Aim: To investigate the gene expression profile changes in the cerebral cortex of mice injected icv with amyloid beta-protein fragment 25-35 using cDNA microarray. Methods: Balb/c mice were randomly divided into a control group and’ A(3-treated group. The Morris water maze test was performed to detect the effect of A-injection on the learning and memory of mice. Atlas Mouse 1.2 Expression Arrays containing 1176 genes were used to investigate the gene expression pattern of each group. Results: The gene expression profiles showed that 19 genes including TBXl, NF-kB, AP-1/c-Jun, cadherin, integrin, erb-B2, and FGFRl were up-regulated after 2 weeks of icv administration of A; while 12 genes were down-regulated, including NGF, glucose phosphate isomerase 1, AT motif binding factor 1, Na~+/K~+-ATPase, and Akt. Conclusions: The results provide important leads for pursuing a more complete understanding of the molecular events of Ap-injection into mice with Alzheimer disease.

GATA-2, a transcription factor, is expressed in several types of blood cells and in the central nervous system (CNS), and regulates the differentiation of these cells. We have obtained five zebrafish transgenic germlines that carry and express the green fluorescent protein (GFP) gene ligated to various 5’ flanking sepuences of zebrafish GATA-2 gene. The spatial pattern of GFP expression varies, mainly depending on which regulatory sequence is used, among the germlines. In some of the germlines, the expression of GFP is restricted to the CNS and the enveloping layer (EVL) cells, while in some other lines GFP is observed only in the CNS. it is noted that the intensity of GFP in the transgenic fish remain unchanged after a six-generation passage of the transgenes. The transgenic fish could find its uses in the future in generating tissue-specific, even cellspecific mutant fish and in functional study of related genes through transgenesis.

Aim:To investigate the effects of a tissue-specific selective estrogen receptormodulator,idoxifene,on hepatic fibrosis in ***:Hepatic fibrosis wasinduced by dimethylnitrosamine(DMN)in male *** DMN model of hepaticfibrosis and the hepatocytes undergoing oxidative stress were treated withidoxifene *** effect of idoxifene on hepatic fibrosis in the DMNmodel was examined by *** of idoxifene on antioxi-dant enzyme levels of copper,zinc-dependent superoxide dismutase(CuZn-SOD),and cellular glutathione peroxidase(GSHPx)were measured by *** ofidoxifene on activation,proliferation,and apoptosis of culture-activated hepaticstellate cells(HSC)were analysed by immunohistochemistry,bromodeoxyuridine(BrdU)uptake,and flow cytometry, respectively. Results:Idoxifene could mark-edly suppress DMN-induced hepatic fibrosis in male rats.A treatment of 0.4mg·kg-1·d-1of idoxifene reduced the protein levels of collagen in the DMN modelby 41.19%(P<0.05).Protein level of CuZn-SOD and activitiy of GSHPx in livertreated with DMN plus 0.4 ***-1·d-1of idoxifene were 2.65 times(P<0.05)and2.08 times greater(P<<0.05)than that of liver treated with DMN alone *** protein level of CuZn-SOD and activity of GSHPx in cultured rat hepatocytestreated with ferric nitrilotriacetate(FeNTA)plus 1×10-7mol/L of idoxifene were3.43 times(P<0.05)and 2.52 times(P<0.05)greater than that treated with *** could inhibit HSC *** with the control,theuptake of BrdU in HSC cultured with 1×10-7mol/L of idoxifene was reduced by51.87%(P<0.05),and the number of apoptotic HSCs cultured with 1×10-7mol/L ofidoxifene increased by 94.52%(P<0.05).Conclusion:Idoxifene showed inhibi-tory action on hepatic fibrosis in male rats.

Aim:To investigate the movement of α1A-adrenergic receptors(α1A-AR) stimu-lated by agonist,phenylephrine （PE）,and the dynamics of receptor movement inreal time in single living cells with millisecond ***:We labeledα1A-AR using the monoclonal,anti-FLAG （a kind of tag） antibody and Cy3-conju-gated goat anti-mouse IgG and recorded the trajectory of their transport processin living HEK293A cells stimulated by agonist,PE,and then analyzed their dy-namic ***:The specific detection of α1A-AR on the surface ofliving HEK293A-α1Acells was achieved.α1A-AR internalize under the stimulationof *** the cells were stimulated with PE for 20 min,apparent colocalizationwas found between α1A-AR and *** 40 min stimulation of PE,trajecto-ries of approximate linear motion in HEK293A-α1Acells were recorded,and theirvelocity was ***:The specific labeling method on the livingcell surface provides a convenient means of real-time detection of the behavior ofsurface *** this method we were able to specifically detect α1A-AR andrecord the behavior of individual particles of receptors with 50 ms exposure time inreal time in single living cells.

BACKGROUND: Pancreatic cancer remains a devastating disease with a 5-year survival rate of less than 5%. Recent advances in diagnostic methods and therapeutic approaches have increased the possibility of improving the existing poor prognosis. DATA SOURCES: English-language articles reporting early diagnosis and therapy of pancreatic cancer were searched from the MEDLINE and PubMed databases, Chinese-language articles were from CHKD (china Hospital Knowledge Database) RESULT: The current literature about pancreatic cancer was reviewed from three aspects: statistics, screening and early detection, and therapy. CONCLUSIONS: Early detection and screening of pancreatic cancer currently should be limited to high risk patients Surgical resection is the only curative approach available, with some recent improvement in outcomes. Gemcitabine has been a standard treatment during the last decade. Gemcitabine based combination treatment, especially combined with newer molecular targeted agents, is promising. The rationale for radiotherapy is controversial, but with the recent development of modern radiation delivery techniques, radiotherapy should be intensified. Patients with borderline pancreatic cancer could benefit from neoadjuvant therapy but more evidence is needed and the best neoadjuvant regimen is still to be determined.
