The role of XPC protein deficiency in tobacco smok.-induced DNA hypermethylation of tumor suppressor genes

DNA hypermethylation of tumor suppressor genes has been frequently observed in cancer patients, and therefore, may provide a valuable biomark.r for cancer prevention and treatment. DNA hypermethylation may also provide an important mechanism in cancer progression. Lung cancer is strongly associated with exposure to environmental carcinogens, especially tobacco smok.. DNA damage generated by tobacco smok. is believed to play an important role in lung cancer development. XPC is a DNA damage recognition protein required for DNA repair and other DNA damage responses and attenuated XPC protein levels have been detected in many lung cancer patients. We studied the role of XPC protein deficiency in tobacco smok.-caused DNA hypermethylation of important tumor suppressor genes. Using both normal human fibroblasts (NF) and XPC-deficient hu man fibroblasts (XPC), our DNA methylation studies demonstrated that the XPC deficiency caused elevated levels of DNA hypermethylation in both Brca1 and Mlh1 tumor suppressor genes following exposure to tobacco smok. condensate (TSC). The results of our ChIP studies revealed that the XPC deficiency led to an increased binding of DNA methyltransferase 3A (DNMT3A) at the promoter region CpG island-containing sequences of these genes under the TSC treatment;however, this increase was partially diminished with prior treatment with caffeine. The results of our immuno-precipitation (IP) studies demonstrated a protein-protein interaction of the ATR with DNMT3A. Our western blots revealed that the XPC deficiency caused an increase in TSC-induced ATR phosphorylation at serine 428, an indicator of ATR activation. All these results suggest that XPC deficiency causes an accelerated DNA hypermethylation in important tumor suppressor genes under tobacco smok. exposure and activation of the ATR signaling pathway is involved in this DNA hypermethylation process.

STEP AP 242 Managed Model-based 3D Engineering:An Application Towards the Automation of Fixture Planning

Fixture design and planning is one of the most important manufacturing activities, playing a pivotal role in deciding the lead time for product development. Fixture design, which affects the part-quality in terms of geometric accuracy and surface finish, can be enhanced by using the product manufacturing information(PMI) stored in the neutral standard for the exchange of product model data(STEP) file, thereby integrating design and manufacturing. The present paper proposes a unique fixture design approach, to extract the geometry information from STEP application protocol(AP) 242 files of computer aided design(CAD) models, for providing automatic suggestions of locator positions and clamping surfaces. Automatic feature extraction software "FiXplan", developed using the programming language C#, is used to extract the part feature, dimension and geometry information. The information from the STEP AP 242 file is deduced using geometric reasoning techniques, which in turn is utilized for fixture planning. The developed software is observed to be adept in identifying the primary, secondary, and tertiary locating faces and locator position configurations of prismatic components. Structural analysis of the prismatic part under different locator positions was performed using commercial finite element method software, ABAQUS, and the optimized locator position was identified on the basis of minimum deformation of the *** area-ratio(base locator enclosed area(%)/work.piece base area(%)) for the ideal locator configuration was observed as 33%. Experiments were conducted on a prismatic work.iece using a specially designed fixture, for different locator configurations. The surface roughness and waviness of the machined surfaces were analysed using an Alicona non-contact optical profilometer. The best surface characteristics were obtained for the surface machined under the ideal locator positions having an area-ratio of 33%, thus validating the predicted numerical resul

Power system planning is a capital intensive investment-decision problem. The majority of the conven- tional planning conducted since the last half a century has been based on the least cost approach, k.eping in view the optimization of cost and reliability of power supply. Recently, renewable energy sources have found a niche in power system planning owing to concerns arising from fast depletion of fossil fuels, fuel price volatility as well as global climatic changes. Thus, power system planning is under-going a paradigm shift to incorporate such recent technologies. This paper assesses the impact of renewable sources using the portfolio theory to incorporate the effects of fuel price volatility as well as CO2 emissions. An optimization framework.using a robust multi-objective evolutionary algorithm, namely NSGA-II, is developed to obtain Pareto optimal solutions. The performance of the proposed approach is assessed and illustrated using the Indian power system considering real-time design prac- tices. The case study for Indian power system validates the efficacy of the proposed methodology as developing countries are also increasing the investment in green energy to increase awareness about clean energy technologies.

Extent of mismatch between the period of circadian clock. and light/dark.cycles determines time-to-emergence in fruit flies

Circadian clock. time developmental stages of fruit flies Drosophila melanogaster, while light/dark.（LD） cycles delimit emergence of adults, conceding only during the ＂allowed gate.＂ Previous studies have revealed that time-to-emergence can be altered by mutations in the core clock.gene period （per）, or by altering the length of LD cycles. Since this evidence came from studies on genetically manipulated flies, or on flies maintained under LD cycles with limited range of periods, inferences that can be drawn are limited. Moreover, the extent of shortening or lengthening of time-to-emergence re- mains yet unk.own. In order to pursue this further, we assayed time-to-emergence of D. rnelanogaster under 12 different LD cycles as well as in constant light （LL） and constant dark.conditions （DD）. Time-to-emergence in flies occurred earlier under LL than in LD cycles and DD. Among the LD cycles, time-to-emergence occurred earlier under T4-T8, followed by T36-T48, and then T12-T32, suggesting that egg-to-emergence duration in flies becomes shorter when the length of LD cycles deviates from 24 h, bearing a strong positive and a marginally negative correlation with day length, for values shorter and longer than 24 h, respectively. These results suggest that the extent of mismatch between the period of circadian clock. and environmental cycles determines the time-to-emergence in Drosophila.

The correlation between friction coefficient and areal topography parameters for AISI 304 steel sliding against AISI 52100 steel

Dry wear experiments provide an insight detail on how severely contacting surfaces change under unlubricated sliding condition. The theory of dry sliding wear is used for understanding mixed-lubrication regime in which asperity interactions play a significant role in controlling of the friction coefficient(f). The purpose of this work.is to study the tribological behavior of AISI 304 steel in contact with AISI 52100 steel during wear. Both materials are used in rolling element bearings commanly. Experiments are carried out using a pin-on-disc tribometer under dry friction condition. The areal(three dimensional, 3D) topography parameters are measured using a 3D white light interferometer(WLI) with a 10× objective. After wear tests, wear mechanisms are analyzed utilizing scanning electron microscope(SEM). Factorial design with custom response surface design(C-RSD) is used to study the mutual effect of load and speed on response variables such as f and topography parameters. It is observed that the root mean square roughness(S_(q)) decreases with an increase in sliding time. Within the range of sliding time, S_(q) decreases with an increase in the normal load. Within the range of sliding speed and normal load, it is found that S_(q), mean summit curvature(S_(sc)), and root mean square slope(S_(dq)) are positively correlated with f. Whereas, negative correlation is found between f and correlation length(Sal), mean summit radius(R), and core roughness depth(S_(k.).

Experimental and numerical investigation of liquid-solid binary fluidized beds： Radioactive particle track.ng technique and dense discrete phase model simulations

Liquid-solid binary fluidized beds are widely used in many industries. However, the flow behavior of such beds is not well understood due to the lack.of accurate experimental and numerical data. In the current study, the behavior of monodisperse and binary liquid-solid fluidized beds of the same density but dif- ferent sizes is investigated using radioactive particle track.ng （RPT） technique and a dense discrete phase model （DDPM）. Experiments and simulations are performed in monodisperse fluidized beds containing two different sizes of glass beads （0.6 and I mm） and a binary fluidized bed of the same particles for vari- ous bed compositions. The results show that both RPT and DDPM can predict the mixing and segregation pattern in liquid-solid binary fluidized beds. The mean velocity predictions of DDPM are in good agree- ment with the experimental findings for both monodisperse and binary fluidized beds. However, the axial root mean square velocity predictions are only reasonable for bigger particles. Particle-particle interac- tions are found to be critical for predicting the flow behavior of solids in liquid-solid binary fluidized beds.

Ce-Co-doped BiFeO_3 multiferroic for optoelectronic and photovoltaic applications

The Ce-Co-doped BiFeO3 multiferroic, Bi（1-x）Ce）xFe（1-x）CoxO3（x = 0.00, 0.01, 0.03, and 0.05）, has been prepared by a sol-gel auto-combustion method and analyzed through Raman spectroscopy, photoluminescence, and UV-visible spectroscopy. We have observed an anomalous intensity of the second-order Raman mode at - 1260 cm^-1 in pure BFO and suppressed intensity in doped samples, which indicates the presence of spin two-phonon coupling in these *** photoluminescence spectra show reduction in the intensity of emission with the increasing dopant concentration, which indicates the high charge separation efficiency. A sharp absorption with three charge transfer（C-T） and two d-d transitions are shown by UV-visible spectra in the visible region. The band gap of BiFeO3（BFO） is decreasing with increasing dopant concentrations and the materials are suitable for photovoltaic applications.

Effects of Large-Scale Climatic Oscillations on the Variability of the Indian Summer Monsoon Rainfall

In India,large-scale climatic oscillations have strong influences on the Indian summer monsoon rainfall(ISMR),which plays a crucial role in India’s agricultural production and economic ***,there are limited studies in India that explore the influences of decadal and multidecadal oscillations on the ISMR and associated El Niño–Southern Oscillation(ENSO).Therefore,in this study we carried out a comprehensive and detailed investigation to understand the influences of ENSO,Pacific decadal oscillation(PDO),and Atlantic multidecadal oscillation(AMO)on ISMR across different regions in *** statistical significance of ISMR associated with different phases(positive/warm and negative/cold)of ENSO,PDO,and AMO(individual analysis),and combined ENSO–AMO,and ENSO–PDO(coupled analysis)were analysed by using the nonparametric Wilcoxon Rank.Sum(WRS)*** individual analysis results indicate that in addition to the ENSO teleconnection,AMO and PDO significantly affect the spatial patterns of *** analysis was performed to understand how the phase shift of PDO and AMO has modulated the rainfall during El Niño and La Niña *** results indicate that the La Niña associated with a positive PDO phase caused excessive precipitation of about 21%–150%in the peninsular,west–central,and hilly regions compared to the individual effect of ENSO/PDO/AMO on ISMR;similarly,the west–central,coastal,and northwest regions received 15%–56%of excessive ***,during the El Niño combined with PDO positive(AMO positive),above-normal precipitation was observed in hilly,northeast,and coastal(hilly,northeast,west–central,and coastal)regions,opposite to the results obtained from the individual ENSO *** study emphasizes the importance of accounting the decadal and multidecadal forcing when examining variations in the ISMR during different phases of ENSO events.

Microwave-assisted reduction roasting–magnetic separation studies of two mineralogically different low-grade iron ores

The microwave-assisted reduction behaviours of two low-grade iron ores having a similar Fe content of 49wt%but distinctly different mineralogical and liberation characteristics were *** performances in terms of the iron grade and recovery as obtained from statistically designed microwave(MW)roasting followed by low-intensity magnetic separation(LIMS)experiments were *** respective optimum conditions,the titano-magnetite ore(O1)could yield an iron concentrate of 62.57%Fe grade and 60.01%Fe recovery,while the goethitic ore(O2)could be upgraded to a concentrate of 64.4%Fe grade and 33.3%Fe *** with the goethitic ore,the titanomagnetite ore responded better to MW *** characterization studies of the feed and roasted products obtained at different power and time conditions using X-ray diffraction,optical microscopy,vibrating-sample magnetometry,and electron-probe microanalysis explain the sequential reduction in the iron oxide ***,tak.ng advantage of the MW absorbing character of the titano-magnetite ore,a blend of the same with the goethite-rich ore at a weight ratio of 60:40(O2:O1)was subjected to MW roasting that resulted in a concentrate of 61.57%Fe grade with a Fe recovery of 64.47%.

Synthesis of Aligned ZnO Nanorod Array on Silicon and Sapphire Substrates by Thermal Evaporation Technique

High density ZnO nanorods were grown by thermal evaporation of Zn powder at 700℃ on Si （100） and sapphire （0001） substrates at atmospheric pressure without adding any catalyst. The nanorods were characterizated in terms of their structural and optical properties. The nanorods grown on Si have a diameter of 350-400 nm and a length of 1.2 μm while those on sapphire have a diameter of 600-800 nm and a length of 2.5 μm. During the structural characterization, it is noticed that the rods grow along the （0002） plane with perfect hexagonal facet. The room temperature photoluminescence spectrum showed a strong UV emission peak.at 385 nm with a weak.green band emission, which confirms that nanorods have good optical properties. It is observed that the oxygen partial pressure plays an important role to control the shape and size of the nanorods in thermal evaporation growth technique.
