AIM： To investigate the mechanism underlying the loss of responsiveness to anti-vascular endothelial growth factor（VEGF） treatment after repeated injections for choroidal neovascularization, VEGF and VEGF receptor（VEGFR） expressions were evaluated following repeated bevacizumab treatments in hypoxic human umbilical vein endothelial cells（HUVECs） in ***： HUVECs were incubated under hypoxic conditions in two media of different bevacizumab concentrations（1.0 or 2.5 mg/m L） for 17 h, and then in a new medium without bevacizumab for 7h. This procedure was repeated twice more. A culture with an identical volume of excipients served as the control. Cytotoxicity and cell proliferation were assessed using 3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyl tetrazolium bromide and Ki-67 assays, respectively. Levels of VEGF and VEGFR were assessed using enzyme-linked immunosorbent assay and Western blot ***： Cytotoxic effects were not reported for either bevacizumab concentration. Cell proliferation was not reduced after anti-VEGF treatments. VEGF level after single treatment was significantly higher than that of the control and after repeated treatments. Phosphorylated VEGFR-2 expression increased significantly after singleand repeated bevacizumab treatments compared with the control. The 1.0 mg/m L bevacizumab induced significantly higher expressions of VEGFR-2 than the 2.5 mg/m L in single and repeated treatment ***： Bevacizumab treatment of HUVECs elevated VEGFR expression in both single and repeated treatments, indicating a mechanism for the reduced efficacy of anti-VEGF therapy in ocular neovascular disorders.

The antitumor capabilities of agonistic anti-4-1BB mAbs have made them an attractive target for tumor ***,the adverse side effects associated with agonist antibodies have hindered their clinical ***,we aimed to study the immune-related adverse events of repeated doses and long-term use of agonistic anti-4-1BB *** show that chronic activation of 4-1BB signals induced the accumulation of IFN-γ-producing PD-1^(+)CD8^(+)T cells in the secondary lymphoid organs of tumor-bearing mice by increasing the number of dividing CD8^(+)T cells,which was beneficial for suppressing tumor growth in the early phase of anti-4-1BB ***,repeated exposure to anti-4-1BB mAbs led to granuloma development in tumor-draining lymph nodes(TDLNs)of mice due to recruitment and accumulation of macrophages via the CD8^(+)T cell-IFN-γ*** was accompanied by excessive lymph node swelling,which impaired the sequential activation of CD8^(+)T *** data provide insights into the immune-related adverse events of long-term agonist 4-1BB antibody dosing,which should be considered during the clinical development of immunomodulating therapy.

＜正＞Recently,sand/gravel materials,instead of crushed rock materials,are also utilized as a main rockfill material to overcome geological and environmental *** this paper,an experimental scheme for centrifuge modeling of concrete faced gravel-fill dam was developed and two centrifuge tests were performed in order to investigate the effect of drainage zoning on seepage *** dam considered in this study was designed to have Zone 3Bs of high permeability in main gravel-fill zone to enhance safety against accidental water infiltration into the *** centrifuge tests with and without Zone 3Bs were performed in order to investigate the effect of Zone 3Bs on seepage flow. Model dams were made of soils sieved to have equivalent permeability,because the grain size of original soil material is too big to construct model *** infiltration was simulated by rising water table over pre-implemented cracks on the *** behaviors of model dams were monitored by pore water pressure transducers and cameras during the *** tests showed that the drainage zone of CFGD drains off the infiltrated water out of the dam in a short *** modeling performed on the testing models also supported the effectiveness of the drainage zone.

根据亚洲减灾中心(ADRC,2016)提供的1987～2016年自然灾害统计数据,水(如洪水和暴雨)引发的灾害为第一大灾害,其发生次数、死亡人数、受灾人数和经济损失的占比分别达到了70.9%、78.9%、84.5%和38.7%,而韩国的灾害变化趋势与之相同。在此背景下,各国灾害管理工作的重点自然都集中在应对洪灾、雨灾和震灾而非其他类型的灾害上。因此,本研究提出了一个涉水基础设施灾害管理平台(WINS)。WINS可以提供灾害管理全过程所需的多种功能,即预防—准备—响应—恢复、实时安全评估、损失信息传送以及通过单一平台为受损涉水基础设施提供应急恢复行动。本灾害管理平台是基于《联合国减灾风险行动计划:可持续发展的风险指引和综合方法》的理念而建,预计其应用将有助于提高城市和涉水基础设施本身的复原力。

大坝一旦完建,大坝业主和大坝工程师的首要职责就是通过实施检查和安全评估对大坝进行合理的维护,这也是相关条例、法规和法案的要求。之前,韩国的大坝安全评估都是基于规范和定性的方法,没有考虑自然的不确定性。最近,基于风险的大坝安全管理被认为是大坝安全管理下一代范例中真正可靠的选择。笔者介绍了目前传统的制度体系在大坝安全管理中是如何运作的,以及基于风险的大坝安全管理是如何作为一种新的大坝安全范例来引进的。笔者还会介绍一些大坝安全导则和法规来追述韩国大坝安全管理的变化历史。最后,笔者将简要介绍某大坝在大坝风险评估方面开展的工作,这是新的大坝安全范例。
