To study the inhibitory effects of plasmid-derived small interfering RNA(siRNA)andsynthetic siRNA on the expression of the hepatitis B virus surface(HBs)gene,three plasmid-derived siRNAsand one synthetic siRNA that complement the coding region of the HBs gene were *** HBsexpression plasmid pHBs-EGFP was also *** cells were co-transfected with pHBs-EGFPand the above *** HBs mRNA quantities were measured by reverse-transcription PCR,and thelevel of HBs-EGFP fusion protein was quantified by fluorescent *** concentrations of thehepatitis B virus surface antigen(HBsAg)derived from the culture supernatant of transfected HepG2.2.15cells were measured by an enzyme-linked immunosorbent assay(ELISA)*** results showed that thethree plasmid-derived siRNAs and the synthetic siRNA can effectively reduce the quantities of HBs mRNAand *** plasmid-derived siRNA psiRNA1 was found to be the most effective inhibitor of *** can inhibit HBs-EGFP expression by 63.3% and suppress HBs mRNA by 75.6%.To furthersubstantiate the above observations,psiRNA1 was transfected into HepG2.2.15 cells(an HBV secreting cellline).The transfections resulted in almost complete blockage of HBsAg production,whereas control vector-transfected cells secreted high levels of HBsAg 7 days *** conclusion,our data suggeststhat RNA interference(RNAi)is an efficient approach for reducing the level of HBs transcripts and proteinsand for suppressing HBsAg production.

The present study was designed to determine the changes of phosphorylation of cAMP- response element binding protein (CREB) in hippocampus induced by ohmefentanyl stereoisomers (F9202 and F9204) in conditioned place preference (CPP) paradigm. The results showed that mice receiving F9202 and F9204 displayed obvious CPP. They could all significantly stimulate CREB phosphorylation and maintained for a long time without affecting total CREB protein levels. The effect of F9204 was similar to morphine which effect was more potent and longer than F9202. We also examined the effects of ketamine, a noncompetitive N-mthyl-D-aspartate receptor (NR) antagonist, on morphine-, F9202- and F9204- induced CPP and phosphorylation of CREB in hippocampus. Ketamine could suppress not only the place preference but also the phosphorylation of CREB produced by morphine, F9202 and F9204. Peroxisome proliferator-activated receptor (PPAR) isoforms (α and γ) are known to beexpressed in pancreatic islets as well as in insulin-producing cell *** of PPAR have been shoWn toenhance glucose-induced insulin secretion in rat pancreatic ***,their effect on insulin secretionis still *** understand the molecular mechanism by which PPAR7 exerts its effect on glucose-induced insulin secretion,we examined the endogenous activity of PPAR isoforms,and studied the PPARyfunction and its target gene expression in INS-1 *** found that:(1)endogenous PPARγ was activatedin a ligand-dependent manner in INS-1 cells;(2)overexpression of PPARy in the absence of PPARγ ligandsenhanced glucose-induced insulin secretion,which indicates that the increased glucose-induced insulin secretionis a PPARγ-mediated event;(3)the addition of both PPARγ and retinoid X receptor (RXR) ligands showed asynergistic effect on the augmentation of reporter activity,suggesting that the hetero-dimerization of PPAR7and RXR is required for the regulation of the target genes;(4)PPARs upregulated both the glucose transporter2 (GLUT2) and Cbl-associated protein (CAP) genes in INS-1 *** findings suggest an importantmechanistic pathway in which PPARγ enhances glucose-induced insulin secretion by activating the expressionof GLUT2 and CAP genes in a ligand-dependent manner.

Aim:To explore the effects and mechanisms of caffeine and coffee on bladderdysfunction in streptozotocin-induced diabetic ***:Sprague-Dawleymale rats were divided randomly into 4 groups:control,diabetes mellitus(DM),DM with coffee treatment,and DM with caffeine *** diabetic rat wasinduced by intraperitoneal injection of streptozotocin(60 mg/kg).After 7 weeks oftreatment with coffee and caffeine,cystometrogram,contractile responses to elec-trical field stimulation(EFS)and acetylcholine(ACh),and cyclic AMP(cAMP)concentration of the bladder body and base were ***:The bladderweight,volume threshold for micturition and post-void residual volume(PVR)inthe diabetic rats were significantly higher compared to those in the control *** or caffeine treatment significantly reduced the bladder weight,bladdercapacity and PVR in the diabetic *** caused significant decreases in cAMPconcentration of the bladder and coffee and caffeine caused upregulation of cAMPcontent in the diabetic *** addition,coffee and caffeine tended to normal-ize the altered detrusor contractile responses to EFS and ACh in the diabetic ***:These results indicate that caffeine and coffee may have beneficialeffects on bladder dysfunction in the early stage of diabetes by increasing cAMPcontent in the lower urinary tract,recovering the micturition reflex and improvingthe detrusor contractility.
