Low-density lipoprotein receptor genetic polymorphism in chronic hepatitis c virus Egyptian patients affects treatment response

AIM: To correlate a genetic polymorphism of the low-density lipoprotein(LDL) receptor with antiviral responses in Egyptian chronic hepatitis c virus(HcV) ***: Our study included 657 HcV-infected patients with genotype 4 who received interferonbased combination therapy. Patients were divided into two groups based on their response to therapy: 356 were responders, and 301 were non-responders. Patients were compared to 160 healthy controls. All patients and controls underwent a thorough physical examination, measurement of body mass index(BMI) and the following laboratory tests: serum alanine aminotransferase, aspartate aminotransferase, alkaline phosphatase, albumin, total bilirubin, direct bilirubin, prothrombin time, prothrombin concentration, INR, complete blood count, serum creatinine, fasting blood sugar, HcV antibody, and hepatitis B surface antigen. All HcV patients were further subjected to the following laboratory tests: HcV-RNA using quantitative polymerase chain reaction(PcR), antinuclear antibodies, thyroid-stimulating hormone, an LDL receptor(LDLR) genotype study of LDLR exon8 c.1171G>A and exon-10 c.1413G>A using real-time PcR-based assays, abdominal ultrasonography, ultrasonographic-guided liver biopsy, and histopathological examination of liver biopsies. correlations of LDL receptor polymorphisms with HAI, METAVIR score, presence of steatosis, and BMI were performed in all ***: There were no statistically significant differences in response rates between the different types of interferon used or LDLR exon10 c.1413G>A. However, there was a significant difference in the frequency of the LDL receptor exon8 c.1171G>A genotype between cases(AA: 25.9%, GA: 22.2%, GG: 51.9%) and controls(AA: 3.8%, GA: 53.1% and GG: 43.1%)(P cally significant difference in the frequency of the LDLR exon 8c:1171 G>A polymorphism between responders(AA: 3.6%, GA: 15.2%, GG: 81.2%) and nonresponders(AA: 52.2%, GA: 30.6%, GG: 17.

BAcKGROUND childhood cancer survivors are potentially at a higher risk of infection with hepatitis c virus (HcV).The effects of all-oral direct-acting antiviral therapy (DAA) on both the HcV infection as well as the state of cancer remission have not been well investigated in this *** To test the effects of dual sofosbuvir/daclatasvir (SOF/DcV) therapy in the treatment of chronic HcV in survivors of hematologic malignancy in pediatric age *** We conducted a prospective,uncontrolled,open-label multicenter study.A total of 20 eligible,chronic HcV,genotype-4,infected children who had been in continuous complete remission from hematologic cancer (leukemia/lymphoma) for at least one year were included in the *** patients were treated with combined SOF/DcV for 12 *** were monitored throughout the study till 12 wk after end of treatment for safety and efficacy outcomes including the sustained virologic response 12 (SVR12) rate,hematological indices,liver and kidney *** The intent-to-treat SVR12 rate was 20 of 20 (100%;95%cI: 84%-100%).All patients showed normalized liver enzymes from *** hematological indices,liver and kidney functions were kept normal throughout the *** fatalities or treatment-emergent serious or severe adverse events were reported throughout the *** SOF/DcV combined therapy could be used safely and effectively in the treatment of chronic HcV genotype-4 infection in leukemia/lymphoma treated *** relapses were detected during treatment and throughout the follow up period for either the original malignant disease or the HcV infection.

The Antiviral Efficacy of Withania somnifera(Ashwagandha) against Hepatitis c Virus Activity: In Vitroand in SilicoStudy

Objective: Evaluation antiviral effects of Withania somnifera (Ashwagandha) leaf extract against HcV. Methods: cell proliferation was assessed using MTT assay after isolation of lymphocyte cells and treated with Ashwagandha water extract (ASH-WX) (6.25 mg/ml - 100 mg/ml). Assessment of quantitative Real-time PcR, colony forming assay, TNF-α and molecular docking studies after infection of normal lymphocyte cells with 1 ml (1.5 × 106 HcV) serum then incubated with ASH-WX at concentration 25 mg/ml & 50 mg/ml. Results: MTT assay revealed a significant increase (p cyte proliferation at all concentration’s particularity at 25 mg/ml with SI (6.06) and at 50 mg/ml with (5.8). While TNF-α significantly decreased following ASH-WX treatment compared with control untreated infected cells (p cR results showed a marked viral load reduction after treatment by ASH-WX at concentration 25 mg/ml to 6.241 × 103 IU/mL. colony formation assay test revealed colony formation reduction compared to positive untreated control. Molecular docking analysis revealed good prediction of binding between Ashwagandha and NS5B and PKN2 compared to Sovaldi. conclusion: ASH-WX may be a powerful antiviral against HcV infection.

Highly efficient syntheses of novel fluorine bearing quinoline-4-carboxylic acids and the related compounds had been achieved from cyclocondensation of 2-amino-5-fluorophenyl glyoxylic acid 1 with benzoyle asetanilides 2 in boiling DMF, to give the target 3. Decarboxylation of 3 produced 6-fluoro-2-phenyl-3-(substituted amino)-keto-quinolines 4, while that reaction underwent refluxing, afforded 7-fluoro-1-(aryl)-3-phenyl-pyrrolo[3,4-c] quinoline-2,9-diones 5. Structure of the products has been established from their elemental and spectral analysis. All targets exhibited a high to moderate activity against some Aspergillus fungi as amylolytic agents.

Association of Hepatitis c Virus Infection and Type 2 Diabetes in Egypt: A Hospital-Based Study

Background: The relation between hepatitis c virus (HcV) infection and the development of type 2 diabetes mellitus (T2DM) remains unknown. The aim of this study was to investigate the presence of an association between T2DM and HcV. Methods: A case control study was conducted at the outpatients’ clinics of Kasr El-Aini Hospital (KAH), from October 2013 till March 2014. In this study, 389 HcV patients were selected as cases and 389 healthy controls were also included. Demographic and clinical data were collected using a structured questionnaire. Laboratory investigations including liver function tests (LFT), blood glucose level and radioimmunoblot assay (ELISA) were performed. Results: Out of 389 HcV cases, 219 (56.3%) were diabetic, whereas 145 (37.3%) were diabetic among the healthy controls. Occurrence of diabetes among cirrhotic patients was 1.7 times higher than non-cirrhotic. Logistic regression showed that residence (P ccupation (P = 0.03, OR = 1.8), and smoking (P = 0.04, OR = 2) were the predictive factors for occurrence of T2DM in HcV patients. conclusions: In this study, we found a positive association between HcV infection and T2DM. Residence, occupation and smoking were the predictive factors for the association of T2DM in HcV patients, whereas hypertension and BMI were only adjunctive factors.

The combination of endoglin and FIB-4 increases the accuracy of detection of hepatic fibrosis in chronic hepatitis c patients

Background and aim: In patients infected with chronic hepatitis c virus, liver biopsy is the gold standard method of staging fibrosis. Different combinations of serum markers attempted to correlate with hepatic fibrosis in place of liver biopsy and have shown encouraging results. The aim of our study is to evaluate the diagnostic value of endoglin and FIB-4 as non-invasive markers of hepatic fibrosis in HcV patients. Methods: We estimated serum endoglin & FIB-4 index in 40 infected chronic hepatitis c patients. Histological staging of hepatic fibrosis was done according to the METAVIR scoring system. Results: Both endoglin and FIB-4 index showed positive correlation with age and aspartate transaminase and inverse correlation with albumin. The diagnostic performance determined by AUROcs for early fibrosis (≤F2), was 0.868 for endoglin and 0.887 for FIB-4, at cut off va- lues of 5.5 & 0.98 with sensitivity of 64.3% & 82.1%, and specificity of 100% & 85% respectively. For ad-vanced fibrosis (>F2), the AUROc was 0.98 for endoglin and 0.967 for FIB-4, obtained at cut off values of 6.29 & 1.6, with sensitivity of 100% & 91.7%, and specificity of 89.3% & 92.9%, respectively. conclusion: Both serum endoglin and FIB-4 index are fairly accurate in differentiating stages of hepatic fibrosis;their combination in a single equation enhanced the accuracy of fibrosis detection in chronic HcV patients.

Introduction: Rotavirus infection is a leading cause of severe diarrhea culminating to dehydration among children under five years of age. Understanding trends and factors that could assist towards devising effective preventive strategies of Rotavirus infection beyond vaccination is crucial. Objectives: This study was done in an attempt to determine the prevalence and associated factors of Rotavirus infection among vaccinated children aged between 6 weeks and 24 months admitted with acute diarrhea Mwanza, Tanzania. Material and Methods: Across sectional study involving vaccinated children aged 6 weeks to 24 months was conducted in three selected hospitals from July 2017 to January 2018. Socio-demographic and other relevant clinical information were collected using a standardized data collection tool adopted from WHO Rotavirus surveillance tool. Rotavirus infection from the stool was detected using an enzyme immunoassay. Data were analyzed using STATA version 13. Results: A total of 301 vaccinated children with acute diarrhea with a median age of 12 [IQR: 8 - 17] months were enrolled. Nine (3.0%) and 292 (97.0%) had received one dose and two doses of Rotavirus vaccine, respectively. The prevalence of Rotavirus infection was 74 (24.6%) [95% CI: 20.0 - 29.8]. A new biocidal agents fluorine substituted-3-thioxopyrimidine-5-carbonitriles (2-9) and/or the related fluorine substi- tuted pyrimido (4,5-d) pyrimidines (10-14) were synthesized by the cycloaddition of fluorinated β- arylidine malo- nonitriles (1a-c) followed by a nucleophilic attack against α,β-bifunctional reagents in different conditions. Structures of the fluorine targets were characterized by their elemental analysis and spectral data (UV, IR, 1H NMR, 13c NMR and mass measurements) and further evaluated as photochemical probe for inhibition of Vitiligo, it was found that compounds 5, 9, 11 and 12 exhibited high potency over the investigated compounds.

In response to the global food crisis and the imperative to address soil degradation, the international agricultural policy is actively working to alleviate the adverse impacts of soil salinity. As part of this initiative, a field trial spanning two consecutive seasons (2019/20-2020/21) was conducted under saline conditions. The primary objective was to evaluate the influence of various compost sources, including vermicompost at a rate of 0.5 ton·fed-1 and plant residues compost at a rate of 5.0 ton·fed-1, as main plots. Subplots were established by applying agricultural gypsum, both in the presence and absence of gypsum requirements. Additionally, sub-subplots were created by externally applying cobalt at a rate of 10.0 mg·L-1, with one sub-subplot receiving foliar cobalt application and the other not. The trial sought to assess the growth performance, chemical composition, enzymatic antioxidants, yield, and quality of cabbage plants (Brassica oleracea var. capitata L.) cultivated in saline soil. According to the findings, cabbage plants exhibited the most favorable response in terms of plant height, chlorophyll content, carotene levels, leaf area, nitrogen (N), phosphorus (P), potassium (K), head yield, vitamin c, and total dissolved solids (TDS) when treated with vermicompost, followed by plant compost. conversely, plants grown without compost exhibited the least improvement in performance. cabbage treated with agricultural gypsum requirements showed better performance than those without gypsum amendment. Moreover, plants subjected to cobalt spray demonstrated the highest growth, yield, and quality parameters compared to those without cobalt foliar application. In contrast, the control group (plants without the studied treatments) displayed the highest levels of enzymatic antioxidants, specifically catalase and peroxidase. This indicates that soil salinity stress led to an increase in catalase and peroxidase production in cabbage
