The genus Echeveria has about 143 species that are unique to the Americas, 117 of which are presented in the mexican Republic, mainly in the states of Hidalgo, Puebla and Oaxaca. Propagation can be done by cuttings, leaf cuttings or seeds, but these methods are insufficient to avoid the predation of many species. NOm-059-ECOL-2001 shows Echeveria elegans as species reported endangered. The objectives in this study were achieved in vitro propagation from axillary buds, as an alternative to multiplication and preservation species. We evaluated different concentrations and combinations of two growth regulators, 6-BAP （2.22, 4.44, 6.66 and 8.88 μm） and u-NAA （1.35 and 2.70 μm） on shoot formation from axillary buds and two culture media with different concentrations of mS salts, to achieve root plants grown in vitro. It was found by combining 6.66 μm of 6-BAP and 1.35μm of α-NAA favored the formation and development of new shoots. In the rooting stage, the best results are achieved with the treatment containing 50% of mS salts. Then the plants were transplanted into greenhouses and achieved a successful acclimatization. During this last phase of development, there were no phenotypic changes or presence of somaclonal variants.

Objective: To evaluate the role of regulatory T cells (Tregs) at late stages of stroke. methods:Anti-CD25 antibody (or PBS as a control) was injected to reduce the pool of Tregs in Wistar rats;then, ischemia was induced transiently by middle cerebral artery occlusion during 60 min and reperfusion was allowed for 7 d. Then, Treg frequency was analyzed in peripheral blood, spleen and lymph nodes. Neurological score (0-6) and infarct volume were also determined. Results: Nine days after injection, the CD4+CD25+ T cells were reduced by 70.4%, 44.8% and 57.9% in peripheral blood, spleen and lymph nodes, respectively compared to PBS-treated rats. In contrast, the reduction of CD4+FOXP3+ T cells was lower in the same compartments (38.6%, 12.5%, and 29.5%, respectively). The strongest reduction of CD25+CD4+ T cells was observed in those FOXP3-negative cells in blood, spleen and lymph nodes (77.8%, 52.8%, and 60.7%, respectively), most likely corresponding to activated T cells. Anti-CD25-treated transient middle cerebral artery occlusion rats had a lower neurological deficit and did not develop tissue damage compared with PBS-treated animals. Conclusions: These findings suggest that treatment with anti-CD25 in our model preferentially reduce the T cell population with an activated phenotype, rather than the Treg population, leading to neuroprotection by suppressing the pathogenic response of effector T cells.

In this work, a method for quantification of heavy metals Inductively Coupled Plasma Sector Field mass Spectrometry (ICP-SFmS) in soil samples of El Bierzo district (Spain) has been optimized and validated. Optimization was carried out for elements: Cr, mn, Co, Ni, Cu, Zn, Se, As, Cd, Hg, Pb and U. Validation of the method was performed with Certified and Standard Reference materials (CRms and SRms);SRm2709, CRm020-051 and CRm050-051. Results obtained under optimized conditions can be summarized as follows: a) the Limits of Detection (LODs) were in the order of sub fg·g-1 for Cr, mn, Cu, Co, As, Cd, Hg, Pb and U, and few fg·g-1for Ni, Zn and Se;b) precision measurement, in terms of relative standard deviation (RSD), was been below 5%;c) the average recovery of CRm was between 81.3% and 98%. In conclusion, the method offers several advantages: fast, good accuracy, very low values of Limits of Quantification (LOQs) and high sensitivity on measurement of heavy metal.

Helplessness-like escape deficits of NIH-HS rats predict passive behavior in the forced swimming test:Relevance for the concurrent validity of rat models of depression

The genetically heterogeneous NIH-HS rat stock has been characterized by its response to anxiety- and fear-inducing situations, thus leading to the conclusion that they are a rather anxious and passive coping type of rats. Taking advantage of these profiles, and knowing that they show very poor performance in the two-way active (shuttle box) escape/avoidance task, we have tested NIH-HS rats (n = 80) in the forced swimming test (FST) as well as we have studied escape response deficits (i.e. response failures) of the same animals in the two-way shuttle box task. They were also tested for anxiety in the elevated zero-maze. The goal of such a study was that of investigating whether there are associations or relationships among helplessness-like or passive coping responses between both models of depression, i.e. the FST and the helplessness-like escape deficits in the shuttle box task. The results for the first time show associations among responses from both depression models and that selecting rats for displaying extreme (active or passive) responses in one of the models predict in a coherent manner (according to the hypothesis) their behaviour in the other model. These findings are discussed in the context of the concurrent validity of both models of depression as well as concerning the possible relevance of NIH-HS rats as a tool for future studies on this field.

The “National Institutes of Health” genetically heterogeneous (NIH-HS) rat stock was created in the 1980s through an eight-way cross of as much as possible separate inbred rat strains (i.e. the mR/N, WN/N, WKY/N, m520/N, F344/N, ACI/N, BN/SsN and BUF/N strains) which were readily available at that time. Hansen and Spuhler [1] developed a more naturalistic, genetically heterogeneous rat stock with the aim of optimizing the distribution of genotypic frequencies and recombination and under the hypothesis that the NIH-HS stock could yield a broad-range distribution of responses (broader than commonly used laboratory rat strains) to experimental conditions, and thus serve as a base population for selection studies. Along the last decade, in a series of studies we have phenotypically characterized the NIH-HS rat stock (a colony exists at our laboratory since 2004) for their anxiety/fearfulness profiles (using a battery of both unconditioned and conditioned tests/tasks), as well as regarding their stress-induced hormonal responses, coping style under inescapable stress and spatial learning ability. We have also compared the phenotypic profiles of NIH-HS rats with those of the low anxious RHA-I and the high anxious RLA-I rat strains. The NIH-HS rat stock is, as a population, a rather anxious type of rat, with predominantly reactive/passive coping style in unlearned and learned anxiety/fear tests, and elevated stress hormone responses (as well as enhanced “depressive” symptoms in the forced swimming test). Genetic studies currently under way have thus far revealed that the genetically heterogeneous NIH-HS rat stock constitutes a unique tool for fine mapping of QTL (for multiple behavioural and biological complex traits) to megabase resolution levels, thus enabling candidate gene identification. Introduction: In bile duct injuries (BDI), cholestasis and cholangitis can alter the fibrinolytic system by promoting an increase of extracellular matrix depositions which favor an imbalance between metalloproteinases (mmPs) and their tissue inhibitors (TImPs). materials and methods: Levels of PAI-1, mmP-3, mmP-8, TImP-1 and TImP-2 in 35 patients with post-cholecystectomy BDI by complete biliary obstruction were measured and compared to a healthy control group. Sirius red staining and immune staining for mmP-3 and mmP-8 were also undertaken in liver biopsies. Results: Levels of PAI-1, TImP-1, TImP-2 and mmP-8 were higher in BDI than healthy controls: 15 ± 2 ng/mL vs 7.1 ± 2 ng/mL (p 0.024);539 ± 64 ng/mL vs 256 ± 13 ng/mL (p p p 2 vs. 22865.7 ± 3865 μm2 in healthy controls (p 2 vs. 30744.2 ± 5810.2 μm2 (p 2 vs. 116337.9 ± 24803.3 μm2 (p 0.55). These results suggest an imbalance between fibrogenic/fibrinolytic protein levels. Interestingly, expression of the fibrinolytic protein mmP-8 was increased in serum and liver biopsies in BDI. Conclusion: We found an imbalance of profibrogenic molecules which promote extracellular matrix deposition. The over-expression of fibrinolytic proteins such as mmP-8 could limit liver fibrosis, preventing hepatic dysfunction in post-cholecystectomy BDI.
