AIM： To investigate the non-he/icobacterpy/ori （H. pylori） bacterial flora concurrent with H. pylori infection.METHODS： A total of 103 gastric biopsy specimens from H. pylori positive patients were selected for bacterial culture. All the non-H, pylori bacterial isolates were identified by matrix assisted laser desorption ionization time-of-flight mass spectrometry （MALdi-TOF MS）.RESULTS： A total of 201 non-H, pyiori bacterial isolates were cultivated from 67 （65.0%） of the 103 gastric samples, including 153 isolates identified successfully at species level and 48 at genus level by MALdi-TOF MS. The dominant species were Streptococcus, Neisseria, Rothla and Staphylococcus, which differed fromthe predominantly acid resistant species reported previously in healthy volunteers. The prevalence of non-H. pylori bacteria was higher in non-ulcer dyspepsia group than in gastric ulcer group （100% vs 42.9%, P 〈 0.001）. Six bacterial species with urease activity （Staphylococcus epidermidis, Staphylococcus warneri, Staphylococcus capitis, Staphylococcus aureus, Brevibacteriurn spp. and Klebsiella pneumoniae） were also isolated.CONCLUSION： There is a high prevalence of the non-H, pylori bacteria concurrent with H. pylori infection, and the non-H, pylori bacteria may also play important as-yet-undiscovered roles in the pathogenesis of stomach disorders.

AIM： To investigate whether anti-H pylori antibodies have cross-reaction with antigens of erythrocyte membrane. METHODS： Blood samples were collected from 14 volunteers （8 positive and 6 negative for Hpylori detected by ^13C-urea breath test） of the general population. Erythrocyte membrane proteins of the subjects were examined by Western blot using anti-H pylori serum. The proteins related to the positive bands were identified by mass spectrum analysis. RESULTS： Anti-Hpylori antibodies had cross-reaction with the proteins of about 50 kDa of erythrocyte membranes in all samples independent of H pylori infection. One protein in the positive band was identified as Chain S, the crystal structure of the cytoplasmic domain of human erythrocyte Band-3 protein. CONCLUSION： Anti-HpyloH antibodies cross-react with some antigens of human erythrocyte membrane, which may provide a clue for the relationship between Hpylori infection and vascular disorders.

Objective During 2003-2005, an outbreak of meningitis due to Neisseria meningitidis serogroup C occurred in China. With the aim to find strain clues result in the final epidemics, the ancestral strain 053442, a clinical isolate, and a carrier strain 053426 with different gene type were analyzed. Methods Clinical strain 053442 and carrier strain 053426 were cultured on GC agar plates under the same condition. Two-dimensional electrophoresis was performed using the pH 3–10 nonlinear IPG strips of 24 cm length, and all the protein spots were identified by matrix-assisted laser desorption/ionization time of flight spectrometry. Results 502 and 380 protein spots were identified in 053426 and 053442 respectively, relating to 266 and 202 different genes covering a wide range of cellular functions. The express volume and number of proteins involved in energy metabolism, protein synthesis and amino acid biosynthesis in 053426 were higher than in 053442. Virulence factor Opa, Opc and a series of proteins involved in pilus assembly and retraction were identified in 053442, which appear to be of primary importance in colonization and invasion of human cells. Compared to 053442, virulence protein species were less in 053426, with lower express volumes too. No Opa and Opc were detected in 053426. Conclusion The different protein expression profiles of the clinical strain 053442 and carrier strain 053426 in the present study provide some clues of the different pathogenicity of the two strains, which may account for result in the final epidemics.

The Large sky Area Multi-Object Fiber Spectroscopic Telescope（LAMOST） general survey is a spectroscopic survey that will eventually cover approximately half of the celestial sphere and collect 10 million spectra of stars, galaxies and QSOs. Objects in both the pilot survey and the first year regular survey are included in the LAMOST DR1. The pilot survey started in October 2011 and ended in June 2012, and the data have been released to the public as the LAMOST Pilot Data Release in August 2012. The regular survey started in September 2012, and completed its first year of operation in June 2013. The LAMOST DR1 includes a total of 1202 plates containing 2 955 336 spectra, of which 1 790 879 spectra have observed signalto-noise ratio（SNR） ≥ 10. All data with SNR ≥ 2 are formally released as LAMOST DR1 under the LAMOST data policy. This data release contains a total of 2 204 696 spectra, of which 1 944 329 are stellar spectra, 12 082 are galaxy spectra and 5017 are quasars. The DR1 not only includes spectra, but also three stellar catalogs with measured parameters： late A,FGK-type stars with high quality spectra（1 061 918 entries）, A-type stars（100 073 entries）, and M-type stars（121 522 entries）. This paper introduces the survey design, the observational and instrumental limitations, data reduction and analysis, and some caveats. A description of the FITS structure of spectral files and parameter catalogs is also provided.

目的研究幽门螺杆菌（helicobacter pylori）作用后人胃腺癌上皮细胞（AGS）中微量磷酸化蛋白的变化情况。方法采用金属离子亲和吸附富集技术富集幽门螺杆菌与AGS细胞相互作用以及AGS细胞的磷酸化蛋白，除盐纯化后利用二维凝胶电泳技术分离磷酸化蛋白，MALdi—TOF／TOF质谱鉴定确认蛋白。结果幽门螺杆菌作用后AGS细胞有21种蛋白出现差异表达，其中15种蛋白表达量下调，4种蛋白出现新表达，1种蛋白表达量上调，1种蛋白不表达。差异表达蛋白涉及细胞的钙离子调节、转录、翻译、蛋白折叠与运输、核糖体的装配、中心体复制、染色体稳定、细胞结构形态、细胞增殖与凋亡等。结论幽门螺杆菌能引起广泛的胃腺癌黏膜上皮细胞蛋白磷酸化特征的变化，这种变化谱特征对进一步全面认识幽门螺杆菌的致病作用有重要意义。

目的:分析幽门螺杆菌(Hpylori)细胞毒素相关蛋白A(CagA)对人胃腺癌黏膜上皮细胞(AGS)Ca2+相关蛋白磷酸化的影响,进一步揭示Hpylori的致病机制.方法:采用金属离子亲和吸附富集技术富集Hpylori、Hpylori CagA缺失株(Hpylori△CagA)与AGS细胞相互作用4h,以及培养相同时间的AGS细胞的磷酸化蛋白,利用二维凝胶电泳技术分离磷酸化蛋白,ImageMaster 2D分析软件识别差异蛋白,MALdi-TOF/TOF质谱鉴定确认蛋白.结果:Hpylori△CagA作用的AGS细胞,与培养相同时间的AGS细胞比较表达量不变,而Hpylori△CagA作用的AGS细胞表达量发生了明显变化,表明此蛋白的变化是单纯由CagA引起的;此类蛋白点共鉴定出19个,其中3个蛋白点与Ca2+相关.钙离子结合蛋白(nucleobindin-2 precursor,CALNUC)在AGS细胞以及Hpylori△CagA与AGS相互作用的2-D胶中表达量接近,而Hpylori与AGS相互作用后该蛋白表达量明显降低.结论:Hpylori CagA进入AGS细胞可能会影响内质网、线粒体及高尔基体的钙稳态,诱发内质网、线粒体、高尔基体凋亡或增殖途径,而成为胃炎、胃溃疡、胃癌发生的诱因之一.

目的:分析幽门螺杆菌(Hpylori)与人胃腺癌上皮细胞(humangastric adenocarcinoma epithelial,AGS)的相互作用蛋白质组学差异.方法:采用双向凝胶电泳分离Hpylori26695株与AGS细胞相互作用0.5、2、4h时间点的细胞和黏附菌的全蛋白样品,考马斯亮兰染色,ImageMster2D图像分析软件比较分析,识别差异蛋白,将差异蛋白点进行胶内酶解,经基质辅助电离解析飞行时间质谱(MALdi-TOF-TOF/MS)获得肽质量指纹图谱(PMF),通过搜寻蛋白质数据库完成对差异蛋白的鉴定.结果:对0.5、2、4h时间点图谱差异蛋白点分析共发现主要差异蛋白点66个,对其中34个(共对应16种蛋白)最终完成准确鉴定,2h后8种蛋白开始上调,其中2种是细胞来源,6种Hpylori来源;4h时4种细胞来源蛋白下调,并有4种新的蛋白开始被观察到.在共感染的早期,细胞的琥珀酸脱氢酶、Hpylori的抗氧化酶类、黏附素以及HSP60表达上调.感染晚期主要是细胞来源的亲环素A、新生多肽相关复合物α多肽开始表达,Hpylori来源的尿素酶、非血红素铁蛋白等蛋白明显出现在黏附菌成分中.结论:Hpylori与AGS细胞相互作用过程中,细菌和细胞蛋白表达均存在明显时序性变化,早期相互作用则主要表现为以与黏附等有关的蛋白表达变化,后期向有利细菌存活和增殖的方向发展,并呈现出与免疫逃逸和病理损伤相关的变化.

目的构建空肠弯曲菌omp18基因的重组表达质粒、克隆表达rOMP18重组蛋白、对表达产物进行鉴定并评价其抗原性。方法 PCR扩增空肠弯曲菌的omp18基因片段,将其连接到表达载体pET32a(+)中并转化至大肠杆菌(***-liBL21(DE3)),诱导表达后SDS-PAGE分析其表达产物并纯化目的蛋白。应用空肠弯曲菌感染免疫血清,利用Western-blot方法分析其抗原性。结果与结论单、双酶鉴定结果显示已成功构建rOMP18的重组表达载体pET32a-omp18,IPTG诱导表达产物的相对分子质量与理论相符34kD(HIS16kD)。Western-blot检测结果表明重组蛋白rOMP18具有抗原性,为空肠弯曲菌感染的特异抗体的检测提供候选抗原。
