Safety and Efficacy of Racotumomab-Alum Vaccine as Second-Line Therapy for Advanced Non-Small Cell Lung Cancer

Despite extensive clinical research in non-small cell lung cancer (NSCLC), overall survival is still poor. Racotumomab-alum is an anti-idiotypic cancer vaccine that targets NeuGcGM3 tumor associated ganglioside. The aim of this study was to evaluate safety and efficacy of racotumomab-alum in advanced NSCLC patients with progressive disease. This expanded access program included 86 histologically confirmed NSCLC patients, 18 years or older age, with advanced disease and without therapeutic option, with ECOG performance status ≤3, adequate organ functions and signed informed consent. The primary endpoint was overall survival and toxicity was measure assessed treatment-related toxicity according CTCAEv3. The study was approved by ethical review boards of participant institutions. Racotumomab-alum treatment consisted in 5 biweekly intradermal doses (1 mg/mL) during the induction phase of treatment (2 months). The maintenance phase consisted in monthly re-immunizations until unacceptable toxicity or PS worsening. The median overall survival time of all patients treated with racotumomab-alum was 8.96 months. The survival rates at 12 and 24 months were 42.8% and 28.0%, respectively. Patients that completed the induction phase of treatment (five doses or more) reached a median OS of 12.1 months. The most common adverse events were injection site reaction, bone pain, cough and asthenia. Racotumomab-alum cancer vaccine could be considered an effective and safe treatment option as second-line therapy for advanced NSCLC. Further clinical studies should be conducted to confirm this result.

Cervical uterine cancer represents the fourth most common malignant neoplasm worldwide in the female sex in terms of incidence, principally from epithelial origen. The high expression of EGFR in this tumor leads to the search for therapeutic alternatives. An Expanded Access Clinical Program was carried out in parallel groups, randomized, multicenter and prospective study, to evaluate the survival of patients with advanced cervical carcinoma, without therapeutic alternative, who would be treated with the therapeutic vaccine CIMAvax-EGF®, the humanized mAb nimotuzumab or the combination of both products, which targeted EGF and EGFR respectively. Modeling and numerical investigation of slow crack growth and crack arrest in ceramic polycrystals

ntergranular slow crack growth in zirconia polycrystal is described with a cohesive zone model that simulate mechanically the reaction-rupture mechanism underlying stress and environ- mentally assisted failure. A 2D polycrystal is considered with cohesive surfaces inserted along the grain boundaries. The anisotropic elastic modulus and grain-to-grain misorientation are accounted for together with an initial stress state related to the processing. A minimum load threshold is shown to originate from the onset of the reaction-rupture mechanism to proceed where a minimum traction is reached locally and from the magnitude of the initial compression stresses. This work aims at providing reliable predictions in long lasting applications of ceramics.

Flexural Strength and Weibull Analysis of Bulk Metallic Glasses

The flexural strength reliability of bulk metallic glasses （BMGs） plates is analyzed using Weibull statistics. The Weibull modulus （m） and characteristic strength （σ0） of the Zr48Cu45AI7 BMG are 34 and 2630 MPa, respectively, which are much higher than the values of fine ceramics （m 〈 30, σ0 〈 1600 MPa）. In particular, the m values obtained by flexural strength and compressive strength statistics of the Mg61Cu28Gd11 BMG are 5 and 33, respectively, indicating that the m values of BMGs are test method dependent, and only the m values obtained by flexural strength statistics can be used to make a convincible comparison with those of ceramics.

Extensins （EXTs） are highly repetitive plant O-glycoproteins that require several post-translational modifi- cations （PTMs） to become functional in plant cell walls. First, they are hydroxylated on contiguous proline residues; then they are O-glycosylated on hydroxyproline and serine. After secretion into the apoplast, O-glycosylated EXTs form a tridimensional network organized by inter- and intra-Tyr linkages. Recent studies have made significant progress in the identification of the enzymatic machinery required to process EXTs, which includes prolyl 4-hydroxylases, glycosyltransferases, papain-type cysteine endopeptidases, and peroxidases. EXTs are abundant in plant tissues and are particularly important in rapidly expanding root hairs and pollen tubes, which grow in a polar manner. Small changes in EXT PTMs affect fastgrowing cells, although the molecular mechanisms underlying this regulation are unknown. In this review, we highlight recent advances in our understanding of EXT modifications throughout the secretory pathway, EXT assembly in cell walls, and possible sensing mechanisms involving the Catharanthus roseus cell surface sensor receptor-like kinases located at the interface between the apoplast and the cytoplasmic side of the plasma membrane.
