Hypoxia is a common phenomenon in hepatocellular carcinoma （HCC）. Hypoxia stabilizes transcription factor, hypoxia-inducible factor （HIF）, to activ.te gene transcription. Expression of HIF is closely associated with metastasis and poor prognosis in HCC. HIF mediates expression of genes that are inv.lv.d in ev.ry step of HCC metastasis including epithelial-mesenchymal transition, inv.sion of the extracellular matrix, intrav.sation, extrav.sation, and secondary growth of the metastases. Because HIF is the central regulator of HCC metastasis, HIF inhibitors are attractiv. tools when used alone or as combined treatment to curb HCC metastasis. This rev.ew will summarize the current findings on the impact of hypoxia/HIF in HCC, with a particular focus on cancer metastasis.

A pulsed,picosecond Nd:YAG laser with a wav.length of 532 nm is used to texture the surface of grade 5 titanium alloy(Ti–6Al–4v.for minimizing its wear *** wear properties of the base samples and laser surface textured samples are analyzed by conducting wear tests under a sliding condition using pin-on-disk *** wear tests are conducted based on the Box–Benhken design,and the interaction of the process parameters is analyzed using response surface *** wear analysis is conducted by v.rying the load,rotating speed of the disc,and track diameter at room temperature with a sliding distance of 1500 *** results demonstrate that the laser textured surfaces exhibited a lower coefficient of friction and good anti-wear properties as compared with the non-textured surfaces.A regression model is dev.loped for the wear analysis of titanium alloy using the analysis of v.riance *** is also observ.d from the analysis that the applied load and sliding distance are the parameters that hav. the greatest effect on the wear behav.or followed by the wear track *** optimum operating conditions hav. been suggested based on the results obtained from the numerical optimization approach.

AIM Influence of chronic hyperglycemia on chemical coding of enteric neurons in stomach using pig as a model for human diabetic *** Ten pigs were div.ded into two groups: diabetic(D group,n = 5) and control(C group,n = 5). Pigs constituting the experimental group were giv.n streptozotocin(150 mg/kg). Animals were euthanized six weeks after the induction of diabetes. The samples of stomach were collected from animals of both groups. The cryostat sections were processed for double immunofluorescence staining using primary antisera directed towards pan-neuronal marker(Hu C/D) proteins and/or neuronal isoform of nitric oxide synthase(n NOS),v.soactiv. intestinal peptide(v.P) and galanin(GAL). RESULTS In the control group in the myenteric ganglia(MG) of the corpus we hav. noted 22.28% ± 1.19% of n NOS positiv. neurons,while in diabetic group we hav. found 40.74% ± 2.22% of n NOS immunoreactiv. perikarya(increase by 82.85 %). In turn in the pylorus we hav. observ.d 15.91% ± 0.58% n NOS containing neurons in control animals and 35.38% ± 1.54% in the diabetes group(increase by 122.37%). In the MG of the antrum and submucosal ganglion(SG) in the corpus hyperglycemia did not cause statistically significant changes. With regard to v.P-positiv. cell bodies in the antrum MG in the control animals we hav. noted 18.38 ± 1.39% and 40.74% ± 1.77% in the experimental group(increase by 121.65%). While in the corpus we hav. observ.d 23.20% ± 0.23% in the control and 30.93% ± 0.86% in the diabetes group(increase by 33.31%). In turn in the pylorus v.P positiv. cells bodies constituted 23.64% ± 1.56% in the control group and 31.20% ± 1.10% in the experimental group(increase by 31.97%). In the submucosal ganglion in the corpus we hav. noted 43.61% ± 1.06% in the control animals and 37.00% ± 1.77% in the experimental group(decrease by 15.15%). A systematic study has been made on the behav.or of Al/n-CdS thin film junction on flexible polymer substrate(polyethylene terephthalate, PET)grown using thermal ev.poration method. Temperature dependence of Ⅰ-Ⅴ measurements for this junction has been done which closely follow the equations of Schottky barrier junction dominated by thermionic emission mechanism. Intrinsic and contact properties such as barrier height, ideality factor and series resistance hav. been calculated from Ⅰ-Ⅴ characteristics. The barrier height of Al/n-CdS junction is found to increase with increase in temperature whereas ideality factor and series resistance decrease with increase in temperature.

A highly selectiv. and sensitiv. liquid chromatography-tandem mass spectrometry （LC-MS/MS） assay has been described for the determination of asenapine （ASE） in presence of its inactiv. metabolites N-desmethyl asenapine （DMA） and asenapine-N-glucuronide （ASG）. ASE, and ASE 13C-d3, used as internal standard （IS）, were extracted from 300 μL human plasma by a simple and precise liquid-liquid extraction procedure using methyl tert-butyl ether. Baseline separation of ASE from its inactiv. metabolites was achiev.d on Chromolith Performance RPse （100 mm ×4.6 mm） column using acetonitrile- 5.0 mM ammonium acetate-10% formic acid （90：10：0.1, v.v.v. within 4.5 min. Quantitation of ASE was done on a triple quadrupole mass spectrometer equipped with electrospray ionization in the positiv. mode. The protonated precursor to product ion transitions monitored for ASE and ASE 13C-d3 were m/z 286.1 → 166.0 and m/z 290.0 → 166.1, respectiv.ly. The limit of detection （LOD） and limit of quantitation （LOQ） of the method were 0.0025 ng/mL and 0.050 ng/mL respectiv.ly in a linear concentration range of 0.050-20.0 ng/mL for ASE. The intra-batch and inter-batch precision （% Cv. and mean relativ. recov.ry across quality control lev.ls were 〈 5.8% and 87.3%, respectiv.ly. Matrix effect, ev.luated as IS-normalized matrix factor, ranged from 1.03 to 1.05. The stability of ASE under different storage conditions was ascertained in presence of the metabolites. The dev.loped method is much simpler, matrix free, rapid and economical compared to the existing methods. The method was suc- cessfully used for a bioequiv.lence study of asenapine in healthy Indian subjects for the first time.

A selectiv., sensitiv. and rugged liquid chromatography–tandem mass spectrometry(LC–MS/MS) assay has been dev.loped for the simultaneous determination of doxepin(Dox) and its pharmacologically activ. metabolite, nordoxepin(NDox) in human plasma. The analytes and their internal standards(IS)were extracted from 500 m L of human plasma by liquid-liquid extraction using methyl tert-butyl *** separation was achiev.d on Hypurity C8 column(100 mm ? 4.6 mm, 5 mm) using a mixture of acetonitrile-methanol(95:5, v.v. and 2.0 mM ammonium formate in 93:7(v.v. ratio. Detection was accomplished by tandem mass spectrometry in the positiv. ionization and multiple reaction monitoring acquisition mode. The protonated precursor to product ion transitions studied for Dox, NDox,and their corresponding ISs, propranolol and desipramine, were m/z 280.1-107.0, 266.0-107.0,260.1-116.1 and 267.1-72.1, respectiv.ly. A linear dynamic range of 15.0–3900 pg/mL for Dox and 5.00–1300 pg/mL for NDox was established with mean correlation coefficient(r2) of 0.9991 and 0.9993, respectiv.ly. The extraction recov.ry ranged from 86.6%–90.4% and 88.0%–99.1% for Dox and NDox, respectiv.ly. The intra-batch and inter-batch precision(% Cv. across quality control lev.ls was r 8.3% for both the analytes. Stability ev.luated under different storage conditions showed no ev.dence of degradation and the % change in stability samples compared to nominal concentration ranged from 4.7% to12.3%. The method was successfully applied to a bioequiv.lence study of 6 mg doxepin hydrochloride orally disintegrating tablet in 41 healthy Indian subjects under fasting and fed conditions.
