N-terminal truncated RHT-1 proteins generated by translational reinitiation cause semi-dwarfing of wheat Green Revolution alleles

The unprecedented wheat yield increases during the Green Revolution were achieved through the introduc-tion of the Reduced height(Rht)-B1b and Rht-D1b semi-dwarfing *** Rht-1 alleles encode growth-repressing DELLA genes containing a stop codon within their open reading frame that confers gibberellin(GA)-insensitive *** this study,we successfully took the hurdle of detecting wild-type RHT-1 proteins in different wheat organs and confirmed their degradation in response to *** further demonstrated that Rht-B1b and Rht-D1b produce N-terminal truncated proteins through trans-lational *** of these N-terminal truncated proteins in transgenic lines and in Rht-D1c,an allele containing multiple Rht-D1b copies,demonstrated their ability to cause strong dwarfism,resulting from their insensitivity to GA-mediated degradation.N-terminal truncated proteins were detected in spikes and nodes,but not in the aleurone *** Rht-B 1b and Rht-D1b alleles cause dwarfism but have wild-type dormancy,this finding suggests that tissue-specific differences in translational reinitiation may explain why the Rht-1 alleles reduce plant height without affecting *** together,our findings not only reveal the molecular mechanism underlying the Green Revolution but also demonstrate that trans-lational reinitiation in the main open reading frame occurs in plants.

The combined influence of maturation,ripening,and climate on the profile of bioactive compounds was studied in banana(Musa acuminata,AAA,Cavendish,*** Naine).Their bioactive compounds were determined by the Folin-Ciocalteu assay and high-performance thin layer chromatographic(HPTLC) *** polyphenol content of bananas harvested after 400 degree days remained unchanged during ripening,while bananas harvested after 600 and 900 degree days exhibited a significant polyphenol *** dopamine was the polyphenol with the highest concentration in banana peels during the green developmental stage and ripening,its kinetics differed from the total polyphenol *** results showed that this matrix of choice(maturation,ripening,and climate) may allow selection of the banana(***,AAA,Cavendish,*** Naine) status that will produce optimal concentrations of identified compounds with human health relevance.

An Integrated Geochemical and Mineralogical Approach for the Evaluation of Zn Distribution in Long-Term Sludge-Amended Soil

This research work was designed to compare the Zn distribution in a long-term sludge-amended soil with that in a control soil. Two complementary approaches were performed: 1) a geochemical approach at the metric scale of the bulk soil horizons and 2) a mineralogical approach at the micrometric scale of the primary minerals weathering microsites. The geochemical approach revealed that Zn in the control soil was inherited from the weathering parent-rock. Its concentration was always lower than in the amended soil where Zn was supplied at the surface by the spread sludges and moves downwards. The mineralogical approach showed that the clay minerals, produced by the weathering of the primary minerals (amphiboles and plagioclases), or filling the fissure network were made up of smectites (saponite and montmorillonite) at the bottom and kaolinite at the top of the two soil profiles. Each clay mineral, with its specific sorption capacity, controlled the Zn distribution within the soil: the smectites produced by the amphiboles had high sorption capacity and favored Zn retention in the upper horizons of the soil. Conversely, the kaolinites produced by the plagioclases had lower sorption capacity, did not retain Zn in the surface horizons, and allowed it to migrate to deeper horizons where it was sorbed onto the montmorillonites.

Local seasonal migrations have long been common in France,and the migrations can be throughout the whole *** identification and assessment of such migration in European countries trace back to 1805 under the Napoleonic Empire regime,when nationality was based on jus sanguinis(Latin,right of blood)with the ultimate purpose of ensuring sufficient recruits in the Napoleonic armies,which is unlike the Republican France today that defines nationality on the basis of jus soli(Latin,right of the soil,also known as birthright citizenship).At the beginning of the 21st century,immigration has become a major challenge to the French domestic policy,and is closely linked to the issues of public safety,national identity,labor market,development of the European Union public policy,and the integration of the second generation by the secular,republican French *** migration within and outside of the Europe demonstrates four conceptual issues:European residents’country of origin,French nationality,the exercising of French citizenship,and the construction of a personal identity in the context of a more and more complex family ethnic composition after the restructuring.

1 Background Congenital heart disease(CHD)is the most common major congenital anomaly,affecting approximately one in every 100 live births[1].Among congenital anomalies,66%of preventable deaths are due to CHD,and 58%of the avertable morbidity and mortality due to congenital anomalies would result from scaling congenital heart surgery services[2].Every year,nearly 300,000 children and adults die from CHD,the majority of whom live in low-and middle-income countries(LMICs)[3].Approximately 49%of all individuals with CHD will require surgical or interventional care at some point in their lifetime[4];as a result of advances in access to and the delivery of such services,over 95%of children born with CHD in high-income countries now live into adulthood[3].Here,adults have surpassed children in the number of CHD cases at a ratio of 2:1[5].

目的了解中法两国对早发型新生儿脓毒症（early—onsetneonatalsepsis，EONS）在临床表现、诊断、治疗方面的不同，为提高重庆医科大学附属儿童医院（简称重医）对EONS的诊治水平提供依据。方法对法国某医院和重医共146例EONS患儿的细菌谱、临床表现、抗菌药使用、产前危险因素记录和筛查情况进行回顾性分析。结果重医常见致病菌为凝固酶阴性葡萄球菌（69．2％）和大肠杆菌（15．4％）；法国医院为B族链球菌（33．3％）和大肠杆菌（33．3％）。法国医院胃液和周围拭子培养常见大肠杆菌（33．3％），B族链球菌（21．2％），凝固酶阴性葡萄球菌（18．2％）等。重医抗菌药使用总天数11．4±7．2（d）、平均每个患儿抗菌药使用的种类（3．1±0．9）和换抗菌药人数比例（70．2％）均大于法国医院6．2±2．5（d），2．2±0．8（d），（9．9％）。两院首剂抗菌药常联用两种抗菌药（重医二代头孢＋半合成青霉素，法国医院阿莫西林＋阿米卡星）。重医二、三线抗菌药常见碳青霉烯类和万古霉素，法国医院常见三代头孢和万古霉素。重医对多种产前危险因素（绒毛膜羊膜炎、母亲发热、破膜到胎儿娩出时间、孕期是否行阴道拭子或尿路感染情况、羊水污染、产前抗菌药使用、EONS的既往史）的记录率均低于法国医院；产前危险因素筛查阳性率两院差异不大。在生后立即出现异常而入院的患儿中，两院最常见的异常均为呼吸困难（96．5％VS．88．2％）；在生后72h之内出现异常的患儿中，两院常见入院原因构成比不同：重医常见原因为呼吸困难（44．4％），反应差（22．2％）等；法国医院常见原因为C反应蛋白升高（78．2％），发热（5．5％）等。两所医院CRP诊断EONS的假阴性率无统计学差异。结论两所医院对EONS诊断、治疗有很大不同。重视产前危险因素的筛查与记录、加强产科对初生新生儿的监护可提高重医对EONS患儿的早期诊断的准确性。对具有产前／产时危险因素的新生儿积极进行胃液／周围拭子培养并行相应的药敏试验可能帮助临床医生进行抗菌药的选用．减少患儿的换药次数和抗菌药使用时间。

Partial Beclin 1 silencing aggravates doxorubicin-and Fasinduced apoptosis in HepG2 cells

AIM： To investigate the role of Beclin 1 on the susceptibility of HepG2 cells to undergo apoptosis after anti-Fas antibody or doxorubicin treatment. METHODS： Beclin 1 silencing was achieved using RNA interference. DNA ploidy, the percentage of apoptotic cells and the mitochondrial membrane potential were assessed by flow cytometry. Levels of Beclin 1, BCI-XL and cytochrome c, and the cleavage of poly （ADP-ribose） polymerase （PARP） were assayed by using Western blots. RESULTS： Beclin 1 expression decreased by 75% 72 h after Beclin 1 siRNA transfection. Partial Beclin 1 silencing significantly increased the percentage of subG1 cells 24 and 40 h after treatment with doxorubicin or anti-Fas antibody, respectively, and this potentiation was abrogated by treatment with a pan-caspase inhibitor. Partial Beclin 1 silencing also increased PARP cleavage, mitochondrial membrane depolarization and cytosolic cytochrome c. The pro-apoptotic consequences of partial Beclin 1 silencing were not associated with a decline in Bcl-XL expression.CONCLUSION： Partial Beclin 1 silencing aggravates mitochondrial permeabilization and apoptosis in HepG2 cells treated with an anti-Fas antibody or with doxorubicin.

Nutritional and vitamin status in patients with neuroendocrine neoplasms

Symptoms of gastroenteropancreatic located neuroendocrine neoplasms(GEPNENs) are often related to food intake and manifest as abdominal pain or diarrhoea which can influence patients nutritional status. Malnutrition is common in cancer patients and influences quality of life, treatment options and survival but is also present in up to 40% of patients with GEP-NENs. As part of malnutrition there are often deficiencies in fat-soluble vitamins, mainly vitamin D. Little knowledge exists on trace elements. Several factors influence the development of malnutrition such as size and localisation of the primary tumour as well as metastases, side effects from treatment but also hormone production of the tumour itself. One of the main influencing factors leading to malnutrition is diarrhoea which leads to dehydration and electrolyte disturbances. Treatment of diarrhoea should be guided by its cause. Screening for malnutrition should be part of routine care in every GEP-NEN patient. Multidisciplinary treatment including dietician support is necessary for all malnourished patients with GEP-NENs.
