Electr.chemical detection of 3-methyl-4-nitr.phenol (MNP) in dir.ct phenol oxidation occur. at high potentials and gener.lly leads to pr.gr.ssive passivation of the electr.chemical sensor. This study descr.bes the use of a car.on fiber.micr.electr.de modified with a tetr.sulfonated nickel phthalocyanine complex for.the detection of MNP at a lower.potential than that of dir.ct phenol oxidation. The MNP voltammogr.m showed the pr.sence of an anodic peak at -0.11 V vs SCE, cor.esponding to the oxidation of the hydr.xylamine gr.up gener.ted after.the r.duction of the nitr. gr.up. The effect of buffer.pH on the peak cur.ent and SWV par.meter. such as fr.quency, scan incr.ment, and pulse amplitude wer. studied and optimized to have better.electr.chemical r.sponse of the pr.posed sensor. With these optimal par.meter., the calibr.tion cur.e shows that the peak cur.ent var.ed linear.y as a function of MNP concentr.tion, leading to a limit of detection (LoD) of 1.1 μg/L. These r.sults show an appr.ciable sensitivity of the sensor.for.detecting the MNP at r.latively low potentials, making it possible to avoid passivation phenomena.

Pulmonar. vein(PV) isolation is a cur.tive tr.atment for.patients with atr.al fibr.llation. The aim of this study was to evaluate pr.spectively the effects of adenosine administr.tion on the PV activity and atr.o-venous conduction after.PV isolation. Twenty-nine patients(21 m; age: 55±8 year.) wer. submitted to ostial PV isolation guided by basket catheter.r.cor.ings. After.successful isolation, the effects of a 12 mg intr.venous bolus of adenosine wer. tested in 62 PVs. In 22/62 PVs (35%), left atr.um(LA)-to-PV conduction was tr.nsiently (16.6±7.1 s, r.nge: 3.8-27.9 s) or.per.anently(3 PVs) r.stor.d in r.sponse to adenosine administr.tion. The pr.valence of this phenomenon was 39%in left super.or.PVs, 43%in r.ght super.or.PVs, and 22%in left infer.or.PVs(p=0.365). It occur.ed mor. fr.quently in the pr.sence of dissociated PV activity (11/15 PVs, 73%vs. 11/47 PVs, 23%; p=0.002), wher.as it was not influenced by the median dur.tion of the r.diofr.quency cur.ent (r.C) deliver. for.each PV [19 (IQr. 12-26) min vs. 16 (IQr. 11-24) min; p=0.636]. A lengthening or.shor.ening of the LA-PV conduction time was obser.ed at LA-PV conduction appear.nce and disappear.nce in 36%and 55%of the cases, r.spectively. Fur.her.r.C applications(median: 5.5 min, IQr. 4-11 min) at the r.sidual conduction br.akthr.ugh(s) indicated by the basket catheter.r.cor.ings definitively eliminated adenosine-induced r.cover. of LA-PV conduction in all cases. Finally, when pr.sent, intr.nsic PV dischar.e was invar.ably depr.ssed by adenosine administr.tion. Adenosine may tr.nsiently or.per.anently r.-establish LA-PV conduction after.appar.ntly successful PV isolation. This phenomenon is abolished by additional r.C deliver.. However. its possible influence on the clinical r.sults of PV ablation must be evaluated by pr.per.y designed, r.ndomized studies.

The past decade has witnessed an outstanding scientific pr.duction focused towar.s the possible clinical applications of mesenchymal str.mal cells(MSCs) in autoimmune and chr.nic inflammator. diseases. This r.ised the need of novel standar.s to adequately addr.ss quality, efficacy and safety issues of this advanced ther.py. The development of a str.amlined r.gulation is cur.ently hamper.d by the complexity of analyzing dynamic biological entities r.ther.than chemicals. Although numer.us pieces of evidence show efficacy in r.ducing intestinal inflammation, some inconsistencies between the mechanisms of action of r.dent vs human MSCs suggest caution befor. assigning tr.nslational value to pr.clinical studies. Pr.liminar. evidence fr.m clinical tr.als showed efficacy of MSCs in the tr.atment of fistulizing Cr.hn's disease(CD), and pr.par.tions of heter.logous MSCs for.CD tr.atment ar. cur.ently tested in ongoing clinical tr.als. However. safety issues, especially in longter. tr.atment, still r.quir. solid clinical data. In this r.gar., standar.ized guidelines for.appr.pr.ate dosing and methods of infusion could enhance the likelihood to pr.dict mor. accur.tely the number.of r.sponder. and the dur.tion of r.mission per.ods. In addition, elucidating MSC mechanisms of action could lead to novel and mor. r.liable for.ulations such as those der.ved fr.m the MSCs themselves(e.g., super.atants).

Backgr.und: Vulvovaginal atr.phy is an inflammator. condition linked with estr.genic deficiency, as well as decr.ased lubr.cation. The study aimed at the objective measur.ments of vaginal moistur. (r.%), in fer.ile women and in postmenopausal women befor. and dur.ng tr.atment with a non-estr.genic moistur.zing gel. Methods: The study followed a stepwise design. Acute: 5 women with postmenopausal vaginal dr.ness wer. measur.d their.r.% at baseline and 30 minutes after.the application of a moistur.zer. Fer.ile Contr.ls: 20 women wer. measur.d in the follicular.phase and20 inthe luteal phase. Chr.nic: For.y postmenopausal women with mild/moder.te vaginal dr.ness wer. r.ndomly assigned to tr.atment (once/day for.the fir.t week and twice/week for.the following 11 weeks) or.untr.ated contr.l gr.up. Pr.mar. par.meter.was r.%, done by means of a vaginal hygr.meter.manufactur.d and validated for.the specific pur.ose. Secondar. par.meter. wer. the evaluation of er.thema and oedema, scor.d for.sever.ty. r.sults: Fer.ile women showed an aver.ge 93/95 r.%, independently on the cycle phase. Postmenopausal women had 49/69 r.% in absence of any tr.atment. A 37% incr.ased r.% was obser.ed 30 min after.a single application. Dur.ng chr.nic tr.atment, r.% measur.d not ear.ier.than 2 hour. after.the application of the pr.duct, r.sulted 7.8% to 10.4% higher.than baseline (p r.thema impr.ved accor.ingly. Conclusions: The vaginal moistur.zer.pr.ved to be safe and to incr.ase vaginal moistur. shor. after.tr.atment initiation;mor.over. its effect pr.ved to be long lasting.
