Background:Nasopharyngeal carcinoma(NPC)is one of the most prevalent cancers in Southeast Asia.Sirtuin 2(SIRT2)is a member of the NAD+-dependent deacetylase family and has been shown to play important roles in numerous biological processes.However,Its function in NPC remains uncertain.The primary aim of this study is to clarify the role of SIRT2 in NPC.Methods:In this research,we examined the effect of SIRT2 silencing on NPC cell proliferation and colony formation using vitro NPC cell lines.Co-immunoprecipitation and mass spectrometry was applied to identify SIRT2-interacting proteins in NPC cells.Results:In comparison to nasopharyngeal epithelial NP69 cells,SIRT2 was up-regulated in multiple NPC cell lines,particularly in CNE2 cells.SIRT2 knockdown abrogated CNE2 cell proliferation and colony formation,whereas SIRT2 overexpression promoted HNE1 cell proliferation and colony formation.The SIRT2-interacting proteins were gathered in gene expression and regulation processes including RNA processing and translation.Among the SIRT2-interacting proteins,there were multiple DEAD-box(DDX)family members.Of note,silencing of DDX24 phenocopied the effect of SIRT2 knockdown on NPC growth.Overexpression of DDX24 restored SIRT2-depleted CNE2 cells to proliferative and colony formation.Conclusions:Our study indicates that SIRT2 can interact with DDX24 to enhance NPC growth.The clinical relevance of SIRT2 and DDX24 in NPC warrants further investigation.

Mild Renal Dysfunction and Risk of Thromboembolism and Bleeding in Patients with Atrial Fibrillation:The Chinese Atrial Fibrillation Registry Study

Objective:Previous studies indicated that patients with atrial fibrillation(AF)and moderate-to-severe chronic kidney disease(CKD)are at a higher risk of thromboembolism and bleeding during anticoagulation.Whether mild CKD is associated with an increased risk of thromboembolism and bleeding in AF patients remains unknown.This study aimed to evaluate the impact of mild CKD on thromboembolism and major bleeding among patients with AF.Methods:Baseline serum creatinine was available in 17,559 of 25,512 patients enrolled in the China-AF study between August 2011 and December 2018.After excluding those who underwent AF ablation or with moderate-to-severe CKD,7191 non-valvular AF patients(2059 with mild CKD and 5132 with normal renal function)with regular follow-up for at least 6 months were included.Primary outcomes were the time to the first occurrence of thromboembolic and major bleeding events.Results:Over a mean follow-up of(44.4±23.4)months,639 thromboembolism and 231 major bleeding events occurred.The crude incidence rates of thromboembolism were higher in the mild CKD group than that of the normal renal function group(3.0/100 person-years vs.2.2/100 person-years,P<0.0001),while the crude incidence rates of major bleeding were comparable between the two groups(1.0/100 person-years vs.0.8/100 person-years,P=0.076).After multivariate analyses,mild CKD was not associated with an increased risk of thromboembolism(HR=1.05,95%CI:0.89-1.25,P=0.547)or major bleeding(HR=1.11,95%CI:0.84–1.47,P=0.476).Conclusions:Mild CKD was not an independent risk factor of thromboembolism or major bleeding in patients with AF.

On 20 July 2021,a sudden rainstorm happened in central and northern Henan Province,China,killing at least 302people.This extreme precipitation event incurred substantial socioeconomic impacts and resulted in serious losses.Accurate monitoring of such rainstorm events is crucial.In this study,qualitative and quantitative methods are used to comprehensively evaluate the abilities of 10 high-resolution satellite precipitation products[CMORPH-Raw(Climate Prediction Center morphing technique),CMORPH-RT,PERSIANN-CCS(Precipitation Estimation from Remotely Sensed Information Using Artificial Neural Networks),GPM IMERG-Early(Integrated Multisatellite Retrievals for Global Precipitation Measurement),GPM IMERG-Late,GSMaP-Now(Global Satellite Mapping of Precipitation),GSMaP-NRT,FY-2F,FY-2G,and FY-2H]in capturing this extreme rainstorm event,as well as their performances in monitoring different precipitation intensities.The results show that these satellite precipitation products are able to capture the spatial distributions of the rainstorm(e.g.,its location in central and northern Henan),but all products have underestimated the amount of precipitation in the rainstorm center.With the increase in precipitation intensity,the hit rate decreases,the threat score decreases,and the false alarm rate increases.CMORPH-RT is better at capturing the rainstorm than CMORPH-Raw,and it depictes the rainstorm process well;GPM IMERG-Late is more accurate than GPM IMERG-Early;GSMaP-NRT has performed better than GSMaP-Now;and PERSIANNCCS and FY-2F perform poorly.Among the products,CMORPH-RT performs the best,which has accurately captured the center of the rainstorm,and is also the closest to the station-based observations.In general,the satellite precipitation products that integrate infrared and passive microwave data are found to be better than those that only make use of infrared data.The satellite precipitation retrieval algorithm and the amount of passive microwave data have a relatively greater impact on the accuracy of satellite precip

The effect of beta-lactam antibiotics on shoot induction and plantlet regeneration from cotyledonary nodes was tested using two peanut cultivars.The culture media contained 4 mg/L 6-benzylaminopurine(BAP)as the main growth regulator.Various concentrations(100–600 mg/L)of cefotaxime,carbenicillin,and timentin were applied in the culture media.In all the tested media,there were no significant differences in the shoot induction as compared to the control.However,little phytotoxic effect was observed at higher concentrations of these antibiotics in the shoot elongation media.Under shoot elongation medium,shoots turned brownish and partly died at higher concentrations where shooting rates were not affected by the treatments.In cefotaxime,timentin,and carbenicillin-containing media,levels of antibiotics greater than 400,300,and 200 mg/L,respectively resulted in the brown coloration of plantlets.Moreover,the mean shoot number and shoot weight significantly decreased as their dosage increased.The results indicate that maximum levels of antibiotics have an adverse effect on the growth and development of peanuts.Also,cefotaxime(100–300 mg/L)and timentin(100–300 mg/L)will be sufficient in controlling Agrobacterium growth in the culture media with the least phytotoxic effect on the peanut plants.

Comprehensive Summary Carbon nanodots(CDs)with high photothermal performance are ideal candidates for tumor photothermal therapy(PTT).Herein,we investigated the photothermal performance of CDs synthesized from urea and citric acid via solvothermal method in dimethyl sulfoxide(DMSO)at different temperatures.Photothermal conversion efficiency(PCE)of up to 61.3%was obtained in the CDs synthesized at 150℃(150-CDs),which is much better than the CDs synthesized at 180℃(180-CDs).By analyzing the morphologies,chemical structures,and absorption spectra of these CDs,we found that the photothermal effect of the CDs was due to the lattice vibration of their carbonized cores.

Background:Oral anti-coagulants(OAC)are the intervention for the prevention of stroke,which consistently improve clinical outcomes and survival among patients with atrial fibrillation(AF).The main purpose of this study is to identify problems in OAC utilization among hospitalized patients with AF in China.Methods:Using data from the Improving Care for Cardiovascular Disease in China-Atrial Fibrillation(CCC-AF)registry,guideline-recommended OAC use in eligible patients was assessed.Results:A total of 52,530 patients with non-valvular AF were enrolled from February 2015 to December 2019,of whom 38,203 were at a high risk of stroke,9717 were at a moderate risk,and 4610 were at a low risk.On admission,only 20.0%(6075/30,420)of patients with a diagnosed AF and a high risk of stroke were taking OAC.The use of pre-hospital OAC on admission was associated with a lower risk of new-onset ischemic stroke/transient ischemic attack among the diagnosed AF population(adjusted odds ratio:0.54,95%confidence interval:0.43-0.68;P70%of AF patients were at a high risk of stroke.Although poor performance on guideline-recommended OAC use was found in this study,over time the CCC-AF project has made progress in stroke prevention in the Chinese AF population.Registration:ClinicalTrials.gov,NCT02309398.

Quantitative Trait Loci(QTL)Mapping and Marker Analysis of Fatty Acids in Peanut

Peanut,with high oil content,has been a major oil and food crop globally.The compositions of the fatty acids are the common factors in determining the oil quality.In the present study,an F2 segregated population with 140 individuals derived from the cross of Weihua8(a cultivar)and 12L49(a line with high oleic acid concentration)was used to construct a genetic map and conduct QTL mapping analysis.A total of 103 polymorphic SSR primers were utilized for genotyping the RILs and finally generating the SSR loci.Within the 103 SSR loci,a genetic linkage map,covering a total length of 3592.35 cM of the whole peanut genome,was constructed.Based on the genetic map,sixteen QTLs located on nine linkage groups related to peanut fatty acids were finally identified.Among them,four QTLs were detected associated with various traits simultaneously,which showed genetic stability in relation to fatty acids of peanut.Except for the QTLs for oleic acid,linoleic acid,and linolenic acid,three novel QTLs for arachidic acid and behenic acid were also detected.These QTLs might be helpful for further fine mapping analysis and marker-assisted selection of fatty acids in peanut.

Starch digestion rate and location in the gastrointestinal tract(GIT)are critical for human health.This review aims to present a comprehensive summary on our current understanding of physiological,biochemical,anatomical and geometrical factors of human digestive system that are related to in vivo starch digestibility.It is shown that all digestive compartments including mouth,stomach,small intestine,and large intestine play critical roles in regulating the overall starch digestion process.A proper investigation of starch digestion pattern should thus be based on the consideration of all these compartments.Main biological factors are summarized as oral mastication and salivation,gastric emptying and motility,small intestinal enzymes and motility,large intestinal resistant starch(RS)-microbiota interactions and gut-brain feedback control,as well as glucose adsorption and hormonal feedback control.However,connections among these biological factors in determining starch digestive behaviors remain elusive.This is due to the inherent complexity of human GIT anatomy,motility and biochemical conditions,as well as ethical,financial and technical issues in conducting clinical studies.Much technological and scientific efforts from both clinical studies and in vitro simulation models are required for a better understanding of in vivo starch digestion behaviors.
