Healing of the nasal septal mucosa in an experimental rabbit model of mucosal injury

UNASSIGNED:The aim of this study was to investigate the regeneration process of the nasal mucosa after a surgically created mucosal defect in the rabbit nasal septum, and to evaluate the effects of different interventions.;UNASSIGNED:A 7 mm-diameter circular mucosal defect was made in the septum of forty New Zealand white rabbits. The rabbits were divided into four groups (ten rabbits in each group) according to the type of intervention; no treatment (control), silastic sheet (SS), hyaluronic acid (HA), and silastic sheet and hyaluronic acid (SS + HA) group. The diameter of the defect, mucosal thickness, epithelial thickness, and ciliated cell count were evaluated every week for five weeks.;UNASSIGNED:The average diameter of the defect in the control group were 5.1, 3.65, 1.2, 0.75, and 0.05 mm at postoperative 1, 2, 3, 4, and 5 weeks. In the SS group, the diameter decreased to 4.35, 2.1, 0.35, 0.15, and 0 mm at postoperative 1, 2, 3, 4, and 5 weeks, respectively, in which the mean diameter of the postoperative week 2 was significantly smaller compared to control (3.65 mm vs. 2.1 mm, = 0.039). For the HA group and SS + HA group, the diameter of the defect did not show a significant difference from the control group during the five weeks. The mucosal thickness, epithelial thickness, and ciliated cell count of the regenerated mucosa were not significantly different among the groups.;UNASSIGNED:The regeneration process of the nasal septal mucosa was identified using a novel rabbit model. Mucosal regeneration can be accelerated by applying silastic sheets.

Background No study has thoroughly compared the effectiveness of combined antiplatelet treatments(other than clopidogrel–aspirin)versus clopidogrel–aspirin or aspirin alone for early secondary prevention in acute ischaemic *** We identified patients with acute,minor,non-cardiogenic ischaemic stroke treated with aspirin alone,clopidogrel–aspirin or other combination *** scores considering the inverse probability of treatment weighting were used to adjust for baseline *** primary outcome was the composite of all strokes(ischaemic or haemorrhagic),myocardial infarction and all-cause mortality at 3 *** Among 12234 patients(male:61.9%;age:65.5±13 years)who met the eligibility criteria,aspirin,clopidogrel–aspirin and other combination treatments were administered in 52.2%,42.9%and 4.9%of patients,*** the crude analysis,the primary outcome event at 3 months occurred in 14.5%of the other combination group,14.4%of the aspirin group and 13.0%of the clopidogrel–aspirin *** the weighted Cox proportional hazards analysis,the 3-month primary outcome event occurred less frequently in the clopidogrel–aspirin group than in the other combination group(weighted HR:0.82(0.59–1.13)),while no association was found between the aspirin group(weighted HR:1.04(0.76–1.44))or other combination group and the 3-month primary *** Other combined antiplatelet treatment,compared with aspirin alone or clopidogrel–aspirin,was not associated with reduced risks of primary composite vascular events or recurrent stroke during the first 3 months after ***,the results suggest that other combination treatments,particularly the cilostazol-based combination,may not be effective alternatives for clopidogrel–aspirin to prevent early vascular events in patients with acute minor *** exploration in clinical trials will be needed.

Antiproliferative Effects of Zinc-Citrate Compound on Hormone Refractory Prostate Cancer

Objective： To investigate the antiproliferative effects of zinc-citrate compound on hormone refractory prostate cancer （HRPC）. Methods： HRPC cell line （DU145） and normal prostate cell line （RWPE-1） were treated with zinc, citrate and zinc-citrate compound at different time intervals and concentrations to investigate the effect of zinc-citrate compound. Mitochondrial （m）-aconitase activity was determined using aconitase assay. DNA laddering analysis was performed to investigate apoptosis of DU145 cells. Molecular mechanism of apoptosis was investigated by Western blot analysis of P53, P21w^fl, Bcl-2, Bcl-xL and Bax, and also caspase-3 activity analysis. Results： Treatment with zinc-citrate compound resulted in a time- and dose-dependent decrease in cell number of DU145 cells in comparison with RWPE-1. M-aconitase activity was significantly decreased. DNA laddering analysis indicated apoptosis of DU145 cells. Zinc-citrate compound increased the expression of P21wall and P53, and reduced the expression of Bcl-2 and Bcl-xL proteins but induced the expression of Bax protein. Zinc-citrate compound induced apoptosis of DU145 cells by activation of the caspase-3 pathway. Conclusion： Zinc-citrate compound can induce apoptotic cell death in DU145, by caspase-3 activation through up-regulation of apoptotic proteins and down-regulation of antiapoptotic proteins.

Efficacy and safety of tadalafil taken as needed for the treatment of erectile dysfunction in Asian men： results of an integrated analysis

We evaluated the efficacy and safety of as-needed tadalafil in a diverse clinical population （with varying patient demographics, disease severity, and comorbid medical conditions） of Asian men with erectile dysfunction （ED）. An integrated analysis of five double-blind, placebo-controlled trials （N = 1 046） was performed. Patients were randomly assigned to receive 10 mg tadalafil （N = 185）, 20 mg tadalafil （N = 510）, or placebo （N = 351）. Efficacy assessments included the International Index of Erectile Function （IIEF）, Sexual Encounter Profile （SEP） diary and Global Assessment Question （GAQ）. Patients receiving 10 mg or 20 mg tadalafil showed significant improvement from baseline-to-end point on the Erectile Function domain of the International Index of Erectile Function （IIEF-EF） domain score in all clinical sub-populations analyzed, compared with patients receiving placebo （P 〈 0.001）. The 10-mg and 20-mg tadalafil groups showed a mean success rate of 64.1% and 70.5% for sexual intercourse attempts （SEP3, successful intercourse）, respectively, compared with 33.4% in the placebo group （P 〈 0.001）, and 85.5% and 85.4% reported improved erections at end point GAQ, respectively, versus 43.5% in the placebo group （P 〈 0.001）. Tadalafil was well tolerated across all groups studied. Headache and back pain were the most frequently reported adverse events. Overall, tadalafil was effective and well tolerated across a diverse clinical spectrum of Asian men with ED.

Piglet gut microbial shifts early in life:causes and effects

The gut microbiome has long been known to play fundamentally important roles in the animal health and the well-being of its host. As such, the establishment and maintenance of a beneficial gut microbiota early in life is crucial in pigs, since early gut colonizers are pivotal in the establishment of permanent microbial community structures affecting the health and growth performance of pigs later in life. Emphasizing this importance of early gut colonizers, it is critical to understand the factors impacting the establishment of the piglet gut microbiome at weaning. Factors include, among others, diet, in-feed antibiotics, probiotics and prebiotic administration. The impact of these factors on establishment of the gut microbiome of piglets at weaning includes effects on piglet gut microbial diversity, structure, and succession. In this review, we thoroughly reviewed the most recent findings on the piglet gut microbiome shifts as influenced by weaning, and how these microbiome changes brought about by various factors that have been shown to affect the development of microbiota in piglets. This review will provide a general overview of recent studies that can help to facilitate the design of new strategies to modulate the gut microbiome in order to enhance gastrointestinal health, growth performance and well-being of piglets.

Role of surfactants on particle deposition, wear, and corrosion behavior of TaC particle incorporated electroless nickel-phosphorus coating

The coatings prepared by incorporating submicron-sized TaC particles while depositing Ni and P atoms on the steel substrate via electroless method showed mixed results of surface finishing, microstructural properties,mechanical, tribological, and corrosion behavior with the addition of various surfactants during the coating process. The surfactant sodium dodecyl sulfate(SDS) enhanced the microhardness when added during the coating process of Ni–P–TaC composite, whereas the cetyltrimethyl ammonium bromide(CTAB) lowered the microhardness as compared to the surfactant-free coating. Moreover, the TaC particle incorporation was hindered by the surfactants CTAB and Triton X-100. An excellent wear resistance behavior has been observed in the surfactantfree Ni–P–TaC coating in comparison to particle-free Ni–P coating. The Ni–P–TaC coatings showed the dependency of wear behavior on the material properties of the counter sliding surface although this dependency has not been observed in Ni–P coatings. The wear damage in Ni–P coating is immense irrespective of the properties of counter-sliding materials. Various surfactant-used Ni–P–TaC coating surfaces deteriorated much more due to corrosion than surfactant-free Ni–P–TaC coating. Among the various surfactants, the CTAB used Ni–P–TaC coating showed weak corrosion resistance.

Ex vivo relaxation effect of Cuscuta chinensis extract on rabbit corpus cavernosum

The effect of Cuscuta chinensis extract on the rabbit penile corpus cavernosum （PCC） was evaluated in the present study. Penises obtained from healthy male New Zealand white rabbits （2.5-3.0 kg） were precontracted with phenylephrine （Phe, 10 pmol I^-1） and then treated with various concentrations of Cuscuta chinensis extract （1, 2, 3, 4 and 5 mg ml^-1）. The change in penile tension was recorded, and cyclic nucleotides in the PCC were measured by radioimmunoassay （RIA）. The interaction between Cuscuta chinensis and sildenafil was also evaluated. The result indicated that the PCC relaxation induced by Cuscuta chinensis extract was concentration-dependent. Pre-treatment with an nitric oxide synthase （NOS） inhibitor （No〉 nitro-L-arginine-methyl ester, L-NAME）, a guanylyl cyclase inhibitor （1H-[1,2,4]oxadiazolo[4,3-a]quinoxalin-l-one, ODQ）, or a protein kinase A inhibitor （KT 5720） did not completely inhibit the relaxation. Incubation of penile cavernous tissue with the Cuscuta chinensis extract significantly increased cyclic guanosine monophosphate （cGMP） and cyclic adenosine monophosphate （cAMP） in the PCC. Moreover, the Cuscuta chinensis extract significantly enhanced sildenafil-induced PCC relaxation. In conclusion, the Cuscuta chinensis extract exerts a relaxing effect on penile cavernous tissue in part by activating the NO-cGMP pathway, and it may improve erectile dysfunction （ED）, which does not completely respond to sildenafil citrate.

Microstructural evolution and mechanical properties of binary Mg-xBi(x=2,5,and 8 wt%)alloys

The microstructures and mechanical properties of Mg-x Bi(x=2,5,and 8 wt%)were investigated compared with pure *** ascast Mg-Bi billets consist of dendriticα-Mg grain matrix,divorced eutectic Mg_(3)Bi_(2) phase and secondary precipitated Mg_(3)Bi_(2) *** homogenization,secondary precipitated Mg_(3)Bi_(2) particles were all dissolved into the matrix,while most of the divorced eutectic Mg_(3)Bi_(2) intermetallic compounds *** the as-extruded samples exhibit fully dynamic recrystallization(DRX)and the average grain size decreases and the amount of nano-scale Mg_(3)Bi_(2) precipitates increases with increasing Bi ***,Bi alloying has little influence on the texture of the as-extruded pure Mg,with all the samples showing typical basal *** the results of grain refinement and precipitation hardening,the strengths of the as-extruded samples increase under both tensile and compressive ***,the yield asymmetry significantly decreases with Bi ***,all the as-extruded samples represent similar ductility under compression,while the tensile elongation first increases and then decreases with more Bi *** as-extruded Mg-5Bi alloy demonstrates good combination of strength and ***,Mg-8Bi alloy displays lower tensile ductility than that of Mg-5Bi alloy due to the presence of abundant undissolved coarse Mg_(3)Bi_(2) particles,which act as cracking sources during tensile ***,the strain hardening rate,strain hardening exponent n,and hardening capacity Hc are greatly reduced by Bi addition,mainly owing to the enhanced synergy effects of grain refinement and numerous Mg_(3)Bi_(2) precipitates.
