Molecular docking studies of human MCT8 protein with soy isoflavones in Allan-Herndon-Dudley syndrome(AHDS)

Monocarboxylate transporter-8 （MCT8） is a specific thyroid hormone transporter, essential for the uptake of thyroid hormone into target tissues. Mutations in the MCT8 gene have been identified as the cause of Allan-Herndon-Dudley syndrome （AHDS）. It has been reported that soy isoflavones influence thyroid hormone system and can interact with thyroid hormone transporter proteins. Therefore, the present study aimed to find out whether soy isoflavones （genistein, daidzein and glycitein） can be used as a natural inhibitor to target MCT8 in AHDS. Docking studies were performed for soy isoflavones in order to evaluate their binding affinity to MCT8 protein using AutoDock4 （version 4.2.6） and AutoDock Vina. After docking, the ligands were ranked according to their binding energy and the best lead compound was selected based on the least binding energy. The docking results indicated that daidzein possesses the lowest binding energy against MCT8. Moreover, it was found that the residues PRO-338, HIS-341, and GLU-348 were involved in hydrogen bond interactions with genistein and daidzein. This study suggests that daidzein is a promising natural inhibitor to target MCT8 in AHDS.

Rocks encountered in foundations of heavy structures are invariably intersected by discontinuities(joints).In the past,several studies have been performed by researchers to incorporate the effect of fully persistent joints in the assessment of the load-carrying capacity of ***,in the field,the joints are non-persistent,and an assumption of full persistency will underestimate the ***,Shaunik&Singh have studied the influence of non-persistency,number of joint segments and discontinuity orientation on the strength behaviour of rock specimens(Shaunik and Singh,2019).Bell’s approach can be used to obtain the bearing capacity of shallow foundations placed in jointed *** the present study,results of the experimental work(Shaunik and Singh,2019)conducted by Shaunik&Singh have been used to suggest expressions by extending Bell’s approach for computing bearing capacity of the foundation placed near the crown of a rock *** to use design charts are also presented for field ***,a real-life problem from Indian Garhwal Himalayas is considered,and the approach suggested in this study is utilised to obtain the bearing capacity of a bridge foundation as a function of uniaxial compressive strength(UCS)of intact rock,joint friction,spacing and orientation of joint,nonpersistency and number of joint segments.

Chemotherapy is an effortless and frequently used approach in cancer ***,in most cases,it can only prolong life expectancy and does not guarantee a complete ***,chemotherapy is associated with severe adverse effects,one of the major complications of effective cancer *** addition,newly published research outputs show that cancer stem cells are involved in cancer disease progression,drug resistance,metastasis,and recurrence and that they are functional in the trans-differentiation capacity of cancer stem cells to cancer cells in response to *** strategies are therefore required for better management of cancer *** prime approach would be to synthesize and develop novel drugs that need extensive resources,time,and endurance to be brought into therapeutic *** subsequent approach would be to screen the anti-cancer activity of avail-able non-cancerous *** concept of repurposing non-cancer drugs as an alternative to current cancer therapy has become popular in recent years because using existing anticancer drugs has several adverse *** have also been investigated for cancer ther-apy due to their significant anti-cancer effects with negligible or no side effects and availabil-ity in food *** this paper,we discuss an ideal hypothesis for screening available non-cancerous drugs with anticancer activity,with a focus on cancer stem cells and their clinical application for cancer ***,drug repurposing and the combination of micronu-trients that can target both cancers and cancer stem cells may result in a better therapeutic approach leading to maximum tumor growth control.

Several constructions in the field of civil engineering quite often need to deal with *** behaviour of rock intersected by a discontinuity or a set of discontinuities has been a topic of keen interest for engineering *** popular attributes of discontinuities that have been given due importance are their frequency,orientation and surface ***-persistency,however,has been given little *** article presents an experimental study wherein focus has been made on the effect of non-persistency of the joint on the uniaxial compressive strength(UCS)of a model rock for various geometries such as orientation,discontinuity length ratio and number of joint *** applicability of single plane of weakness theory(SPWT)to assess the strength of jointed specimens has also been *** has been noticed that SPWT captures the strength behaviour only for a narrow range of discontinuity *** an improvement,an approach is suggested by extending concepts of degree of persistence and joint factor to have a better understanding towards strength behaviour of rocks intersected by non-persistent joints.

The disruptive effects of the COVID-19 pandemic has rapidly shifted how individuals navigate in *** are concerned that travel behavior will shift toward a car-driven and homeworking future,shifting demand away from public transport *** concerns place the recovery of public transport in a possible crisis.A resilience perspective may aid the discussion around recovery-particularly one that deviates from pre-pandemic *** paper presents an empirical study of London’s public transport demand and introduces a perspective of spatial resilience to the existing body of research on post-pandemic public transport *** study defines spatial resilience as the rate of recovery in public transport demand within census boundaries over a period after lockdown restrictions were *** relationship between spatial resilience and urban socioeconomic factors was investigated by a global spatial regression model and a localized perspective through Geographically Weighted Regression(GWR)*** this case study of London,the analysis focuses on the period after the first COVID-19 lockdown restrictions were lifted(June 2020)and before the new restrictions in mid-September *** analysis shows that outer London generally recovered faster than inner *** of income,car ownership and density of public transport infrastructure were found to have the greatest influence on spatial patterns in ***,influential relationships vary locally,inviting future research to examine the drivers of this spatial ***,this research recommends transport policymakers capture the influences of homeworking,ensure funding for a minimum level of service,and advocate for a polycentric recovery post-pandemic.

We propose to investigate the THz field generation using nonlinear mixing mechanism of laser beat wave with inclined rippled density *** laser pulses with frequencies(ω_(1),ω_(2)) and wave vectors(k_(1),k_(2)) co-propagate and resultant laser beat wave forms at beat frequency(ω_(1)-ω_(2)).Laser beat wave imparts a nonlinear force on the ambient electrons and pushes them outward with nonlinear velocity v_(NL).Coupling of induced density perturbation and nonlinear velocity v_(NL)generates nonlinear currents at laser beat frequency that further generates electromagnetic field E_((ω_(1)-ω_(2))) in terahertz(THz)*** the present scheme,density ripples are introduced at an angle with respect to laser propagation and flat Gaussian index(f) is introduced in laser field profile that transform curved top of Gaussian field envelope into flat top field *** combined effect of flat laser pulses with inclined density ripples in plasmas shows 10-fold enhancement in THz field amplitude when flat-Gaussian index(f) varies from 1 to ***,the THz field intensifies when density ripples inclination increases upto a certain angle and then decreases.

We are currently faced with an aging population, which is rapidly growing worldwide. Two thirds of cancer occurs in the over 65-year age group. Societal conceptions from the past have created ageist stereotypes; old age is associated with frailty and the elderly are perceived to be destined for deterioration and loss of independence. Cancer within the elderly is also subject to these stereotypes, with elderly cancer patients considered by some not as likely to recover as younger patients with cancer. We summarise and review the current concerns regarding elderly management and treatments utilised for the management of oncological disease in the elderly, and discuss the impact of under-treatment within this population.

Demand for graphite in the forthcoming years to develop Li-ion batteries(LIBs)with the goal of driving electric vehicles(EV)and its requirement in multifarious energy storage applications as an *** emerging sector of LIB-based EVs,along with portable electronics,produces an inevitable volume of batteries in the e-waste *** main reason for the lower percentage of recycling(at present,<5%)is due to the recovery of economically rich metals like Li,Ni,and ***,complete recycling technologies,including the strategic material graphite,which is available in a massive amount of spent LIBs,are urgently needed to be updated to ensure the reuse of all *** approach lifts the recycling process to develop an economic one besides the geostrategic and environmental policy ***,we summarize the importance of graphite and its demand and specify the reasons to recycle the graphite from spent LIBs along with its development as an anode in ***,the approach of the current recycling process of graphite in lab-scale and industries for various applications,including energy storage,are discussed with the highlights of future progress.

In recent years,machine learning algorithms and in particular deep learning has shown promising results when used in the field of legal *** legal field is strongly affected by the problem of information overload,due to the large amount of legal material stored in textual *** text processing is essential in the legal domain to analyze the texts of the court events to automatically predict smart *** an increasing number of digitally available documents,legal text processing is essential to analyze documents which helps to automate various legal domain *** document classification is a valuable tool in legal services for enhancing the quality and efficiency of legal document *** this paper,we propose Sammon Keyword Mapping-based Quadratic Discriminant Recurrent Multilayer Perceptive Deep Neural Classifier(SKM-QDRMPDNC),a system that applies deep neural methods to the problem of legal document *** SKM-QDRMPDNC technique consists of many layers to perform the keyword extraction and ***,the set of legal documents are collected from the *** the keyword extraction is performed using SammonMapping technique based on the distance *** the extracted features,Quadratic Discriminant analysis is applied to performthe document classification based on the likelihood ratio ***,the classified legal documents are obtained at the output *** process is repeated until minimum error is *** experimental assessment is carried out using various performance metrics such as accuracy,precision,recall,F-measure,and computational time based on several legal documents collected from the *** observed results validated that the proposed SKM-QDRMPDNC technique provides improved performance in terms of achieving higher accuracy,precision,recall,and F-measure with minimum computation time when compared to existing methods.
