Inhibitory immune receptors set thresholds for immune cell activation,and their deficiency predisposes a person to autoimmune ***,the agonists of inhibitory immune receptors remain largely unknown,representing untapped sources of treatments for autoimmune ***,we show that V-set and transmembrane domain-containing 1(VSTM1)is an inhibitory receptor and that its binding by the competent ligand soluble galectin-1(Gal1)is essential for maintaining neutrophil viability mediated by downregulated reactive oxygen species ***,in patients with systemic lupus erythematosus(SLE),circulating Gal1 is oxidized and cannot be recognized by VSTM1,leading to increased intracellular reactive oxygen species levels and reduced neutrophil *** neutrophil function or death contributes significantly to the pathogenesis of SLE by providing danger molecules and autoantigens that drive the production of inflammatory cytokines and the activation of autoreactive ***,serum levels of glutathione,an antioxidant able to convert oxidized Gal1 to its reduced form,were negatively correlated with SLE disease *** together,our findings reveal failed inhibitory Gal1/VSTM1 pathway activation in patients with SLE and provide important insights for the development of effective targeted therapies.

The Janus kinases(JAKs)are a family of intracellular tyrosine kinases that play an essential role in many basic biological processes,such as apoptosis and *** any dysfunction of the proteins in this pathway may lead to a variety of diseases,including cancer and diseases that affect the immune system,such as severe combined immune deficient(SCID).Marine biological resources have become an important source in new drug research and development due to their diversity,complexity and *** this study,Marine alkaloid Neobacillamide A was isolated from the greedy and stubborn sponge symbiotic Bacillus atrophicus C89 in the South China *** 24 novel marine alkaloid Neobacillamide A derivatives were designed and synthesized,which were evaluated for their inhibitory activity against JAK/STAT signaling pathway and their cytotoxicity to A549 *** 13c,13o,14d,14g and 14h showed potent JAK/STAT inhibition capability(concentration of 25μmol L^(-1),all inhibitory potencies were above 60%),especially compound 14g exhibited superior JAK/STAT inhibition effect(89.70%inhibition).In addition,all these compounds with a concentration of 25μmol L^(-1)displayed weak or no cytotoxicity to A549 cells,which means that these Neobacillamide A derivatives with JAK/STAT inhibition capability may have potential anti-inflammatory function.

SNP-based identification of QTL for resistance to black point caused by Bipolaris sorokiniana in bread wheat

Black point disease caused by Bipolaris sorokiniana is a problem in wheat production *** aimed to identify major quantitative trait loci(QTL)for resistance to black point and develop molecular markers for marker-assisted selection(MAS).A recombinant inbred line(RIL)population derived from a cross between Wanyuanbai 1(susceptible)and SN4143(resistant)was evaluated for black point response at three locations during two years under artificial inoculation with ***,providing data for six *** resistant and 30 susceptible RILs were selected to form resistant and susceptible bulks,respectively,that were genotyped by the wheat 660 K SNP array;685 singlenucleotide polymorphisms(SNPs)were identified,among which 385(56.2%)and 115(16.8%)were located on chromosomes 4 A and 2 B,*** segregant RNA-Seq analysis identified candidate regions on chromosomes 4 A(4.60–40.28 Mb)and 5 A(1.22–48.47 Mb).Genetic linkage maps were constructed for chromosomes 2 B,4 A,and 5 A using 59 polymorphic d CAPS and SSR ***,two QTL,designated ***-4 A and ***-5 A,were detected on chromosomes 4 A and 5 A,*** resistance allele of ***-4 A was derived from SN4143,and that of ***-5 A came from Wanyuanbai ***-4 A with a large and consistent effect(15.1%)is likely to be a new locus for black point *** markers linked to ***-4 A and ***-5 A have potential application in MASbased breeding.

Neuronal apoptosis and interleukin 1-beta converting enzyme expression in the frontal cortex and hippocampus of mice with ischemia/reperfusion injury

BACKGROUND： Interleukin 1β-converting enzyme （ICE） gene expression can induce neuronal apoptosis. However, the dynamic changes in ICE gene expression and its effects on neuronal apoptosis under cerebral ischemia/reperfusion conditions remain unclear. OBJECTIVE： To observe neuronal apoptosis and changes in ICE gene expression in the frontal cortex and hippocampus following ischemia/reperfusion injury. deSIGN, TIME AND SETTING： A randomized, controlled animal study was conducted at the Laboratory of Experimental Animal Center, the Second Hospital of Jilin University and Central Laboratory, the Second Hospital of Jilin University, China, from November 2008 to September 2009. MATERIALS： The ICE gene primer sequence （TaKaPa Co., Dalian, China）, FACScan Flow cytometer （Becton Dickinson, Franklin Lakes, N J, USA）, and Perkin Elmer GeneAmp PCR system 2400 （Perkin Elmer, Waltham, MA, USA） were used in this study. METHODS： A total of 45 healthy, adult, male, Kunming mice were randomly assigned to normal control （n = 5）, sham surgery （n = 5）, and model （n = 35） groups. The mice in the model group were equally and randomly subdivided into seven subgroups according to various reperfusion time points （1 hour, 1,3, 7, 14, 28, and 42 days）. Animal models of ischemia/reperfusion injury were established by bilateral carotid artery ligation in the model group. The mice in the sham surgery group only received saline perfusion and surgery for carotid artery exposure. MAIN OUTCOME MEASURES： Neuronal apoptosis in the frontal cortex and hippocampus of mice was measured using flow cytometry. The time course of ICE mRNA levels in the frontal cortex and hippocampus were quantified using reverse transcription-polymerase chain reaction. RESULTS： Neuronal apoptosis in the frontal cortex and hippocampus peaked at 3 days following ischemia/reperfusion injury （P 〈 0.05）. Characteristic investigation of trilayered Cu/Al8011/Al1060 composite:Interface morphology, microstructure, and in-situ tensile deformation

In this work, Cu/Al8011/Al1060 trilayered composite with an ultrathin copper layer was fabricated successfully by the hybrid process of cast-rolling and hot-roll-bonding. The effects of heat treatment were investigated on the microstructure and mechanical behavior of the composite. The results showed that annealing temperatures above 300°C led to the formation of new intermetallic compounds that joined their initial counterparts. Also, the interface growth was fast above 340°C. It was found that the annealing temperature of 320°C led to the optimal tensile properties of the composite. Through rolling, hard-brittle IMCs imposed intense plastic strains on the surrounding regions, resulting in further grain refinement of both metals in the vicinity of the interface. Although the kernel average misorientation of the trilayered composite was almost identical with the Cu/Al8011 bimetal,its amount was considerably decreased after heat treatment, as the distorted structure changed into a nearly strain-free one using the restoration phenomenon. The SEM-based in-situ tensile test showed that crack initiation occurred within the IMCs and propagated into the Cu strip, leading to its early fracture. However, the strengthdisplacement curve during in-situ tensile deformation did not decrease significantly due to the ultrathin copper layer.

In tea plants,the abundant flavonoid compounds are responsible for the health benefits for the human body and define the astringent flavor *** the downstream mechanisms of flavonoid biosynthesis have been extensively studied,the role of chalcone synthase(CHS)in this secondary metabolic process in tea plants remains less *** this study,we compared the evolutionary profile of the flavonoid metabolism pathway and discovered that gene duplication of CHS occurred in tea *** identified three CsCHS genes,along with a CsCHS-like gene,as potential candidates for further functional *** the CsCHS-like gene,the CsCHS genes effectively restored flavonoid production in Arabidopsis ***,CsCHS transgenic tobacco plants exhibited higher flavonoid compound accumulation compared to their wild-type *** notably,our examination of promoter and gene expression levels for the selected CHS genes revealed distinct responses to UV-B stress in tea *** findings suggest that environmental factors such as UV-B exposure could have been the key drivers behind the gene duplication events in CHS.

Gene expression profiling of HepG_(2) cells after treatment with black tea polyphenols

This study aimed to determine the effects of black tea polyphenols on gene expression in hepatocellular cancer *** total RNA from HepG_(2) hepatocellular cancer cells treated with black tea polyphenols was subjected to Human 14K cDNA microarray ***-time PCR and Western blot analysis were conducted to verify microarray *** tea polyphenols treatment at the dose of 20 mg/L,40 mg/L or 80 mg/L for one to three days inhibited the growth of HepG_(2) cells in a dose and time dependent manner.A total of 48 genes showed more than two-fold change after black tea polyphenols treatment,including 17 upregulated genes and 31 downregulated genes,and they were involved in the regulation of cell growth,cell cycle,apoptosis,signaling,angiogenesis,tumor invasion and ***-time PCR analysis of the selected genes showed that their mRNA expression changes were consistent with the microarray *** addition,Western blot analysis of the selected genes showed that their protein expression changes were consistent with mRNA *** conclusion,gene expression profiles provide comprehensive molecular mechanisms by which black tea polyphenols exerts growth inhibition effects on cancer *** novel molecular targets identified in this study may be further exploited as therapeutic strategies for hepatocellular cancer.

Low Temperature degradation of Alumina-toughened Zirconia in Artificial Saliva

The long-term aging behavior of an alumina-toughened zirconia （ATZ） ceramic in artificial saliva hydrothermal atmosphere was determined and compared with that of tetragonal zirconia polycrystals doped with 3mo1% Y203 （3Y-TZP）. The specimens of 3Y-TZP and ATZ were aged in distilled water and artificial salivary respectively at 134 ~C. Monoclinic phase content was evaluated by X-ray diffraction （XRD） analysis. Specimen strength was determined by a biaxial bending test. Surface screening was done using an atomic force microscope （AFM）. No statistically significant influence of any treatment on strength was demonstrated for either material. However, XRD measurements revealed ATZ＇s anti-aging properties were much better than TZP after the aging treatment. Therefore, ATZ can be applied to the field of dentistry. Artificial saliva in ceramic surface sediments may be useful for stren^heninl ceramics.

Spectral sensitivity of the compound eyes of Anomala corpulenta motschulsky(Coleoptera:Scarabaeoidea)

The scarab beetle, Anomala corpulenta Motschulsky（Coleoptera： Scarabaeoidea）, is a widespread and destructive pest in China. Vision is one of the most important means of acquiring information about the external environment. In order to contribute to the understanding of the perception of visual stimuli in this species, the light sensitivity and spectral responses of the scarab beetle, A. corpulenta, were measured by using an electroretinogram（ERG） technique. In total, 14 monochromatic light intensities, between 340 and 605 nm, were applied to the compound eyes of A. corpulenta under varying levels of adaptation to dark and light conditions. The results showed that all light stimuli induced an ERG response, with varied amplitudes. The spectral sensitivity curve of dark-adapted eyes showed one major peak（~400 nm; near-ultraviolet）, a secondary peak（from 498 to 562 nm; yellow-green） and the third peakat 460 nm. By contrast, in light-adapted eyes, only a near-UV peak was observed. From these results, we conclude that the compound eye of A. corpulenta is likely to have at least three spectral types of photoreceptor. Significance of differences were also recorded in the responses of male and female compound eyes, as well as diurnally and nocturnally. The amplitude of ERG in response to white-light stimuli varied with the light intensity： The stronger the luminance, the higher the ERG value. This suggests that the compound eye of A. corpulenta adapts quickly to changing light conditions, enabling A. corpulenta to maintain nocturnal activities.
