Lack of the fragile X mental retardation protein leads to Fragile X syndrome(FXS)while increased levels of FmR1 mRNA,as those observed in premutation carriers can lead to Fragile X-associated tremor ataxia syndrome(FXTAS).Until recently,FXTAS had been observed only in carriers of an FmR1 premutation(55–200 CGG repeats);however the disorder has now been described in individuals carriers of an intermediate allele(45–54 CGG repeats)as well as in a subject with a full mutation with ***,we report on molecular and clinical data of a male FmR1 mosaic individual with full and premutation *** analysis of FmR1 and FmRP expression in this subject is consistent with a FXS *** observed reduced expression of FmRP in both peripheral blood and brain leading to the FXS *** addition,a dramatic 90%depletion of both FmR1 mRNA and FmRP levels was observed in the blood,as normally observed in FXS cases,and an even greater depletion in the brain.A clinical report of this patient,at age 71,described neurodegenerative signs of parkinsonism that were likely,in retrospect,part of a FXTAS scenario as post-mortem examination shows the presence of intranuclear inclusions,the hallmark pathology of *** findings presented in this study indicate co-morbidity for both FXS and FXTAS in this individual carrying both full and premutation FmR1 *** addition,based on symptoms and pathological and molecular evidence,this report suggests the need to redefine the diagnostic criteria of FXTAS.

Peptide biosensor reagents are emerging as an alternative to typical antibody-based detection methods. Peptides can be rapidly isolated using bacterial display methods for new and emerging biothreats and can be chemically synthesized for rapid, large-scale production. With the emergence of peptide biosensor reagents, there is a growing need to develop methods for characterizing binding interactions. Capillary electrophoresis (CE) is a free-solution separation method that is able to determine target and analyte binding association (Kb) and dissociation constants (Kd). In this study, the Kb, Kd, and peptide specificity of an isolated peptide binding reagent to protective antigen (PA) of Bacillus anthracis were evaluated using capillary electrophoresis at 10 and 20 kV. The relative binding specificity was rapidly assessed by measuring the peptide relative mobility shift at 20 kV at nonequilibrium using bovine serum albumin (BSA), horseradish peroxidase (HRP), and an anti-PA monoclonal antibody (mAb). The αPA peptide was shown to be highly specific for PA, with a Kd = 177 nm measured at 20 kV and Kd = 312 nm measured at 10 kV. These results show that peptides from bacterial display libraries can be rapidly tested for specificity and binding affinity, in solution, for use as a potential biosensor reagent against new and emerging biothreats.

Water Source Quality in Northern and Central Tanzania: Implications for Rural Communities

Limited water availability coupled with the lack of hygienic and reliable water sources plague rural areas throughout the developing world. Tanzaniahas abundant fresh water sources, yet delivery, disinfection, and conservation outside of large towns is lacking or minimal at best. Here we examine drinking water sources in two climatologically distinct regions in Tanzania. We consider their chemical and microbiological characteristics specifically with respect to the potential for ultraviolet (UV) disinfection treatment. Interviews with local villagers provided information on collection means, frequency, storage, and incidences of water related illnesses. Total suspended solids, iron (Fe) and manganese (mn) concentrations, total bacterial abundances, presence of fecal coliforms, and evidence of Escherichia coli contamination were measured in 31 water sources. Total suspended solids exceeded 15 mg/L, the limit for effective UV treatment, in more than half the sources. Principal component analysis indicated a positive correlation of bacterial abundances with levels of Fe. Water with elevated levels of mn was associated with greater incidences of diseases. Levels of both Fe and mn appeared to be more dependent on water source than on climatic differences with the chemical composition of the source rock and redox conditions of the water source at time of collection likely contributing to measured concentrations. E. coli was detected more frequently in water sources in the drier Kondoa district than in the wetter Arusha region. Water quality and socio-economic conditions within villages were linked to incidences of water-borne diseases. The maximum risk of exposure to diarrhea-causing pathogens, for example was strongly related to household income level. Nonetheless, incidences of diarrhea were reduced by more than 40% when the average monthly household expenses increased from US $10 to $20. Aims Understanding succession in tropical forest is an important aspect of vegetation science,but to date,successional processes in sea-sonally dry tropical forests(SDtFs)have received much less atten-tion than evergreen humid tropical *** aim to fill this knowledge *** We investigated vegetation succession in SDtF areas consisting of three different successional stages(early,intermediate,late),and a SDtF-savanna ecotone in the municipal district of Juramento,north of minas Gerais State,*** twelve 400 m2 plots in each area,we compared vegetation parameters and structural variables(absolute density and basal area)and examined the floristic composition of the tree component to find gradients of *** Findings We found evidence of species turnover along a successional gradi-ent,with the intermediate stage showing the highest species rich-ness and *** was accompanied with a significant increase in the number of tree individuals and basal area from the early to intermediate successional ***,the intermediate and late SDtF successional stages were more similar in structure and *** ecotone was the most species rich and was similar to the intermediate SDtF and early successional stage in species richness and floristic composition *** results will have implications for guiding SDtF management and recovery programs.

Single incision laparoscopic surgery(SILS) is a minimally invasive platform with specific benefits over traditional multiport laparoscopic surgery. The safety and feasibility of SILS has been proven, and the applications continue to grow with experience. After 500 cases at a high-volume, single-institution, we were able to standardize instrumentation and operative steps, as well as develop adaptations in technique to help overcome technical and ergonomic challenges. These technical adaptations have allowed the successful application of SILS to technically difficult patient populations, such as pelvic cases, inflammatory bowel disease cases, and high body mass index patients. This review is a frame of reference for the application and wider integration of the single incision laparoscopic platform in colorectal surgery.

Emergence and Seedling Characteristics of maize Native to the Southwestern US

Locally adapted maize landraces, which are associated with Native American groups, were traditionally planted deeply, reportedly up to 45 cm deep. Crop resources such as these should be evaluated for possible use in future sustainable farming practices. Cold temperatures often delay maize (Zea mays L.) planting in the Corn Belt, possibly reducing yield potential, and spring frost and hail can damage early plantings. If producers could plant deeper and earlier in the spring, the planting season period could be extended and the potential for frost damage reduced because the growing point would be insulated below the soil surface for a longer period of time. The emergence capabilities of eleven Native American landraces were evaluated at various planting depths and compared to one Corn Belt line, BSSS-53. Emergence from depths between 5 and 45 cm was evaluated in a growth chamber study. Seedling dry matter partitioning and morphological characteristics were also examined. A field study was then performed to further test those landraces that successfully emerged (≥ 75%) from the 25 cm depth in the growth chamber. Results indicate that some of the evaluated Native American landraces have a greater capacity to emerge from depth than BSSS-53. Emergence capacity was not related to initial seed weight. mesocotyl elongation largely accounted for successful emergence from greater planting depths. The landraces partitioned relatively more dry matter to roots than shoots compared to BSSS-53. These results suggest that several of these Native American landraces may be useful for the development of maize varieties more tolerant to deep planting.

The aim of this study was to evaluate the effect of chewing simulation on wear of artificial enamel abraded against zirconia-based crowns. Fifteen crown preparations were scanned for the manufacturing of crowns using computer-aided-design/computer-aided-machining technique (CAD/CAm), according to the following (n = 5): Polished (Pm) and glazed (Gm) monolithic zirconia (1.5 mm uniform thickness), and Bilayer (BL - 0.8 mm zirconia coping, 0.7 mm porcelain veneer) crowns. The samples were cemented and chewing simulation (2.5 million cycles/0-80N/artificial saliva/37°C) was performed with steatite indenters (6 mm diameter) as antagonists. Assuming the uniformity of the unaged samples, antagonists were scanned using a surface profilometer and the material loss volume was calculated. Roughness of the crowns’ occlusal surface was also analyzed using the profilometer. Scanning electron microscopy was used to characterize the abraded surface. One-way ANOVA and Tukey test (p = 0.05) were employed for analysis of wear results. A significant difference was observed among the groups (p 3 ± 0.015) than those abraded against monolithic zirconia, polished (Pm - 0.167 mm3 ± 0.02) and glazed (0.101 mm3 ± 0.03), which were similar to each other. Veneering porcelain results in more pronounced wear of the artificial enamel than monolithic zirconia. However, mastication against monolithic Y-TZP also imposes wear to the opposing teeth.
