DNA methylation is an important epigenetic change affecting gene expression in plants in both normal and stress conditions. The organelles, mitochondria and chloroplast play a significant role in sensing and initiating stress response. In this study, we report the methylation pattern in chloroplast and mitochondrial genomes in irrigated and water stressed conditions and its relationship with gene expression of a drought tolerant Zea mays cultivar, Z59. Whole genome bisulfite sequencing was done to analyze the pattern of methylation in both the conditions. Mapping of bisulfite reads to B73 reference of mitochondrial and chloroplast genomes showed hypomethylation in water stressed plants when compared to irrigated plants. Sliding window approach to the methylation count data showed highest peak at 419,800 to 420,800 bp region in mitochondria and at 36,900 to 37,900 bp region in chloroplast genomes in both samples. Annotation of the methylated genomes showed that, genes related to photosystem I & II in chloroplast and nad4 gene in mitochondria were hypo methylated in the water stressed sample. RNA-seq analysis of transcriptomics reads mapped to the same reference showed regulation of rps3, rps2A, ccmFC, atp1 and many uncharacterized genes in mitochondria and psbA, psbD, psbc, psaA, and atpA, genes in chloroplast.

Glycemic Indices of Multiple Oral Nutritional Supplements: A Randomized Cross-Over Study in Indian Adults

background: A randomized cross-over study was conducted to assess the glycemic index (GI) of seven Oral Nutritional Supplements (ONSs). These ONSs are designed to support the nutritional requirements of different age-groups, physiological states, or health conditions among Indian adults. Methods: The study had two phases viz., phase1 (n = 18) studied two ONSs: A1 and b1 and phase 2 (n = 20) studied five ONSs: A2, b2, C2, D2 & E2. The subjects were healthy, non-diabetic adults, aged between 20 - 44 years with a mean body Mass Index of 21.2 ± 1.52 kg/m2 (Phase 1) and 21.0 ± 1.45 kg/m2 (Phase 2). All these ONSs were compared with reference drinks (glucose). The carbohydrates in one serving of each ONS were matched to carbohydrates from 25 grams of glucose following ISO 2010 guidelines. Capillary blood was assessed for blood glucose response at baseline, 15, 30, 45, 60, 90 and 120 minutes. GI was calculated as the incremental area under the curve (iAUC) for the test drinks and expressed as a percentage of the average iAUC from glucose. Results: Phase 1 indicated that the high fiber diabetes-specific nutrition supplement A1 with higher protein (23% energy), higher fat (25% energy) and reduced carbohydrates (40% energy) had a significantly (p = 0.002) lower GI [34 (±6)] as compared to b1 [63 (±7)] (protein 19%, fat 7% and carbohydrates 60% energy) even with similar amount (22%) and type of fiber. Phase 2 reported that all test products [A2 (32 ± 5), b2 (37 ± 4), C2 (31 ± 5), D2 (31 ± 5) and E2 (55 ± 4)] had a low GI. As compared to phase 1, ONSs in phase 2 had lower fiber content (1.6% - 4.6% energy). Conclusion: The glycemic index of oral nutrition supplements is influenced not only by their fiber content, but also by the overall macronutrient composition including protein (≥17% energy), fat (≥10% - 27% energy) and carbohydrates (40% - 57.5% energy).

Recognizing the composition and modulation of the microbiome, a viable therapeutic tool for multi-targeted therapy is a new strategy that has recently been explored. Glucosamine (GS) is being studied for its prebiotic potential in addition to being the most abundant and naturally occurring amino monosaccharide. The current study focuses on glucosamine’s prebiotic potential by assessing the stability of various GS concentrations (1% - 5%) in the gastrointestinal tract (GIT) and its ability to be fermented by the gut microbiota. The results showed that GS stimulated the most growth in L. acidophilus even after a longer incubation time than b. bifidum and L. acidophilus growth was concentration-dependent, with maximum growth at 3% with a simultaneous decrease in pH (5.6 - 1.7). The decrease in GS concentration with time also represented the growth of bacterial species, demonstrating the species’ utilization of GS. Furthermore, at 3%, GS also represented the prebiotic index of 1.9. In addition, the concentration of GS in various simulated GIT fluids was estimated in both fast and fed conditions to examine GS stability at various levels in the gut. The results showed that GS remained unaffected and non-digestible in all of the simulated GIT fluids (salivary, gastric, intestinal, and colonic), but there was a slight decrease in GS concentration (2.8%) in the fasted state of gastric fluid due to low pH levels (1.6). As a result, the findings are conclusive and suggest that GS possesses prebiotic properties.

The present study investigated the antiarrhythmic activity of alcoholic extract of Tinospora cordifolia （T. cordifolia） in CaCl2 induced arrhythmia. CaCl2 （25 mg/kg） was administered by intravenous infusion （iv） to produce arrhythmia in rats. The animals were then treated with T. cordifolia extract （150, 250, and 450 mg/kg） and verapamil （5 mg/kg,iv）. Lead II electrocardiogram was monitored. Plasma calcium, sodium and potassium levels were measured. In CaCl2 induced arrhythmia, heart rate was decreased by 41.10%, T. cordifolia at 150, 300, and 450 mg/kg decreased the heart rate by 26.30%, 29.16%, and 38.29%, respectively, and verapamil reduced the heart rate by 9.70% compared to the normal group. The PQRST waves were normalized and atrial and ventricular fibrillation was controlled in rats treated with verapamil and T. cordifolia. CaCl2 increased calcium and sodium levels and decreased potassium levels in blood. T. cordifolia dose-dependently decreased calcium and sodium levels and increased potassium levels. Hence, T. cordifolia can be used in antiarrhythmic clinical settings and beneficial in atrial and ventricular fibrillation and flutter and may be indicated in ventricular tachyarrhythmia.

Spatio-Temporal Expression Patterns of Aurora Kinases A, b in Mouse Zygotes during the First Mitosis

Objective To investigate the expression and localization of Aurora kinase A （A URKA ） and Aurora kinase b （A URKb） in mouse zygotes during the process of the first mitosis. Methods Quantitative real-time RT-PCR and Western blotting were performed to analyze the expression of AURKA and AURKb. The subcellular location of AURKA and A URKb was studied by confocal microscopy. Results A URKA and A URKb were increasingly expressed from phase G1 and peaked at phase M. After the entrance into mitosis A URKb became the predominant form both in mRNA and protein levels. The proteins of A URKA and A URKb both distributed in the cytoplasm and were associated with nucleus during the first mitosis of mouse zygotes, with some details in different. Conclusion The expression and localization of Aurora kinases A and b was in a cell- cycle regulated manner during the process of the cleavage of mouse zygotes. This discovery will aid in future investigations on their specific roles and molecular mecha- nisms in the regulation of mammalian early embryonic development.

目的:研究Aurora激酶A(AURKA)、Aurora激酶b(AURKb)在小鼠受精卵第一次有丝分裂进程中的表达与亚细胞定位。方法:分别利用实时荧光定量PCR和Western blotting法从mRNA和蛋白水平上检测AURKA、AURKb在小鼠受精卵第一次有丝分裂进程中的表达规律,并用间接免疫荧光法观察AURKA/b蛋白在各时相的细胞定位。结果:AURKA、AURKb在受精卵的G2、M期呈高表达,G1、S期呈低表达,AURKA/b蛋白主要定位于胚胎的细胞质和细胞核中,但细节上各有特点。结论:AURKA、AURKb在小鼠受精卵第一次有丝分裂进程各期呈时空特异性表达,提示其对小鼠早期胚胎发育过程发挥重要的调控作用。
