Transmission restriction and genomic evolution co-shape the genetic diversity patterns of influenza A virus

Influenza A virus (IAV) shows an ex.ensive host range and rapid genomic variations, leading to continuous emergence of novel viruses with significant antigenic variations and the potential for cross-species transmission. This causes global pandemics and seasonal flu outbreaks, posing sustained threats worldwide. Thus, studying all IAVs' evolutionary patterns and underlying mechanisms is crucial for effective prevention and control. We developed FluTyping to identify IAV genotypes, to ex.lore overall genetic diversity patterns and their restriction factors. FluTyping groups isolates based on genetic distance and phylogenetic relationships using whole genomes, enabling identification of each isolate's genotype. Three distinct genetic diversity patterns were observed: one genotype domination pattern comprising only H1N1 and H3N2 seasonal influenza subtypes, multi-genotypes co-circulation pattern including majority avian influenza subtypes and swine influenza H1N2, and hybrid-circulation pattern involving H7N9 and three H5 subtypes of influenza viruses. Furthermore, the IAVs in multi-genotypes co-circulation pattern showed region-specific dominant genotypes, implying the restriction of virus transmission is a key factor contributing to distinct genetic diversity patterns, and the genomic evolution underlying different patterns was more influenced by host-specific factors. In summary, a comprehensive picture of the evolutionary patterns of overall IAVs is provided by the FluTyping's identified genotypes, offering important theoretical foundations for future prevention and control of these viruses. © 2024 The Authors

NSUN2 mediates distinct pathways to regulate enterovirus 71 replication

Increasing evidences suggest that the methyltransferase NSUN2 catalyzes 5-methylcytosine (m5C) modifications on viral RNAs, which are essential for the replication of various viruses. Despite the function of m5C deposition is well characterized, other potential roles of NSUN2 in regulating viral replication remain largely unknown. In this study, the m5C modified residues catalyzed by NSUN2 on enterovirus 71 (EV71) RNAs were mapped. NSUN2, along with m5C modifications, played multiple roles during the EV71 life cycle. Functional m5C modified nucleotides increased the translational efficiency and stability of EV71 RNAs. Additionally, NSUN2 was found to target the viral protein VP1 for binding and promote its stability by inhibiting the ubiquitination. Furthermore, both viral replication and pathogenicity in mice were largely attenuated when functional m5C residues were mutated. Taken together, this study characterizes distinct pathways mediated by NSUN2 in regulating EV71 replication, and highlights the importance of its catalyzed m5C modifications on EV71 RNAs for the viral replication and pathogenicity. © 2024 The Authors

Natural vibration and critical velocity of translating Timoshenko beam with non-homogeneous boundaries

In most practical engineering applications,the translating belt wraps around two fix.d *** boundary conditions of the dynamic model are typically specified as simply supported or fix.d *** this paper,non-homogeneous boundaries are introduced by the support *** the translating belt as the mechanical prototype,the vibration characteristics of translating Timoshenko beam models with nonhomogeneous boundaries are investigated for the first *** governing equations of Timoshenko beam are deduced by employing the generalized Hamilton's *** effects of parameters such as the radius of wheel and the length of belt on vibration characteristics including the equilibrium deformations,critical velocities,natural frequencies,and modes,are numerically calculated and *** numerical results indicate that the beam ex.eriences deformation characterized by varying curvatures near the *** radii of the wheels play a pivotal role in determining the change in trend of the relative difference between two beam *** the results unearths that the relative difference in equilibrium deformations between the two beam models is more pronounced with smaller-sized *** the two wheels are of equal size,the critical velocities of both beam models reach their respective *** addition,the relative difference in natural frequencies between the two beam models ex.ibits nonlinear variation and can easily ex.eed 50%.Furthermore,as the ax.al velocities increase,the impact of non-homogeneous boundaries on modal shape of translating beam becomes more *** dealing with non-homogeneous boundaries is challenging,beam models with non-homogeneous boundaries are more sensitive to parameters,and the differences between the two types of beams undergo some interesting variations under the influence of non-homogeneous boundaries.

The H10 subtype avian influenza virus (AIV) poses an ongoing threat to both birds and humans. Notably, fatal human cases of H10N3 and H10N8 infections have drawn public attention. In 2022, we isolated two H10N3 viruses (A/chicken/Shandong/0101/2022 and A/chicken/Shandong/0603/2022) from diseased chickens in China. Genome analysis revealed that these viruses were genetically associated with human-origin H10N3 virus, with internal genes originating from local H9N2 viruses. Compared to the H10N8 virus (A/chicken/Jiangx./102/2013), the H10N3 viruses ex.ibited enhanced thermostability, increased viral release from erythrocytes, and accumulation of hemagglutinin (HA) protein. Additionally, we evaluated the pathogenicity of both H10N3 and H10N8 viruses in mice. We found that viral titers could be detected in the lungs and nasal turbinates of mice infected with the two H10N3 viruses, whereas H10N8 virus titers were detectable in the lungs and brains of mice. Notably, the proportion of double HA Q222R and G228S mutations in H10N3 viruses has increased since 2019. However, the functional roles of the Q222R and G228S double mutations in the HA gene of H10N3 viruses remain unknown and warrant further investigation. Our study highlights the potential public health risk posed by the H10N3 virus. A spillover event of AIV to humans could be a foretaste of a looming pandemic. Therefore, it is imperative to continuously monitor the evolution of the H10N3 influenza virus to ensure targeted prevention and control measures against influenza outbreaks. © 2024 The Authors

A vertical track nonlinear energy sink

Eliminating the effects of gravity and designing nonlinear energy sinks(NESs)that suppress vibration in the vertical direction is a challenging task with numerous damping *** this paper,the dynamic design of a vertical track nonlinear energy sink(VTNES)with zero linear stiffness in the vertical direction is proposed and realized for the first *** motion differential equations of the VTNES coupled with a linear oscillator(LO)are *** the strong nonlinearity considered of the VTNES,the steady-state response of the system is analyzed with the harmonic balance method(HBM),and the accuracy of the HBM is verified *** this basis,the VTNES prototype is manufactured,and its nonlinear stiffness is *** damping effect and dynamic characteristics of the VTNES are studied theoretically and *** results show that the VTNES has better damping effects when strong modulation responses(SMRs)***,even for small-amplitude vibration,the VTNES also has a good vibration suppression *** sum up,in order to suppress the vertical vibration,an NES is designed and developed,which can suppress the vertical vibration within certain ranges of the resonance frequency and the vibration intensity.

The deep structure,material circulation,and dynamic processes in the Southeast Asia have long been an elusive scientific puzzle due to the lack of systematic scientific observations and recognized theoretical *** on the deep seismic tomography using long-period natural earthquake data,in this study,the deep structure and material circulation of the curved subduction system in Southeast Asia was studied,and the dynamic processes since 100 million years ago was *** is pointed out that challenges still ex.st in the precise reconstruction of deep mantle structures of the study area,the influence of multi-stage subduction on deep material ex.hange and shallow magma activity,as well as the spatiotemporal evolution and coupling mechanism of multi-plate *** work should focus on high-resolution land-sea joint 3-D seismic tomography imaging of the curved subduction system in the Southeast Asia,combined with geochemical analysis and geodynamic modelling works.

Towards human-leveled vision systems

The human visual system is a complex.and interconnected network comprising billions of neurons. It plays an essential role in translating environmental light stimuli into information that guides and shapes human perception and action. Research on the visual system aims to uncover the underlying neural structure principles of human visual perception and their possible ***, there are two main approaches: biological system analysis and simulation, artificial intelligence models based on deep learning. Here we aim to discuss the two approaches to human-level vision systems. Deep learning has significantly impacted the field of vision with achievements in representation, modeling, and hardware design. However, there is still a significant gap between deep learning models and the human visual system in terms of scalability, transferability, and sustainability. The progress of the biological visual system can help fill the gap by further understanding the properties and functions of different components of the system. We take the efforts of reconstructing the retina as an ex.mple to illustrate that even if we are unable to replicate the visual system on a computer right now, we can still learn a lot by combining ex.sting research outcomes in neuroscience. At the end of the paper, we suggest tracing back to gradually build visual systems from the computational counterpart of biological structures to achieve a human-level vision system in the future.
