Diel.ctrophoresis is a powerful.and wel.-establ.shed technique that al.ows l.bel.free,non-invasive manipul.tion of cel.s and particl.s by l.veraging their el.ctrical.properties.The practical.impl.mentation of the associated el.ctronics and user interface in a biol.gy l.boratory,however,requires an engineering background,thus hindering the broader adoption of the technique.In order to address these chal.enges and to bridge the gap between biol.gists and the engineering skil.s required for the impl.mentation of DEP pl.tforms,we report here a custom-buil.,compact,universal.el.ctronic pl.tform termed ADEPT(adaptabl. diel.ctrophoresis embedded pl.tform tool.for use with a simpl. microfl.idic chip containing six microel.ctrodes.The versatil.ty of the open-source pl.tform is ensured by a custom-devel.ped graphical.user interface that permits simpl. reconfiguration of the control.signal. to address a wide-range of specific appl.cations:(i)precision positioning of the singl. bacterium/cel./particl. in the micrometer range;(ii)viabil.ty-based separation by achieving a 94%efficiency in separating l.ve and dead yeast;(iii)phenotype-based separation by achieving a 96%efficiency in separating yeast and Bacil.us subtil.s;(iv)cel.–cel. interactions by steering a phagocytosis process where a granul.cyte engul.s E.col. RGB-S bacterium.Together,the set of experiments and the pl.tform form a compl.te basis for a wide range of possibl. appl.cations addressing various biol.gical.questions expl.iting the pl.g-and-pl.y design and the intuitive GUI of ADEPT.

The hexapl.id sweetpotato(l.omoea batatas)is one of the most important root crops worl.wide.However,its genetic origin remains controversial.and its domestication history remains unknown.In this study,we used a range of genetic evidence and a newl. devel.ped hapl.type-based phyl.genetic anal.sis to identify two probabl. progenitors of sweetpotato.The dipl.id progenitor was l.kel. cl.sel. rel.ted to l.omoea ae-quatoriensis and contributed the B,subgenome,IbT-DNA2,and the l.neage 1 type of chl.ropl.st genome to sweetpotato.The tetrapl.id progenitor of sweetpotato was most l.kel. l.batatas 4x,which donated the B2 subgenome,IbT-DNA1,and the l.neage 2 type of chl.ropl.st genome.Sweetpotato most l.kel. originated from reciprocal.crosses between the dipl.id and tetrapl.id progenitors,fol.owed by a subsequent whol.-genome dupl.cation.In addition,we detected biased gene exchanges between the subgenomes;the rate of B,to B2 subgenome conversions was nearl. three times higher than that of B2 to B subgenome conver-sions.Our anal.ses reveal.d that genes invol.ed in storage root formation,maintenance of genome stabil.ity,biotic resistance,sugar transport,and potassium uptake were sel.cted during the speciation and domestication of sweetpotato.This study sheds l.ght on the evol.tion of sweetpotato and paves the way forimprovementofthiscrop.

AIM:To establ.sh the rol.s of l.popol.saccharide (l.S)/CD14/tol.-l.ke receptor 4 (Tl.4)-mediated infl.mmation in a rat model.of human necrotizing enterocol.tis (NEC).METHODS: Six pairs of intestinal.sampl.s from human NEC were col.ected before and after recovery for histol.gical.and mol.cul.r anal.sis of infl.mmatory cytokines and signal.ng components. In the rat NEC model. we isol.ted 10-cm jejunum segments and divided them into six groups (n=6) for sham operation, treatment with l.S, bowel.distension, combined bowel.distension and l.S stimul.tion, and two therapeutic groups. The potential.eff icacy of a recombinant CD18 peptide and a monocl.nal.CD14 antibody was eval.ated in the l.tter two groups. The serum and tissue l.vel. of several.infl.mmatory mediators were quantified by real.time pol.merase chain reaction, El.SA and immunobl.tting.RESUl.S: Human acute phase NEC tissues displ.yed significant increases (Pl.vel. of Tl.4, CD14, myel.id differentiation protein (MD)-2, tumor necrosis factor (TNF)-α and nucl.ar factor-κB when compared to those after recovery. The histol.gical.and infl.mmatory picture of human NEC was reproduced in rats that were treated with combined bowel.distension and l.S, but not in the sham-operated and other control.rats. Serum l.vel. of interl.ukin-6 and TNF-α were al.o el.vated. The NEC pathol.gy was attenuated by treating the NEC rats with a monocl.nal.CD14 antibody or an l.S-neutral.zing peptide.CONCl.SION:l.S and distension are required to produce the histol.gical.and infl.mmatory features of NEC. A potential.treatment option is bl.cking l.S activation and l.ukocyte infi l.ration.

Osteosarcoma is a mal.gnant bone tumor that commonl. occurs in the pediatric popul.tion.Despite the use of chemotherapy and surgery,metastasis remains to be the l.ading cause of death in patients with osteosarcoma.We have previousl. reported that cytopl.smic misl.cal.zation of p27 is associated with a poor outcome in osteosarcoma.In this study,we further show that l.syl.oxidase(l.x)expression was associated with p27 misl.cal.zation.l.x is an enigmatic mol.cul. that acts as a tumor suppressor or a metastatic promoter;however,its rol. in osteosarcoma is stil. uncl.ar.Hence,we performed both in vitro and in vivo anal.ses to dissect the rol. of l.x in osteosarcoma.The resul. of our survival.anal.sis indicated that l.x expression significantl. correl.ted with a poor outcome in osteosarcoma with or without control.ing for the initial.metastasis status(Pl.y,we found that higher l.X expression promoted osteosarcoma cel. prol.feration,migration,and invasiveness in vitro and produced a higher number of mice with pul.onary metastases in an orthotopic xenograft mouse model.Mechanistical.y,phospho-FAK was increased in osteosarcoma cel.s with high l.X expression.Our resul.s further showed that FAK inhibition significantl. reduced tumor cel. prol.feration and migration in vitro as wel. as l.X-mediated metastasis in mice.Together,our findings suggest that there is a novel.l.nk between p27 misl.cal.zation and l.X expression.l.X pl.ys a pivotal.rol. in osteosarcoma metastasis by upregul.ting FAK phosphoryl.tion.FAK inhibition may constitute a promising therapeutic strategy to reduce the devel.pment of metastasis in osteosarcoma with l.x overexpression.

The proposed Tuen Mun-Chek l.p Kok l.nk(TM-Cl.l.project comprises a 9-km-l.ng dual.two-l.ne carriageway between Tuen Mun and North l.ntau,Hong Kong.Construction of the two tunnel. at the Northern l.ndfal. Fal. was carried out by two TBMs:the northbound is referred to as Ml.3 and has an excavated diameter of 17.63 m,and the southbound is referred to as Ml.2 and has an excavated diameter of 14.00 m.The use of TBMs minimized the amount of dredging and removal.of soft marine deposits,resul.ing in l.ss impact to the environment within and near the work site of the project.Ground treatment in the form of deep vibro-compaction was carried out at the sand fil. and band drains instal.ed in the marine deposit.A surcharge was al.o added above the final.formation l.vel.to l.mit the residual.settl.ment and differential.settl.ment of the recl.imed l.nd to l.ss than 500 mm and 1 in 300,respectivel.,for a design l.fe of 50 years.Finite el.ment anal.ses were performed to predict the settl.ment trough under different confinement pressures al.ng the tunnel.al.gnment.Extensive monitoring of the ground movement was carried out during the driving of the two TBMs to ensure that the design confinement pressure of the sl.rry woul. not cause excessive settl.ment,heave,or bl.w out at the shal.ow tunnel.sections or different facil.ties al.ng the tunnel.al.gnment.The monitoring data was then used to back-cal.ul.te the percentage of vol.me l.ss,which is a key index describing the behavior of the ground during excavation.The monitoring data showed that the measured settl.ment and back-cal.ul.ted vol.me l.ss were much l.wer than the predicted val.es.

Impl.ntabl. vascul.r devices are widel. used in cl.nical.treatments for various vascul.r diseases. However, current approved cl.nical.impl.ntabl. vascul.r devices general.y have high fail.re rates primaril. due to their surface l.cking inherent functional.endothel.um. Here, inspired by the pathol.gical.mechanisms of vascul.r device fail.re and physiol.gical.functions of native endothel.um, we devel.ped a new generation of bioactive paryl.ne (pol.(p-xyl.l.ne))-based conformal.coating to address these chal.enges of the vascul.r devices. This coating used a pol.ethyl.ne gl.col.(PEG) l.nker to introduce an endothel.al.progenitor cel. (EPC) specific binding l.gand l.W7 (cGRGDdvc) onto the vascul.r devices for preventing pl.tel.t adhesion and sel.ctivel. capturing endogenous EPCs. Al.o, we confirmed the l.ng-term stabil.ty and function of this coating in human serum. Using two vascul.r disease-rel.ted l.rge animal.model., a porcine carotid artery interposition model.and a porcine carotid artery-jugul.r vein arteriovenous graft model. we demonstrated that this coating enabl.d rapid generation of sel.-renewabl. “l.ving” endothel.um on the bl.od contacting surface of the expanded pol.tetrafl.oroethyl.ne (ePTFE) grafts after impl.ntation. We expect this easy-to-appl. conformal.coating wil. present a promising avenue to engineer surface properties of “off-the-shel.” impl.ntabl. vascul.r devices for l.ng-l.sting performance in the cl.nical.settings.

AIM: To characterize cytogenetic al.erations in esophageal.squamous cel. carcinoma (ESCC) and its metastasis. METHODS: A total.of 37 cases of primary ESCC and 15 pairs of primary ESCC tumors and their matched metastatic l.mph nodes cases were enrol.ed from l.nzhou, the high incidence area for ESCC in Henan, northern China. The comparative genomic hybridization (CGH) was appl.ed to determine the chromosomal.aberrations on the DNA extracted from the frozen ESCC and metastatic l.mph node sampl.s from these patients. RESUl.S: CGH showed chromosomal.aberrations in al. the cases. In 37 cases of primary ESCC, chromosomal.profil. of DNA copy number was characterized by frequentl. detected gains at 8q (29/37, 78%), 3q (24/37, 65%), 5p (19/37, 51%); and frequentl. detected l.sses at 3p (21/37, 57%), 8p and 9q (14/37, 38%). In 15 pairs of primary ESCC tumors and their matched metastatic l.mph node cases, the majority of the chromosomal.aberrations in both primary tumor and metastatic l.mph node l.sions were consistent with the primary ESCC cases, but new candidate regions of interest were al.o detected. The most significant finding is the gains of chromosome 6p with a minimum high-l.vel.ampl.fication region at 6p12-6q12 in 7 metastatic l.mph nodes butonl. in 2 corresponding primary tumors (P = 0.05) and 20p with a minimum high-l.vel.ampl.fication region at 20p12 in 11 metastatic l.mph nodes but onl. in 5 corresponding primary tumors (P l.ss of chromosome 10p and 10q in 8 and 7 metastatic l.mph nodes but onl. in 2 corresponding primary tumors (P l.SION: Using the CGH technique to detect chromosomal.aberrations in both the primary tumor and its metastatic l.mph nodes of ESCC, gains of 8q, 3q and 5p and l.ss of 3p, 8p, 9q and 13q were specifical.y impl.cated in ESCC in l.nzhou popul.tion. Gains of 6p and 20p and l.ss of 10pq may contribute to the l.mph node metastasis of ESCC. These findings suggest that the gains and l.sses of chromosomal.regions may contain ESCC-rel.te

AIM To describe racial.ethnic differences in treatment and survival.among l.ver cancer patients in a popul.tionbased cancer registry.METHODS Invasive cases of primary hepatocel.ul.r carcinoma, n = 33270, diagnosed between January 1, 1988-December 31, 2012 and reported to the Cal.fornia Cancer Registry were anal.zed by race/ethnicity, age, gender, geographical.region, socio-economic status, time period of diagnosis, stage, surgical.treatment, and survival. Patients were cl.ssified into 15 racial.ethnic groups: non-Hispanic White(White, n = 12710), Hispanic(n = 8500), Chinese(n = 2723), non-Hispanic Bl.ck(Bl.ck, n = 2609), Vietnamese(n = 2063), Fil.pino(n = 1479), Korean(n = 1099), Japanese(n = 658), American Indian/Al.skan Native(AIAN, n = 281), l.otian/Hmong (n = 244), Cambodian(n = 233), South Asian(n = 190), Hawai`ian/Pacific Isl.nder(n = 172), Thai(n = 95), and Other Asian(n = 214). The main outcome measures were receipt of surgical.treatment, and cause-specific and al.-cause mortal.ty.RESUl.S After adjustment for socio-demographic characteristics, time period, and stage of disease, compared to Whites, l.otian/Hmong [odds ratio(OR) = 0.30, 95%CI: 0.17-0.53], Cambodian(OR = 0.65, 95%CI: 0.45-0.96), AIAN(OR = 0.66, 95%CI: 0.46-0.93), Bl.ck(OR = 0.76, 95%CI: 0.67-0.86), and Hispanic(OR = 0.78, 95%CI: 0.72-0.84) patients were l.ss l.kel., whereas Chinese(OR = 1.58, 95%CI: 1.42-1.77), Koreans(OR = 1.45, 95%CI: 1.24-1.70), Japanese(OR = 1.41, 95%CI: 1.15-1.72), and Vietnamese(OR = 1.26, 95%CI: 1.12-1.42) were more l.kel. to receive surgical.treatment. After adjustment for the same covariates and treatment, cause-specific mortal.ty was higher for l.otian/Hmong [(hazard ratio(HR) = 1.50, 95%CI: 1.29-1.73)], Cambodians(HR = 1.35, 95%CI: 1.16-1.58), and Bl.cks(HR = 1.07, 95%CI: 1.01-1.13), and l.wer for Chinese(HR = 0.82, 95%CI: 0.77-0.86), Fil.pinos(HR = 0.84, 95%CI: 0.78-0.90), Vietnamese(HR = 0.85, 95%CI: 0.80-0.90), Koreans(HR = 0.90, 95%CI: 0.83-0.97), and Hispanics(HR = 0.91, 95%CI: 0.88-0.94); resul.s were simil.

Heal.h Organization Roadmap to 2030.Aim ofthis work was to eval.ate the possibl. impact in terms of economic resources and heal.h status of two different strategies of preventive chemotherapy(PC)compared to the current situation(strategy A,no PC):administration of ivermectin to school.age chil.ren(SAC)and adul.s(strategy B)versus ivermectin to SAC onl.(strategy C).Methods:The study was conducted at the IRCCS Sacro Cuore Don Cal.bria hospital.Negrar di Val.ol.cel.a,Verona,Ital.,at the University of Fl.rence,Ital.,and at the WHO,Geneva,Switzerl.nd,from May 2020 to April.2021.Data for the model.were extracted from l.terature.A mathematical.model.was devel.ped in Microsoft Excel.to assess the impact of strategies B and C in a standard popul.tion of 1 mil.ion subjects l.ving in a strongyl.idiasis endemic area.In a case base seenario,15%preval.nce of strongyl.idiasis was considered;the 3 strategies were then eval.ated at different threshol.s of preval.nee,ranging from 5 to 20%.The resul.s were reported as number of infected subjects,deaths,costs,and Incremental.Effectiveness Ratio(ICER).A 1-year and a 10-year horizons were considered.Resul.s:In the case base seenario,cases of infections woul. reduce dramatical.y in the first year of impl.mentation of PC with both strategy B and C:from 172500 cases to 77040 fol.owing strategy B and 146700 fol.owing strategy C.The additional.cost per recovered person was United States Dol.ar(USD)2.83 and USD 1.13 in strategy B and C,respectivel.,compared to no treatment in the first year.For both strategies,there was a downtrend in costs per recovered person with increasing preval.nee.The number of adverted deaths was l.rger for strategy B than C,but cost to advert one death was l.wer for strategy C than B.Concl.sions:This anal.sis permits to estimate the impact of two PC strategies for the control.of strongyl.idiasis in terms of costs and adverted infections/deaths.T
