Role of fetuin A in the diagnosis and treatment of joint arthritis

Osteoarthritis is a slowly progressive disease which includes the intervention of several cytokines, macrophage metalleinoproteinases reaction, leading to the degradation of the local cartilage but also having an impact on the serum acute phase proteins(APPs). subsequently, biomarkers seem to be essential to estimate its progression and the need for any surgical intervention such as total arthroplasty, but also can be used as therapeutic agents. Recently, among APPs, fetuin A drew attention regarding its possible anti-inflammatory role in animal models but also as a therapeutic agent in the inflammatory joint disease in clinical trials. In contrast with other APPs such as C-reactive protein, fetuin A appears to be lower in the serum of patients with degenerative joint disease in comparison with the healthy ones, and also acts as an antagonist of the anti-proliferative potential of transforming growth factor-b(TGF-b) cytokines. Because of its lower serum levels in arthritis, an unregulated binding of TGF-b and bone morphogenetic proteins takes place leading to further arthritic lesions. The purpose of the present review is to assess the current evidence regarding the multipotent role of the alpha-2-Hsglycoprotein or as also known Fetuin-a in animal models but also as a biomarker of the degenerative joint arthritis in clinical trials.

Multimodal imaging of experimental choroidal neovascularization

AIM:To compare choroidal neovascularization(CNV)lesion measurements obtained by in vivo imaging modalities,with whole mount histological preparations stained with isolectin Gs-IB4,using a murine laser-induced CNV ***:B6 ***-Tg(Csf1 r-EGFP)1 Hume/J heterozygous adult mice were subjected to laser-induced CNV and were monitored by fluorescein angiography(FA),multicolor(MC)fundus imaging and optical coherence tomography angiography(OCTA)at day 14 after CNV *** pigment epithelium(RPE)whole mounts were prepared at the end of the experiment and were stained with isolectin *** areas were measured in all different imaging modalities at day 14 after CNV from three independent raters and were compared to choroidal-RPE whole *** correlation coefficient(ICC)type 2(2-way random model)and its 95%confidence intervals(CI)were calculated to measure the correlation between different raters’***’s rank correlation coefficient(spearman’s r)was calculated for the comparison between FA,MC and OCTA data and histology ***:FA(early and late)and MC correlates well with the CNV measurements ex vivo with FA having slightly better correlation than MC(FA early spearman’s r=0.7642,FA late spearman’s r=0.7097,and MC spearman’s r=0.7418),while the interobser ver reliability was good for both techniques(FA early ICC=0.976,FA late ICC=0.964,and MC ICC=0.846).In contrast,OCTA showed a poor correlation with ex vivo measurements(spearman’s r=0.05716)and high variability between different raters(ICC=0.603).CONCLUsION:This study suggests that FA and MC imaging could be used for the evaluation of CNV areas in vivo while caution must be taken and comparison studies should be performed when OCTA is employed as a CNV monitoring tool in small rodents.

EEG connectivity analysis in infants:A Beginner's Guide on Preprocessing and Processing Techniques

Over the last decades,infantile brain networks have received increased scientific attention due to the elevated need to understand better the maturational processes of the human brain and the early forms of neural ***(EEG)is becoming a popular tool for the investigation of functional connectivity(FC)of the immature brain,as it is easily applied in awake,non-sedated ***,there are still no universally accepted standards regarding the preprocessing and processing analyses which address the peculiarities of infantile EEG data,resulting in comparability difficulties between different ***,during the last few years,there is a growing effort in overcoming these issues,with the creation of age-appropriate *** FC in infants has been mostly measured via linear metrics and particularly coherence analysis,non-linear methods,such as cross-frequency-coupling(CFC),may be more valuable for the investigation of network communication and early network ***,graph theory analysis often accompanies linear and non-linear FC computation offering a more comprehensive understanding of the infantile network *** current review attempts to gather the basic information on the preprocessing and processing techniques that are usually employed by infantile FC studies,while providing guidelines for future studies.

Management of carbon tetrachloride-induced acute liver injury in rats by syngeneic hepatocyte transplantation in spleen and peritoneal cavity

AIM: Acute hepatitis may seldom have a fulminant *** the treatment of this medical emergency, potential liver support measure must provide immediate and sufficient assistance to the hepatic function. The goal of our study was to study the adequacy of hepatocyte transplantation (HCTx) in two different anatomical sites, splenic parenchyma and peritoneal cavity, in a rat model of reversible acute hepatitis induced by carbon tetrachloride (CCl4).METHODs: After CCl4 intoxication, 84 male Wistar rats used as recipients were divided in to four experimental groups accordingly to their treatment: Group A (n=24): intrasplenic transplantation of 10±10^6 isolated hepatoo/tes, Group B (n=24):intrapedtoneal transplantation of 20×10^6 isolated hepatocytes attached on plastic microcarriers, Group C (n= 18): intrasplenic injection of 1 mL normal saline (sham-operated controls),Group D (n=18): intraperitoneal injection of 2.5 mL normal saline (sham-operated controls). survival, liver function tests (LIT) and histology were studied in all four groups, on d 2,5 and 10 ***: The ten-day survival (and mean survival) in the 4 groups was 72.2% (8.1±3.1), 33.3% (5.4±3.4), 0%(3.1±1.3) and 33.3% (5.4±3.6) in groups A, B, C, D,respectively (PAB＜0.05, PAc＜0.05, P80=Ns). In the final survivors, LIT (except alkaline phosphatase) and hepatic histology retumed to normal, independently of their previous therapy. Viable hepatocytes were identified within splenic parenchyma (in group A on d 2) and both in the native liver and the fatty tissue of abdominal wall (in group B on d 5).CONCLUsION: A significantly better survival of the intrasplenically transplanted animals has been demonstrated,Intraperitoneal hepatocytes failed to promptly engraft. A different timing between liver injury and intraperitoneal HCTx may give better results and merits further investigation.

Heroin-induced changes of CD34-positive rat thymus cells

Heroin is a well known opioid that causes al-terations to the immune system of a number of investigated animals. Only a few studies have explored the effect of heroin on the lymphocytes maturation. Thymocyte progenitors originate from haematopoietic stem cells in the bone marrow. The immature T-cells express neither CD4 nor CD8, and are therefore classed as double-negative (CD4-CD8-) cells. CD34 glycoprotein is the only defined marker of the immature T-lymphocytes. In this study we have investigated the changes induced in CD34+ rat thymocytes after heroin administration by immunofluores-cence in frozen rat thymus sections using the 4H11 monoclonal anti-CD34 antibody. There is a remarkable decrease in the number of CD34+ immature thymocytes when examined 1hour after last heroin injection and a small recover when examination took place 20 days after last injection. The above results suggest a major effect of heroin administration early in the ma- turation process of T lymphocytes probably by increasing the apoptotic cell death and the ne- gative consequences for the immune system responses.

serum Calprotectin and Prealbumin Levels among Ovarian Cancer Patients Aged 30 - 45 Years

Purpose: serum calprotectin and prealbumin have been previously linked with ovarian cancer. However, to date, their effectiveness in the diagnosis of the disease remains uncertain. The purpose of the present study is to determine whether serum prealbumin and calprotectin correlate with the stage and type of histologic diagnosis. Materials-Methods: In the present study, we included 80 women aged 30 - 45 years that had either primary ovarian cancer (stages 1 and 2) with elevated levels of CA-125 (n = 40) or were healthy (n = 40). serum calprotectin and prealbumin levels were assessed using ELIsA kits. statistical analysis was performed with sPss v.20.0 statistical software. Results: Both plasma prealbumin and calprotectin levels were significantly different in patients with ovarian cancer compared to healthy controls. specifically, serum prealbumin levels showed a level towards decreasing as the stage of the disease advanced (21.2 (15.5 - 30.6) in healthy controls, 12.8 (6.9 - 16) for patients with stage 1 disease and 9.9 (4.0 - 15.5) for patients with stage 2 disease p p Conclusions: Both prealbumin and calprotectin seem to be useful markers for the identification of patients suffering from ovarian cancer.

AIM: To determine the mechanical properties of anasto- motic colonic tissue in experimental settings and there- fore give a measure of wound healing. METHODs: Thirty-six male Wistar rats were used as ex- perimental models of anastomotic tissue integrity. On the 5th post-operative day, the tensile strength was measured by application of an axial force, providing a quantitative measure of anastomotic dehiscence and leakage. REsULTs: Diagrams of the load as a function of the time [P = P (t)] and of the displacement also as a func- tion of time [Δs = Δs (t)] were recorded for each test, permitting the design of the load versus the displace- ment diagram and thus providing significant data about the critical values of anastomotic failure. Quantitative data were obtained concerning the anastomotic strength of both control specimens (healthy rats), as well as spec- imens from non-healthy rats for comparison. CONCLUsION: This experimental model provides an excellent method of measuring anastomotic strength. Despite the relative small number of specimens used, this method provides an accurate way of measuring wound repair. More experimental measurements need to be performed to correlate emerging tensile strength val-ues to anastomotic failure.

Pancreatic ductal adenocarcinoma(PDAC)has a high mortality rate and is notoriously refractory to multiple cancer *** recent years,cancer therapy has expanded beyond traditional cytotoxic chemotherapy to targeted agents and immunotherapy which have been successfully implemented in many *** robust pre-clinical research,these novel therapies have only had a small impact on ***,there have been successes with emerging clinical data supporting a potential role for checkpoint inhibitor therapy and targeted therapy with poly(ADP-ribose)polymerase inhibitors for select subsets of PDAC *** this clinical review,we discuss recent pre-clinical evidence for targeting metabolic pathways as well as prevalent intratumoral immune subsets,and focus on clinical trials designed to test novel agents in *** challenge of translating pre-clinical findings to patients remains substantial and many clinical trials yield negative results,but collaborative efforts and renewed focus on novel clinical trials have led to optimism that we will identify additional options for PDAC patients and change outcomes for this deadly disease.

AIM: To investigate the in vivo effects of type Ⅰdiabetes on the mechanical strength of tibial bone in a rodent ***: The biomechanical effect of diabetes on the structural integrity of the tibia in streptozotocin induced diabetic Wistar rats was analysed. Induction of diabetes was achieved by an intra-peritoneal injection and confirmed by measuring serial blood glucose levels(> 150 mg/d L). After 8 wk the tibiae were harvested and compared to a control group. Biomechanical analysis of harvested tibiae was performed using a threepoint bending technique on a servo hydraulic MTs 858 MiniB ionix frame. Maximum force applied to failure(N), stiffness(N × mm) and energy absorbed(N/mm) were recorded and plotted on load displacement curves. A displacement control loading mode of 1 mm/min was selected to simulate quasi-static loading conditions. Measurements from load-displacement curves were directly compared between ***: Fourteen streptozotocin induced diabetic Wistar rats were compared against nineteen non-diabetic controls. An average increase of 155.2 g in body weight was observed in the control group compared with only 5 g in the diabetic group during the experimental study period. Levels of blood glucose increased to 440.25 mg/d L in the diabetic group compared to 116.62 mg/d L in the control *** biomechanical results demonstrate a highly significant reduction in the maximum load to failure from 69.5 N to 58 N in diabetic group compared to control(P = 0.011). Energy absorption to fracture was reduced from 28.2 N in the control group to 23.5 N in the diabetic group(P = 0.082). No significant differences were observed between the groups for bending ***: streptozotocin-induced diabetes in rodents reduces the maximum force and energy absorption to failure of bone, suggesting a predisposition for fracture risk.
