A nonenzymatic electrochemical sensor for the detection of hydrogen peroxide in vitro and in vivo fibrosis models

Fibrosis occurs due to the excessive deposition of extracellular matrix caused by cell injury.After various types of tissue injury,the dysregulation of the internal response can eventually lead to the destruction of organ structure and dysfunction.There is increasing evidence that oxidative stress,which is characterized by excessive production of hydrogen peroxide(H_(2)O_(2)),is an important cause of fibrosis.Therefore,we synthesized a biosensitive and efficient electrochemical H_(2)O_(2)sensor based on PtNi nanoparticle-doped N-reduced graphene oxide(PtNi-N-rGO)to detect H_(2)O_(2)released from transforming growth factorβ1(TGFβ1)-induced myofibroblast.In addition,the sensor could easily detect changes in H_(2)O_(2)in the lung and bronchoalveolar lavage fluid(BALF)of mice with pulmonary fibrosis.Furthermore,the sensor could also detect H_(2)O_(2)in activated hepatic stellate cells and the liver of carbon tetrachloride(CCl_(4))-induced liver fibrosis.Moreover,the alterations in H_(2)O_(2)detected by the sensor were consistent with nicotinamide adenine dinucleotide phosphate oxidase 4(NOX4)protein expression and the staining results of pathological sections.Taken together,these results highlight the use of H_(2)O_(2)sensors for the rapid detection of fibrosis and facilitate the rapid evaluation of antifibrotic drug candidates.

Cellular senescence-driven transcriptional reprogramming of the MAFB/NOTCH3 axis activates the PI3K/AKT pathway and promotes osteosarcoma progression

Osteosarcoma is the most common primary malignancy of bones and primarily occurs in adolescents and young adults.However,a second smaller peak of osteosarcoma incidence was reported in the elderly aged more than 60.Elderly patients with osteosarcoma exhibit different characteristics compared to young patients,which usually results in a poor prognosis.The mechanism underlying osteosarcoma development in elderly patients is intriguing and of significant value in clinical applications.Senescent cells can accelerate tumor progression by metabolic reprogramming.Recent research has shown that methylmalonic acid(MMA)was significantly up-regulated in the serum of older individuals and played a central role in the development of aggressive characteristics.We found that the significant accumulation of MMA in elderly patients imparted proliferative potential to osteosarcoma cells.The expression of MAFB was excessively up-regulated in osteosarcoma specimens and was further enhanced in response to MMA accumulation as the patient aged.Specifically,we first confirmed a novel molecular mechanism between cellular senescence and cancer,in which the MMA-driven transcriptional reprogramming of the MAFB-NOTCH3 axis accelerated osteosarcoma progression via the activation of PI3K-AKT pathways.Moreover,the down-regulation of the MAFB-NOTCH3 axis increased the sensitivity and effect of AKT inhibitors in osteosarcoma through significant inhibition of AKT phosphorylation.In conclusion,we confirmed that MAFB is a novel age-dependent biomarker for osteosarcoma,and targeting the MAFB-NOTCH3 axis in combination with AKT inhibition can serve as a novel therapeutic strategy for elderly patients with osteosarcoma in experimental and clinical trials.

Realizing high-performance Na_(3)V_(2)(PO_(4))_(2)O_(2)F cathode for sodium-ion batteries via Nb-doping

Na_(3)V_(2)(PO_(4))_(2)O_(2)F(NVPOF)has received considerable interest as a promising cathode material for sodium-ion batteries because of its high working voltage and good structural/thermal stability.However,the sluggish electrode reaction resulting from its low intrinsic electronic conductivity significantly restricts its electrochemical performance and thus its practical application.Herein,Nb-doped Na_(3)V_(2-x)Nb_(x)(PO_(4))_(2)O_(2)F/graphene(rGO)composites(x=0,0.05,0.1)were prepared using a solvothermal method followed by calcination.Compared to the un-doped NVPOF/r GO,doping V-site with high-valence Nb element(Nb^(5+))(Na_(3)V_(1.95)Nb_(0.05)(PO_(4))_(2)O_(2)F/r GO(NVN05POF/rGO))can result in the generated V4^(+)/V3^(+)mixed-valence,ensuring the lower bandgap and thus the increased intrinsic electronic conductivity.Besides,the expanded lattice space favors the Na^(+)migration.With the structure feature where NVN05POF particles are attached to the rGO sheets,the electrode reaction kinetics is further accelerated owing to the well-constructed electron conductive network.As a consequence,the as-prepared NVN05POF/r GO sample exhibits a high specific capacity of~72 m Ah·g^(-1)at 10C(capacity retention of 65.2%(vs.0.5C))and excellent long-term cycling stability with the capacity fading rate of~0.099%per cycle in 500 cycles at 5C.

The extraction of effective components and an antioxidant activity study of Tulipa edulis

Plant extracts from natural sources are an excellent choice for food additives and natural antioxidants.In this study,the active components of Tulipa edulis were extracted and analysed,and their antioxidant capacity was measured.Then,the crude extract mixture was separated and purified using a Sephadex LH-20 gel,and the antioxidant activity of the purified products was determined.Human umbilical vein endothelial human umbilical vein endothelial cells(HUVEC)cells were treated with 35 mmol/L glucose to construct a model of oxidative stress.Then,the cells were treated with the active component to observe whether the products of T.edulis could have a good protective effect on HUVEC cells induced by glucose.Transcriptome analysis was also performed on HUVEC cells after same treatment to explore the possible mechanism of the component F2 protecting HUVEC cells from oxidative stress induced by high glucose.The results showed that component F2 obtained from T.edulis has strong antioxidant activity.Moreover,F2 can play a strong antioxidant protective role in HUVEC cells.Meanwhile,the gene expression of heme oxygenase 1(HO-1),γ-glutamyl cysteine ligase catalytic subunit(GCLC)and NAD(P)H quinone oxidoreductase-1(NQO1)in HUVEC cells was up-regulated after treated with F2.This study provides reference value for the further development and application of T.edulis and the d evelopment of functional food.

Natural variation in maize gene ZmSBR1 confers seedling resistance to Fusarium verticillioides

Maize seedling blight caused by Fusarium verticillioides is a widely occurring maize disease,but the genetics and mechanisms of resistance are not well understood.In this study,GWAS performed by MLM and 3VmrMLM identified 40 and 20 QTNs,associated with seedling blight resistance.These methods identified 49 and 36 genes,respectively.Functional verification of candidate gene ZmSBR1 identified by both methods showed that the resistance of a mutant line to seedling blight decreased by 0.37 grade points after inoculation with F.verticillioides,compared with the WT.The length of the stem rot lesion caused by F.verticillioides increased by 86%in mutant seedlings,and the relative length of the adult plant stalk rot increased by 35%in mutant plants compared to the wild type after inoculation with Fusarium graminearum.Transcriptome analysis showed that expression of defense-related genes after inoculation was down-regulated in the mutant compared to the wild type,synthesis of secondary metabolites associated with resistance was reduced,and the immune response triggered by PAMP decreased,resulting in decreased resistance of mutant maize seedlings.Candidate gene association analysis showed that most maize inbred lines carried the susceptible haplotype.A functional PCR marker was developed.The results demonstrated that ZmSBR1 conferred resistance to multiple Fusarium diseases at the seedling and adult growth stages and had important application value in breeding.

大学生失眠及其与压力感知的相关关系

目的探讨大学生失眠及其与压力感知的关系。方法采用失眠严重程度指数(ISI)、压力感知量表(PSS-10)对3702名大学生进行整群抽样调查。结果大学生失眠检出率为31.4%,男生的失眠症状表现较女生更严重(t=2.047,P=0.041),家庭经济状况越差的大学生失眠症状表现越严重(F=20.423,P<0.001);失眠和压力感知及危机知觉因子、应对能力知觉因子呈正相关,相关系数分别为0.42、0.38、0.31(P<0.001);高压力感知水平者的失眠得分更高(F=203.030,P<0.001),失眠检出率更高(χ^(2)=359.784,P<0.001),中、重度失眠者的压力感知水平也更高(F=293.569,P<0.001);大学生压力感知类型表现为失控型15.3%、紧张型8.3%、易感型23.0%、放松型53.5%,失控型者的失眠得分更高,其次为紧张型、易感型、放松型者最低(F=185.969,P<0.001)。不同压力感知类型者的失眠检出率分别为57.3%、43.3%、39.3%和18.7%,各组间均存在差异(χ^(2)=368.876,P<0.001)。结论大学生失眠和压力感知之间关系密切,从压力感知水平和压力感知类型两方面来区分失眠的高危人群有利于更好地进行失眠的管理和防治。

Background Freshwater snails of the genera Bulinus spp.,Biomphalaria spp.,and Oncomelania spp.are the main intermediate hosts of human and animal schistosomiasis.Identification of these snails has long been based on mor-phological and/or genomic criteria,which have their limitations.These limitations include a lack of precision for the morphological tool and cost and time for the DNA-based approach.Recently,Matrix-Assisted Laser Desorp-tion/lonization Time-Of-Flight(MALDI-TOF)mass spectrometry,a new tool used which is routinely in clinical microbi-ology,has emerged in the field of malacology for the identification of freshwater snails.This study aimed to evaluate the ability of MALDI-TOF MS to identify Biomphalaria pfeifferi and Bulinus forskali snail populations according to their geographicalorigin.Methods This study was conducted on 101 Bi.pfeifferi and 81 Bu.forskali snails collected in three distinct geo-graphical areas of Senegal(the North-East,South-East and central part of the country),and supplemented with wild and laboratory strains.Specimens which had previously been morphologically described were identified by MALDl-TOF MS[identification log score values(LSV)≥1.7],after an initial blind test using the pre-existing database.After DNA-based identification,new reference spectra of Bi.pfeiferi(n=10)and Bu.forskali(n=5)from the geographical areas were added to the MALDI-TOF spectral database.The final blind test against this updated database was per-formed to assess identification at the geographic source level.Results MALDI-TOF MS correctly identified 92.1%of 101 Bi.pfeifferi snails and 98.8%of 81 Bu.forskali snails.At the final blind test,88%of 166 specimens were correctly identified according to both their species and sampling site,with LSVs ranging from 1.74 to 2.70.The geographical source was adequately identified in 90.1%of 91 Bi.pfeifferi and 85.3%of 75 Bu.forskalii samples.Conclusions Our findings demonstrate that MALDI-TOF MS can identify and differentiate snail populations according to geographic

梅洛-庞蒂的身体现象学解决了身体在胡塞尔和萨特理论中的困境与矛盾,以含混的“现象身体”确立了身体作为主体的立场,并影响了20世纪90年代以索布切克、劳拉·马科斯和詹妮弗·巴克为代表的电影现象学的兴起,进而生发出影像触感的话语。在此基础上进一步阐述了在21世纪“世界影像”的媒介环境中体触影像的两种可能性,分别是以马克·汉森为代表的强调身体介入和选择的“体感”影像,和以德勒兹为代表的强调身体流动、创造和生成的“形象”影像。马克·汉森沿袭梅洛-庞蒂的理论,在当下数字媒介环境中全面探讨了身体与触感的独特审美经验,而德勒兹却打开了另一条“激进”却又十分恰切的肉身感觉之路,使我们得以重新反思体触影像的可能及主体性的问题。

效用可转移的均衡博弈的核心刻画了参与人形成稳定的合作后收益的分配方式.以参与人合作形成前后所得到的最少支付和最大支付为参考点,本文定义了参与人的最小权力向量和最大权力向量,然后将这两个向量的唯一有效妥协定义为一个新的解,称为核心妥协解.本文分析了均衡博弈的核心妥协解与另一个著名妥协解τ-值以及核心之间的关系.基于最小权力优先性和零规范情形最大权力比例性,公理化刻画了均衡博弈的核心妥协解.将最小权力优先性替换为策略均衡不变性,本文给出了核心妥协解的另一个公理化刻画.最后,以机场跑道成本分摊为例,分析了核心妥协解的应用.
