Current strategies for drug discovery research mainly focus on molecular targets,in which structure biology plays a central role,such as the detailed mechanism of a drug-target *** principle of this approach is the presumed molecular mechanism or genetic background of the disease being *** this approach has brought progress in understanding the mode of action for some drugs,most prevalent illnesses-especially chronic diseases-are multifactorial and present with a group of symptoms,as is often the case in cardiovascular diseases,metabolic disorders,cancers,and so *** newly emerging term“multi-morbidity”(i.e.,multiple co-occurring diseases)describes a grand challenge for this strategy,and successful drug intervention for multimorbidity should be multifaceted[1].So,our research team has shifted its attention to a new direction that views the disease as a whole instead of a single-molecule abnormality,and focusing on drugs with multiple targets.

In antiviral therapy of hepatitis B virus(HBV)infection,drug resistance remains a huge obstacle to the long-term effectiveness of nucleoside/tide analogs(NAs).Primary resistance mutation(rtM204V)contributes to lamivudine(LAM)-resistance,and compensatory mutations(rtL180M and rtV173L)restore viral fitness and increase replication *** evaluation of new anti-viral agents against drug-resistant HBV is limited by the lack of available small-animal *** established LAM-resistance HBV replication mice models based on clinical LAM-resistant HBV ***(rtM204VþrtL180M)or triple(rtM204VþrtL180MþrtV173L)lamivudine-resistant mutations were introduced into HBV expression vector,followed by hydrodynamic injection into tail vein of NOD/SCID *** was detected on days 5,9,13 and 17 and liver HBV DNA was detected on day 17 after *** serum and liver HBV DNA levels in LAM-resistant model carrying triple mutations are the highest among the *** NAs,LAM and entecavir(ETV),were used to test the availability of the *** and ETV inhibited viral replication on wild-type *** was no longer effective on LAM-resistant models,but ETV retains a strong ***,these models can be used to evaluate anti-viral agents against lamivudine-resistance,affording new opportunities to establish other drug-resistant HBV small-animal models.

Anodized metal oxide nanostructures for photoelectrochemical water splitting

Photoelectrochemical(PEC) water splitting offers the capability of harvesting, storing, and converting solar energy into clean and sustainable hydrogen energy. Metal oxides are appealing photoelectrode materials because of their easy manufacturing and relatively high stability. In particular, metal oxides prepared by electrochemical anodization are typical of ordered nanostructures, which are beneficial for light harvesting, charge transfer and transport, and the adsorption and desorption of reactive species due to their high specific surface area and rich channels. However, bare anodic oxides still suffer from low charge separation and sunlight absorption efficiencies. Accordingly, many strategies of modifying anodic oxides have been explored and investigated. In this review, we attempt to summarize the recent advances in the rational design and modifications of these oxides from processes before, during, and after anodization. Rational design strategies are thoroughly addressed for each part with an aim to boost overall PEC performance. The ongoing efforts and challenges for future development of practical PEC electrodes are also presented.

Hepatitis C: From inflammatory pathogenesis to antiinflammatory/hepatoprotective therapy

Hepatitis C virus(HCV) infection commonly causes progressive liver diseases that deteriorate from chronic inflammation to fibrosis, cirrhosis and even to hepatocellular carcinoma. A long-term, persistent and uncontrolled inflammatory response is a hallmark of these diseases and further leads to hepatic injury and more severe disease progression. The levels of inflammatory cytokines and chemokines change with the states of infection and treatment, and therefore, they may serve as candidate biomarkers for disease progression and therapeutic effects. The mechanisms of HCV-induced inflammation involve classic pathogen pattern recognition, inflammasome activation, intrahepatic inflammatory cascade response, and oxidative and endoplasmic reticulum stress. Direct-acting antivirals(DAAs) are the first-choice therapy for effectively eliminating HCV, but DAAs alone are not sufficient to block the uncontrolled inflammation and severe liver injury in HCV-infected individuals. Some patients who achieve a sustained virologic response after DAA therapy are still at a long-term risk for progression to liver cirrhosis and hepatocellular carcinoma. Therefore, coupling with antiinflammatory/hepatoprotective agents with anti-HCV effects is a promising therapeutic regimen for these patients during or after treatment with DAAs. In this review, we discuss the relationship between inflammatory mediators and HCV infection, summarize the mechanismsof HCV-induced inflammation, and describe the potential roles of anti-inflammatory/hepatoprotective drugs with anti-HCV activity in the treatment of advanced HCV infection.

Four stereoisomers of 3,5-bis（2-hydroxybut-3-en-1-yl）-l,2,4-thiadiazole, named insatindigothiadia- zoles A-D （1a-1d）, were isolated from the roots oflsatis indigotica. Their structures were determined by spectroscopic analysis; specifically, the absolute configurations were assigned by using the MPA determination rule based on △δrs values of MPA esters, and supported by electronic CD （ECD） calculations. Proposed biosynthetic pathways and preliminary investigations of the biological activities of la-1d against influenza virus A （H3N2）, Coxsackie virus B3, and/or HSV-1 are also discussed.

Eight new polycyclic phloroglucinol meroterpenoids guajamers A-H(1-8),a methylated benzoylphloroglucinol meroterpenoid guajamer I(9)representing a new skeleton,and two known analogues(10 and 11)were isolated from the leaves of Psidium *** structures of new molecules were elucidated by detailed analysis of spectroscopic data,and those of 1,2,8,and 9 were unambiguously confirmed by single-crystal X-ray diffraction ***,compounds 1-8 were sesquiterpene and monoterpene-based meroterpenoids with rearranged skeletons,while compound 9 was the first case of 3-alkyl-5-formyl-benzoylphloroglucinol-coupled sesquiterpene containing an unusual C-l-spiro-fused 6/6/9/4 polycyclic *** addition,all the isolated compounds were evaluated for their antibacterial activity against three bacterial strains,and most of them(compounds 2-7,10,and 11)showed antibacterial activity against Staphylococcus aureus and Staphylococcus epidermidis with MIC values of 8-32 μmol/*** findings suggested that meroterpenoids isolated from Psidium guajava can be considered as potential antibacterial leading compounds for pharmaceutical industry.

Our first priority was and will always be to serve our readers and the best way is to diligently curate our publications to ensure the high quality of our articles. For our efforts, APSB was included by the Chemical Abstract in 2011, accepted by Pub Med Central in 2015, indexed by Science Citation Index in 2017 and has evolved to become one of the most important international journals in the field of pharmaceutical sciences.
