Expanded natural killer cells augment the antimyeloma effect of daratumumab, bortezomib, and dexamethasone in a mouse model

Background Assessment of minimal erythemal dose(MED) for individual patients has been used to guide the narrow- band UVB(NB-UVB) phototherapy,which sometimes causes discomfort and additional *** L value(the lightness of color in Commission Internationlale de l’Eclairge Lab color scale) measured by colorimeter was shown to be useful for predicting sensitivity to NB-UVB *** aim of the study was to compare the efficacy and safety of NB-UVB phototherapy between 50%of MED and colorimetric L value starting dose regimens for skin phototype III to V Korean patients with psoriasis. Method Twenty seven patients determined starting doses based on colorimetric L value,and 27 patients based on 50% of *** correlation analysis showed that L value had the most significant association with MED compared with skin phototypes,a,and b values,we designated starting doses of L value regimen as follows:300 mJ/cm(L >66), 400 mJ/cm(62 dose,and cumulative dose until achieving the state of near *** proportion of adverse effects was not also significantly different. Conclusions NB-UVB starting dose determination based on colorimetric L value was comparable with conventional MED based regimen in efficacy and safety for skin phototype III-V *** it provides much convenience and ease for both patients and physicians,colorimetric L value could partly substitute the MED checking methods in NB-UVB phototherapy.

The use of natural killer(NK)cells is a promising and safe immunotherapeutic approach in the field of cancer ***,combination treatments are required to enhance the effector functions and therapeutic efficacy of NK *** this study,we investigated the potential of daratumumab(Dara),bortezomib,and dexamethasone(Dvd)to augment the antitumor effects of NK cells in a multiple myeloma(MM)xenograft mouse *** cells were expanded and activated using the K562-OX40 ligand and membrane-bound IL-18 and IL-21 in the presence of IL-2 and IL-15 from peripheral blood mononuclear cells from MM patients.A human MM xenograft model was established using human RPMI8226-RFP-FLuc cells in NOD/SCID IL-2Rγnull(NSG)***-bearing mice were divided into six treatment groups:no treatment,expanded NK cells(eNKs),Dara,Dara+eNKs,Dvd,and Dvd+*** treatment strongly enhanced the cytotoxicity of eNKs by upregulating expression of NK cell activation ligands,downregulating expression of NK cell inhibitory ligands,and promoting antibody-dependent cellular *** combination of eNKs with Dvd significantly prolonged mouse survival and reduced the tumor burden and serum M-protein ***,Dvd pretreatment significantly increased eNK persistence and homing to MM *** findings suggest that Dvd treatment potentiates the antimyeloma effects of NK cells expanded and activated ex vivo by modulating immune responses in MM-bearing mice.

Measuring intrinsic hip external rotator strength (ER) without compensatory pelvic motion and activation of the sartorius is important for preventing or rehabilitating lower extremity injuries. However, the optimal method for measuring intrinsic hip ER muscle strength while minimizing compensatory pelvic motions and activation of the sartorius is unclear. The purpose of this study is to compare measurements of hip ER strength, compensatory pelvic motion, and sartorius activation in the sitting, prone, and sidelying positions. Thirty-one healthy subjects (16 males and 15 females) were recruited for this study. Hip ER strength, pelvic kinematics, and sartorius muscle activation were measured during maximal isometric contraction of the hip ER in the sitting, prone, and sidelying positions. Hip ER strength was measured using a load-cell-type strength-measurement sensor. Pelvic kinematics was measured using an electromagnetic motion-tracking sensor. Electromyography was used to measure sartorius muscle activity. Data were analyzed using one-way repeated-measures analysis of variance. The result showed that hip ER strength and sartorius muscle activation were significantly lower in the sidelying compared with the sitting and prone positions (p < 0.01). Pelvic anteroposterior tilting was significantly greater in the sitting compared with the prone and sidelying positions (p < 0.01). Pelvic rotation differed significantly among positions (p < 0.01). Pelvic lateral tilting was significantly greater in the prone compared with the sitting position (p < 0.017). Compensatory pelvic motion and sartorius muscle activation were lower when hip ER strength measurements were made in the sidelying position. Therefore, the sidelying position is effective for measuring selective intrinsic hip ER strength.

The biosynthesis of host lipids and/or lipid droplets(LDs)has been studied extensively as a putative therapeutic target in diverse viral ***,directly targeting the LD lipolytic catabolism in virus-infected cells has not been widely ***,we show the linkage of the LD-associated lipase activation to the breakdown of LDs for the generation of free fatty acids(FFAs)at the late stage of diverse RNA viral infections,which represents a broad-spectrum antiviral *** of membrane transporter systems due to virus-induced cell injury results in intracellular malnutrition at the late stage of infection,thereby making the virus more dependent on the FFAs generated from LD storage for viral morphogenesis and as a source of *** replication of SARS-CoV-2 and influenza A virus(IAV),which is suppressed by the treatment with LD-associated lipases inhibitors,is rescued by supplementation with *** administration of lipase inhibitors,either individually or in a combination with virus-targeting drugs,protects mice from lethal IAV infection and mitigates severe lung lesions in SARS-CoV-2-infected ***,the lipase inhibitors significantly reduce proinflammatory cytokine levels in the lungs of SARS-CoV-2-and IAV-challenged animals,a cause of a cytokine storm important for the critical infection or mortality of COVID-19 and IAV *** conclusion,the results reveal that lipase-mediated intracellular LD lipolysis is commonly exploited to facilitate RNA virus replication and furthermore suggest that pharmacological inhibitors of LD-associated lipases could be used to curb current COVID-19-and future pandemic outbreaks of potentially troublesome RNA virus infection in humans.

BACKGROUND Gastric ‘indefinite for neoplasm/dysplasia'(IFND) is a borderline lesion that is difficult to diagnose as either regenerative or neoplastic. There is a need for guidance in the identification of a subset of patients, who have an IFND lesion with a higher risk of malignant potential, to enable risk stratification and optimal *** To determine the clinical and pathologic factors for the accurate diagnosis of gastric IFND *** In total, 461 gastric lesions diagnosed via biopsy as IFND lesions were retrospectively evaluated. Endoscopic resection(n = 134), surgery(n = 22), and follow-up endoscopic biopsy(n = 305) were performed to confirm the *** time interval from initial biopsy to cancer diagnosis was measured, and diagnostic delays were categorized as > 2 wk, > 2 mo, > 6 mo, and > 1 year. The IFND lesions presenting as regenerating atypia(60%) or atypical epithelia(40%)at initial biopsy were adenocarcinomas in 22.6%, adenomas in 8.9%, and gastritis in 68.5% of the *** Four clinical factors [age ≥ 60 years(2.445, 95%CI: 1.305-4.580, P = 0.005),endoscopic size ≥ 10 mm(3.519, 95%CI: 1.891-6.548, P < 0.001), single lesion(5.702, 95%CI: 2.212-14.696, P < 0.001), and spontaneous bleeding(4.056, 95%CI:1.792-9.180, P = 0.001)], and two pathologic factors [atypical epithelium(25.575,95%CI: 11.537-56.695, P < 0.001], and repeated IFND diagnosis [6.022, 95%CI:1.822-19.909, P = 0.003)] were independent risk factors for gastric cancer. With two or more clinical factors, the sensitivity and specificity for carcinoma were91.3% and 54.9%, respectively. Ten undifferentiated carcinomas were initially diagnosed as IFND. In the subgroup analysis, fold change(5.594, 95%CI: 1.458-21.462, P = 0.012) predicted undifferentiated or invasive carcinoma in the submucosal layers or deeper. Piezo-scanner used in STMs has limited the scanning range up to several micrometers in two dimensions. Various two dimensional positioners have been tried to cover wider scanning range of STMs in order to locate the probing tip over a position of interest. However, existing positioners require entire modification of the previous STM system. Changing the original STM system is difficult because one should redesign the whole STM head and resize other parts. We constructed a X-Y piezoelectric positioner that requires only minor modification of the original STM system. It is designed to move a sample holder over a distance of some millimeters with a few nanometer steps on a plane. It has one X-aligned shear piezo stack and another Y-aligned shear piezo stack being orthogonal to the former one and its size is about 20×20×10 mm. Slip-stick motion could be realized by a contact consists of a stainless steel ball and a polished alumina surface. This X-Y positioner could be made by modifying only the sample holder and the electrical connection part between the STM head and new sample holder. We could expand scanning range to a few millimeters without modifying original STM system. Because only a slightly modification was needed, it was possible to use the original sample holder. For uniform samples we could use the original sample holder and for patterned samples or samples which have nano-structures, we could use our modified sample holder. Our observations are performed using optical microscope in the air at room temperature. We expect further improvement can be done by adjusting tension between a stainless steel ball and a polished alumina surface or by changing the combination of contact materials.

Objectives To evaluate the age-related one-year major adverse cardiocerebrovascular events （MACCE） after percutaneous coronary intervention （PCI） in acute myocardial infarction （AMI）. We analyzed the association between age and one-year MACCE after AMI. Methods A total of 13,104 AMI patients from Korea Acute Myocardial Infarction Registry-National Institue of Health （KAMIR-NIH） between November 2011 and December 2015 were classified into four groups according to age （Group I, 〈 60 years, n = 4199; Group II, 60-70 years, n = 2577; Group III; 70-80 years, n = 2774; Group IV, ≥ 80 years, n = 1018）. Patients were analyzed for one-year composite of MACCE （cardiac death, myocardial infarction, target vessel revascularization, cerebrovascular events） after AMI. Results The one-year MACCE in AMI were 3.5% （Group I）, 6.3% （Group II）, 9.6% （Group III） and 17.6% （Group IV）. After adjustment for confounding para- meters, the analysis results showed that patients with AMI had incremental risk of one-year MACCE [Group II, adjusted hazard ratios （aHR） = 1.224, 95% CI： 0.965-1.525, P = 0.096; Group III, aHR = 1.316, 95% CI： 1.037-1.671, P= 0.024; Group IV, aHR = 1.975, 95% CI： 1.500-62.601, P〈 0.001） compared to Group I. Especially, cardiac death in the composite of primary end point played a major role in this effect （Group II, aHR = 1.335, 95% CI： 0.941-1.895, P= 0.106; Group III, aHR = 1.575, 95% CI： 1.122-2.210, P= 0.009; Group IV, aHR = 2.803, 95% CI： 1.937-4.054, P 〈 0.001）. Conclusions Despite advanced techniques and medications for PCI in AMI, age still exerts a powerful influence in clinical outcomes. Careful approaches, even in the modem era of developed cardiology are needed for aged-population in AMI intervention.

This study was carried out to understand the long-term effect of organic waste treatment on the fate of heavy metals originated from the organic wastes, together with examination of changes in soil properties. For this, the soils received three different organic wastes (municipal sewage sludge, alcohol fermentation processing sludge, pig manure compost) in three different rates (12.5, 25, 50 ton/ha/yr) for 7 years (1994 - 2000) were used. To see the long-term effect, plant growth study and soil examination were conducted twice in 2000 and 2010, respectively. There was no additional treatment of organic wastes for post ten years after ceasing organic waste treatment for seven years. Soil examination conducted in 2010 showed decreases in soil pH, EC, total nitrogen, organic matter, available phosphorus, exchangeable cations and heavy metal contents in all soils received organic wastes compared to the results obtained in 2000. Speciation of heavy metals in soil through sequential extraction showed that organically bound Cu was the dominant species in all treatment and exchangeable Cu was increased in the plots treated with municipal sewage sludge and alcohol fermentation processing sludge. organically bound Ni increased from 25% - 30% to 32% - 45% in 2010 inall treatment while Pb showed increase in carbonate form in all treatments. Zn existed mainly as sulfide and residual forms, showing increases in organically bound form in all treatment during post ten years.
