻胀变形,而刚度较大的支护结构对基坑的约束作用更强,能有效限制基坑水平冻胀变形。研究成果可为寒区工程支挡结构的设计和施工提供借鉴。
