Objectives: To compare mortality and death or major morbidity (DOMM) among in fants l age (EGA) born during two post- surfacta nt era time periods. Study design and patients: Comparative cohort study of very low birthweight (501- 1500 g) infants l Res earch Network born during two post- surfactant era time periods (group I, 1991 - 1994, n = 1408; group II, 1995- 1998, n = 1348). Perinatal and neonatal fact ors were compared, and group related mortality and DOMM risk were evaluated. Res ults: Mortality was higher for group I (63.1% v 56.7% ; p = 0.0006). Antenata l steroids (ANS) and antenatal antibiotics (AABX), surfactant (p lmonary dysplasia (p = 0.0008) were more prevalent in group II. In a re gression model that controlled for basic and delivery factors only,mortality ris k was greater for group I than for group II (odds ratio (OR) 1.4, 95% confiden ce interval (CI) 1.2 to 1.7); the addition of AABX and surfactant,or ANS (OR 0.9 7, 95% CI 0.79 to 1.2) to the model appeared to account for this difference. There was no difference in DOMM (86.8% v 88.4% ; p = 0.2) , but risk was lower for group I in regression models that included ANS (OR 0.70 , 95% CI 0.52 to 0.94). Conclusion: Survival to discharge was more likely duri ng the more recent period because of group differences in ANS, AABX, and surfact ant. However, this treatment shift may reflect an overall more aggressive manage ment approach. More consistent application of treatment has led to improving sur vival of ly a t the cost of greater risk of major in- hospital morbidities.

Post-traumatic stress disorder(PTSD) is a disabling, potentially chronic disorder that is characterized by re-experience and hyperarousal symptoms as well as the avoidance of trauma-related stimuli. The distress experienced by many veterans of the Vietnam War and their partners prompted a strong interest in developing conjoint interventions that could both alleviate the core symptoms of PTSD and strengthen family bonds. We review the evolution of and evidence base for conjoint PTSD treatments from the Vietnam era through the post-911 era. Our review is particularly focused on the use of treatment strategies that are designed to address the emotions that are generated by the core symptoms of the disorder to reduce their adverse impact on veterans, their partners and the relationship. We present a rationale and evidence to support the direct incorporation of emotion-regulation skills training into conjoint interventions for PTSD. We begin by reviewing emerging evidence suggesting that high levels of emotion dysregulation are characteristic of and predict the severity of both PTSD symptoms and the level of interpersonal/marital difficulties reported by veterans with PTSD and their family members. In doing so, we present a compelling rationale for the inclusion of formal skills training in emotional regulation in couple–/family-based PTSD treatments. We further argue that increased exposure to trauma-related memories and emotions in treatments based on learning theory requires veterans and their partners to learn to manage the uncomfortable emotions that they previously avoided. Conjoint treatments that were developed in the last 30 years all acknowledge the importance of emotions in PTSD but vary widely in their relative emphasis on helping participants to acquire strategies to modulate them compared to other therapeutic tasks such as learning about the disorder or disclosing the trauma to a loved one. The effects of indigenous prescribed fire on riparian vegetation in central California

Introduction:Fire has a long history,but little documented role,as a process in riparian *** millennia California Indians have applied fire to riparian ecosystems for a variety of purposes,but the effects of such fires on riparian vegetation are poorly known outside of traditional knowledge structures of indigenous ***:This study involved the seasonal application of fire based on traditional ecological knowledge versus alternate burn methods to assess fire effects on riparian vegetation in central ***:While the annual variation amongst treatments was not significant,the results of this study suggest that fall burns support higher overall richness and native species richness;spring burns yield a decrease in overall richness,but an increase in native species richness;and summer burns contribute to an initial decline in overall and native species *** evenness indices of treatments increase most with spring and summer ***:These findings are key to understanding the seasonal role of fire in managing native species in areas prone to non-native or other invasive *** research demonstrates the application of traditional ecological knowledge to facilitate an understanding of how prescribed fire could aid in the management and conservation of riparian ecosystems.

BACKGROUND Colorectal cancer(CRC) is the second leading cause of cancer-related *** stem cells(CSCs) in CRC, which are spared by many chemotherapeutics,have tumorigenic capacity and are believed to be the reason behind cancer relapse. So far, there have been no effective drugs to target colon CSCs. Diiminoquinone(DIQ) has shown promising effects on targeting colon ***, there is limited research on the effects of DIQ on eradicating CSCs in *** To investigate the anticancer potential of DIQ on colon CSCs in two-dimensional(2D) and three-dimensional(3D) models using colonospheres and patient-derived *** Various 2D methods have been used to assess the effect and the mechanism of DIQ on HCT116and HT29 cell lines including cell proliferation and viability assays, migration and invasion assays,immunofluorescence staining, and flow cytometry. The potency of DIQ was also assessed in 3D culture using the sphere formation assay and colon cancer patient-derived organoid *** Our results showed that DIQ significantly inhibited cell proliferation, migration, and invasion in HCT116 and HT29 cell lines. DIQ treatment induced apoptosis along with an accumulation of HCT116 and HT29 cancer cells in the sub-G1 region and an increase in reactive oxygen species in both CRC cell lines. DIQ reduced sphere-forming and self-renewal ability of colon cancer HCT116and HT29 stem/progenitor cells at sub-toxic doses of 1 μmol/l. Mechanistically, DIQ targets CSCs by downregulating the main components of stem cell-related-catenin, AKT, and ERK oncogenic signaling pathways. Potently, DIQ displayed a highly significant decrease in both the count and the size of the organoids derived from colon cancer patients as compared to control and 5-fluorouracil *** This study is the first documentation of the molecular mechanism of the novel anticancer therapeutic DIQ via targeting CSC, a promising compound that needs further investigation.

Relation between self-recalled childhood physical activity and adult physical activity: The women’s health initiative

Background: Evidence suggests that childhood physical activity may play a role in the etiology and prevention of adult chronic diseases. Because researchers must often depend on self-recalled physical activity data many years after the exposure, it is important to understand factors which may influence adult recall of childhood physical activity. This study evaluated the influence of adult characteristics on reported childhood physical activity and the association between adult physical activity and self-recalled childhood physical activity. Methods: 48,066 post-menopausal women from the Women’s Health Initiative Observational Study reported their physical activity level during ages 5-9, 10-14, and 15-19. Results: In this cohort, over 65% of the population reported the same category of physical activity over the three childhood age groups. While higher levels of childhood physical activity were significantly associated with higher adult physical activity, this association varied by race/ethnicity, education, smoking, body mass index, history of diabetes or cardiovascular disease, social support and physical functional status. Women who were consistently highly active reported adult physical activity levels that were 2.82 MET-hr/week (95% C.I. = 2.43, 3.20) higher compared to women who were always physically inactive during childhood. Conclusions: It is important for researchers to understand the influence of adult characteristics on reported childhood physical activity.

Metal Uptake by Plants from Soil Contaminated by Thin-Film Solar Panel Material

Various metals, including zinc (Zn), nickel (Ni), aluminum (Al), chromium (Cr), gallium (Ga), lead (Pb), copper (Cu) and indium (In), may be released and cause contamination when scrapped end-of-life (Eol) Cu(InGa)Se2 thin-film solar panel (CIGS TFSP) is buried in the soil. In this study, we grew Brassica parachinensis l. H. Bariley (VegBrassica) in three different types of soils, namely, a commercial soil, a Mollisol, and an Oxisol, which had been contaminated by CIGS TFSP to various extents. The concentrations of contaminants in these soils were positively correlated with both the amount of CIGS TFSP added and the burial period. Plants grew well in commercial soil and Mollisol, but those in Oxisol showed prominent signs of chlorosis and died after 30 days. The bioaccumulation factor (BF) and concentration of Zn in VegBrassica grown in commercial soil with 10% of CIGS TFSP added were 3.61 and 296 mg/kg, respectively, while the BF and concentration of In of VegBrassica grown in Mollisol were 3.80 and 13.72 mg/kg, respectively. The results showed that soils were contaminated by metals released from CIGS TFSP, and different adsorption patterns were observed for VegBrassica?depending on which types of metals associated with the soil properties.

目的观察人血小板(hPlTs)输注给WT小鼠和SCID小鼠后,在2种小鼠体内的复苏情况。方法通过一侧尾静脉分别给11只WT(FVB/NJ)小鼠和10只SCID小鼠输注单采的hPlTs 200μl/只,在输注后5、60、120、240 min从WT和SCID小鼠对侧尾静脉,以及360 min从SCID小鼠对侧尾静脉采集小鼠全血10μl/只(次),用3.8%EDTA抗凝后,立即采用4℃保存的1%多聚甲醛固定,4℃过夜保存;将固定的小鼠全血样本经过洗涤和再悬浮后,采用含有磁珠的TruCOUNT试管,用小鼠抗人FITC-CD61抗体和大鼠抗小鼠PE-CD41抗体染色细胞,流式细胞仪定量分析小鼠全血中复苏的hPlTs:以输注后5 min采集的小鼠全血中的hPlTs数量作为基数值,其余时间点的hPlTs数值分别与之比较,计算hPlTs输注后的复苏率。统计学分析比较hPlTs输注后,在WT和SCID小鼠体内的复苏率。结果WT和SCID小鼠体内hPlTs的复苏率,在输注后120 min分别为(37.15±10.39)%和(54.88±18.33)%(t=0.002,PlTs后120和240 min,SCID小鼠体内hPlTs的复苏率明显高于WT小鼠,提示SCID小鼠可能作为一种动物模型替代人体实验来评价在体hPlTs的功能。

目的建立一种流式细胞术检测动物模型小鼠全血中人血小板(hPlTs)的方法。方法用CO2致死SCID小鼠,立即心脏穿刺取血,用EDTA抗凝小鼠全血,备用。无菌分离5ml人的单采血小板备用。用1%多聚甲醛固定hPlTs、小鼠全血及hPlTs/小鼠全血混合血液(V/V=1∶4),4℃过夜保存。用洗涤溶液(1%PBS+1%FBS)洗涤和重新悬浮血液,将实验分为阳性对照组、阴性对照组和实验组,用相应的样本与相应抗体反应,建立检测模板。将小鼠全血和hPlTs以1∶4比例混合,用小鼠全血以1∶10、1∶100、1∶1000、1∶10000和1∶100000按体积比稀释上述混合血液,固定,4℃过夜保存。用洗涤溶液洗涤并重新悬浮血液样本,在TruCOUNT试管中定量检测hPlTs,称为"血小板检测值"。用血细胞计数仪检测单采血小板中血小板浓度,称为"血小板期望值"。与血细胞计数仪检测到的"血小板期望值"成对比较,观察可靠的检测浓度范围。结果当血小板期望值介于3000～150000/μl时,与血小板检测值具有较好的线性关系。结论采用流式细胞仪方法,可以可靠地检测到hPlTs期望值介于3000～150000/μl浓度范围内混合在小鼠全血中的hPlTs。
