A yeast deletion library was sc.eened based on NADH fluoresc.nc. using a 384-well plate assay to identify a yeast isolate lac.ing a previously identified c.ll surfac. oxidase exhibiting an osc.llatory pattern with a period length of 25 min and resistant to the ENOX1-spec.fic.inhibitor simalikalac.one D (YNOX for yeast-spec.fic.ENOX = ENOX4). The c.NA was c.oned from a yeast over expression library using NADH fluoresc.nc. analyzed by Fast Fourier transform and dec.mposition fits. The objec.ive was to identify and sequenc. an ENOX homologue in Sac.haromyc.s c.revisiae with a 25 min rather than a 24 min period length (YNOX). The finding identified YDR005c.as the yeast ENOX protein with a temperature-independent 25 min period length and insensitive to inhibition by simalikalac.one D. The enc.ded protein was expressed in bac.eria and c.arac.erized. Gel slic.s c.rresponding to 55 kDa and 39 kDa His-tagged proteins exhibited 25 min osc.llatory patterns not inhibited by 1 μM simalikalac.one D for both NADH oxidation and reduc.d c.enzyme Q10 oxidation as well as a protein disulfide-thiol interc.ange ac.ivity whic. alternated with the oxidative ac.ivities. Ac.ivities were phased by low-frequenc. elec.romagnetic.fields but, in c.ntrast, to yeast ENOX1, not by addition of melatonin. The assay in the presenc. of D2O shifted the length of the osc.llatory period from 25 min to 32 min. The YDR005c.deletion mutant c.lls lac.ed the ENOX4 c.oc. output present in the wild type yeast.

Malaria inc.denc. in urban areas has generally been low c.mpared to rural areas but rec.nt data indic.te that urban malaria remains a public.health problem. It is therefore important to understand the fac.ors that promote urban malaria to help formulate future vec.or c.ntrol strategies. This study c.mpared Anopheles gambiae s.l. (A. gambiae s.l.) spec.es c.mposition, distribution and insec.ic.de resistanc. mec.anisms between vegetable and non-vegetable growing areas in Ac.ra Metropolis. Four sites were selec.ed within the c.ty of Ac.ra whic. c.mprised of two vegetable-growing and two non-vegetable growing areas. WHO susc.ptibility tests were c.rried out on adults A. gambiae s.l. reared from larvae c.llec.ed from the sites. Five insec.ic.des were tested and the A. gambiae c.mplex, resistanc. genotypes and enzyme ac.ivities of eac. population were c.arac.erized. All A. gambiae s.l. populations tested were resistant to all the insec.ic.des, but relatively lower mortalities were observed in the vegetable growing areas. The mortality against 0.05% deltamethrin was 2.6% (Opeibea) and 12.5% (Korle-Bu) for the vegetable growing areas and 36.2% (Ac.imota) and 38.9% (Mataheko) in the non-vegetable growing areas. Anopheles gambiae s.s. (95% of Opeibea population) and Anopheles c.luzzii, (98% of Korle-Bu population) were the dominant spec.es in the vegetable growing areas. The voltage-gated sodium c.annel (Vgsc.1014F) frequenc.es of all the populations were similar but the ac.tylc.olinesterase (ac.-1) frequenc.es were signific.ntly lower (p A. gambiae s.l. from Opeibea than from the other areas. The c.ntribution of urban agric.lture in the development of insec.ic.de resistanc. needs to be c.nsidered in the formulation of future vec.or c.ntrol strategies alongside other domestic.usages.

Objec.ives: To evaluate the effec.s of starting therapy with either Truvada (TVD) or c.mbivir (c.V) in c.mbination with Efavirenz (EFV) or a boosted Protease Inhibitor (LPV/r, ATV/r, fAPV/r and SQV/r) on c.inic.l, immunologic.l, and virologic.l parameters in HIV-infec.ed antiretroviral (ARV) naive patients. Methods: Twenty-seven patients were prospec.ively enrolled and rec.ived TVD-EFV, 32 TVD-PI/r, 24 c.V-EFV, and 23 c.V-PI/r. Immunovirologic.l analyses were performed at baseline and after 3, 6, 9 and 12 months after initiation of therapy;a full set of data is available for 51 patients. Results: Median c.4+ c.ll c.unts and HIV-RNA plasma viremia were c.mparable in the four groups of patients (TVD-EFV, TVD-PI/r, c.V-EFV, c.V-PI/r) at baseline. At month 12 HIV RNA plasma viremia was c.s/ml in all patients. Median c.4 c.ll c.unts inc.eases were higher, although not signific.ntly, in c.V-EFV and TVD-EFV patients. Finally, c.8+/c.38+/c.RO+T lymphoc.tes were signific.ntly dec.eased after 12 months of therapy in all patients with, possibly, a faster effec. seen in the EFV groups. c.nc.usions: Immunologic.parameters are similarly affec.ed by different ARV c.mbinations, even if EFV-assoc.ated immunomodulation might be marginally better.

c.mparison of the fatty ac.d c.mposition of the serum phospholipids of c.ntrols, prediabetic. and adults with type 2 diabetes

Objec.ive: Although abnormalities in the fatty ac.d c.mposition of serum and red c.ll membrane phospholipids of patients with type 2 diabetes are well-doc.mented, lac.ing are studies of this issue in prediabetic.individuals. Materials/Methods: For this c.oss-sec.ional study, we rec.uited 180 subjec.s (30 - 80 years), 56 of whom were normal with regard to gluc.se c.ntrol (HbA1c. 6.5%). Serum phospholipids were isolated and analyzed for fatty ac.ds. Results: Most importantly, the fatty ac.d c.mpositions of the c.ntrols and prediabetic.subjec.s were not different for 19 fatty ac.ds. However, the fatty ac.d profile of the phospholipids of the patients with diabetes differed from the other two groups;the 14 to 18-c.rbon saturated fatty ac.ds were dec.eased by 12% - 26% whereas the unsaturated fatty ac.ds 16:1n-7, 18:1n-9, 18:2n-6, 20:3n-6 and 20:4n-6 were inc.eased by 45% - 64%. Of note, the doc.sahexaenoic.ac.d (DHA) status of individuals in all three study groups was remarkably low c.mpared with international values, as indic.ted by DHA proportions in the 1.62% - 2.07% range, and there were no differenc.s between groups. The mean melting point of the phospholipid fatty ac.ds of the diabetic.patients (32.2℃) was signific.ntly lower (p c.subjec.s (38.1℃) and the c.ntrols (39.9℃) whic. were not different from eac. other. c.nc.usion: These observations indic.te that the fatty ac.d c.anges assoc.ated with type 2 diabetes follow the onset of the disease as opposed to being a c.usative fac.or of poor gluc.se c.ntrol and insulin insensitivity.

"菩提伽耶佛像"先于七世纪中叶时,经由高僧玄奘和使者王玄策等人介绍到唐都,其表现是作触地式手印的单体坐佛。这一佛像类型刚传入中国时,被认作是菩提伽耶大菩提寺中圣像的摹本。七世纪末首见"菩提瑞像"的名称,并且在各地广为流传。然而,中国地区的触地印佛像中,很大一部分都装饰有耳环、项链、手镯或王冠,具"装饰佛"或"宝冠佛"的特征。本文认为中国唐代的"触地印装饰佛像",是由"菩提伽耶佛像"与"装饰佛"两种不同的佛教图像融合而成的新图像,并分析它在中国在什么情况下产生、及其后的流传。

香港儿童服用低剂量三聚氰胺后肾脏并发症的现况调查研究

目的调查香港儿童服用小剂量三聚氰胺后的肾脏并发症。设计横断面调查方法。机构香港专业评估中心。调查对象根据全港初级医疗保健中心的记录，研究包括3170例（1748例男童，1422例女童）≤12岁、连续1个月或以上服用被三聚氰胺污染奶制品的儿童。主要评估指标是否存在肾结石和血尿。结果 1例儿童存在肾结石，7例儿童肾脏内可疑存在三聚氰胺结晶，应用尿试纸检查发现208例（6．6％）儿童存在血尿。这些儿童在专业评估中心进一步检查后，仅有7．4％的儿童确实存在显微镜下血尿，所以总体儿童血尿出现率低于1％。结论：在曾服用小剂量三聚氰胺的儿童中，没有急性肾功能衰竭或尿路梗阻等严重不良事件发生。我们的研究结果与全港调查一致。即使将7例肾内可疑存在三聚氰胺结晶的患儿包括在内，应用超声检查发现的可疑三聚氰胺相关不良事件发生率也仅为0．2％，且这些患儿尚不需要进一步治疗。应用尿试纸检测是否存在血尿，可能会过高估计血尿的发生率。研究结果提示对于曾服用小剂量三聚氰胺的儿童进行大范围、积极的检查可能并不会得到更多有意义的资料，所以不值得投入更多的经费去进行这种检查。

The wealth of DNA data generated by the human genome projec. c.upling with rec.ntly invented high-throughput gene expression profiling tec.niques has dramatic.lly sped up the proc.ss for biomedic.l researc.ers on eluc.dating the role of genes in human diseases. One powerful method to reveal insight into gene func.ions is the systematic.analysis of gene expression.
