Group-specific functional patterns of mitochondrion-related organelles shed light on their multiple transitions from mitochondria in ciliated protists

Adaptations of ciliates to hypoxic environments have arisen independently several *** on mitochondrion-related organelle(MRO)metabolisms from distinct anaerobic ciliate groups provide evidence for understanding the transitions from mitochondria to MROs within *** deepen our knowledge about the evolutionary patterns of ciliate anaerobiosis,mass-culture and single-cell transcriptomes of two anaerobic species,Metopus laminarius(class Armophorea)and Plagiopyla ***(class Plagiopylea),were sequenced and their MRO metabolic maps were *** addition,we carried out comparisons using publicly available predicted MRO proteomes from other ciliate classes(i.e.,Armophorea,Litostomatea,Muranotrichea,Oligohymenophorea,Parablepharismea and Plagiopylea).We found that single-cell transcriptomes were similarly comparable to their mass-culture counterparts in predicting MRO metabolic pathways of *** patterns of the components of the MRO metabolic pathways might be divergent among anaerobic ciliates,even among closely related ***,our findings indicate the existence of group-specific functional relics of electron transport chains(eTCs).Detailed group-specific eTC functional patterns are as follows:full oxidative phosphorylation in Oligohymenophorea and Muranotrichea;only electron-transfer machinery in Armophorea;either of these functional types in Parablepharismea;and eTC functional absence in Litostomatea and *** findings suggest that adaptation of ciliates to anaerobic conditions is group-specific and has occurred multiple *** results also show the potential and the limitations of detecting ciliate MRO proteins using single-cell transcriptomes and improve the understanding of the multiple transitions from mitochondria to MROs within ciliates.

A facile,branched DNA assay to quantitatively measure glucocorticoid receptor auto-regulation in T-cell acute lymphoblastic leukemia

Glucocorticoid(GC) steroid hormones are used to treat acute lymphoblastic leukemia(ALL) because of their pro-apoptotic effects in hematopoietic ***,not all leukemia cells are sensitive to GC,and no assay to stratify patients is *** the GC-sensitive T-cell ALL cell line CeM-C7,auto-up-regulation of RNA transcripts for the glucocorticoid receptor(GR) correlates with increased apoptotic *** study aimed to determine if a facile assay of GR transcript levels might be promising for stratifying ALL patients into hormone-sensitive and hormone-resistant *** GR transcript profiles of various lymphoid cell lines and 4 bone marrow samples from patients with T-cell ALL were analyzed using both an optimized branched DNA(bDNA) assay and a real-time quantitative reverse transcription-polymerase chain reaction *** were significant correlations between both assay platforms when measuring total GR(exon 5/6) transcripts in various cell lines and patient samples,but not for a probe set that detects a specific,low abundance GR transcript(exon 1A3).Our results suggest that the bDNA platform is reproducible and precise when measuring total GR transcripts and,with further development,may ultimately offer a simple clinical assay to aid in the prediction of GC-sensitivity in ALL patients.

Paclitaxel-loaded hollow-poly（4-vinylpyridine） nano- particles enhance drug chemotherapeutic efficacy in lung and breast cancer cell lines

Paclitaxel （PTX）, one of the most effective cytotoxins for the treatment of breast and lung cancer, is limited by its severe side effects and low tumor selectivity. In this work hollow-poly（4-vinylpyridine）（hollow-p4VP） nanoparticles （NPs） have been used for the first time to generate PTX@p4VP NPs, employing a novel technique in which a gold core in the center of the NP is further oxidized to produce the hollow structure into which PTX molecules can be incorporated. The hollow-p4VP NPs exhibit good physicochemical properties and displayed excellent biocompatibility when tested on blood （no hemolysis） and cell cultures （no cytotoxicity）. Interestingly, PTX@p4VP NPs significantly increased PTX cytotoxicity in human lung （A-549） and breast （MCF-7） cancer cells with a significant reduction of PTX ICs0 （from 5.9 to 3.6 nM in A-549 and from 13.75 to 4.71 nM in MCF-7）. In addition, PTX@p4VP caused a decrease in volume of A-549 and MCF-7 multicellular tumor spheroids （MTS）, an in vitro system that mimics in vivo tumors, in comparison to free PTX. This increased antitumoral activity is accompanied by efficient cell internalization and increased apoptosis, especially in lung cancer MTS. Our results offer the first evidence that hollow- p4VP NPs can improve the antitumoral activity of PTX. This system can be used as a new nanoplatform to overcome the limitations of current breast and lung cancer treatments.

Probabilistic Modeling of Oil Spills at the exclusive economic Zone of Cuba Using Petromar-3D Model

This article shows the probabilistic modeling of hydrocarbon spills on the surface of the sea, using climatology data of oil spill trajectories yielded by applying the lagrangian model PeTROMAR-3D. To achieve this goal, several computing and statistical tools were used to develop the probabilistic modeling solution based in the methodology of Guo. Solution was implemented using a databases approach and SQL language. A case study is presented which is based on a hypothetical spill in a location inside the exclusive economic Zone of Cuba. Important outputs and products of probabilistic modeling were obtained, which are very useful for decision-makers and operators in charge to face oil spill accidents and prepare contingency plans to minimize its effects. In order to study the relationship between the initial trajectory and the arrival of hydrocarbons spills to the coast, a new approach is introduced as an incoming perspective for modeling. It consists in storage in databases the direction of movement of the oil slick at the first 24 hours. The probabilistic modeling solution presented is of great importance for hazard studies of oil spills in Cuban coastal areas.

Lagrangian Model PeTROMAR-3D to Describe Complex Processes in Marine Oil Spills

The version 2.1 of PeTROMAR-3D model, created in the Center for Marine Meteorology of the Meteorology Institute of Cuba, is presented. This Lagrangian model has been designed to describe the physical processes of marine oil spills in the face of multiple scenarios of the marine environment. Although it is applicable to any part of the world, it is mainly designed for inter-American seas. The novelty has been to integrate the processes of drift and weathering into a model, with updated methods that incorporate 3D phenomena, a very favorable situation to achieve an operating system in Cuba and the region for the immediate and medium term. Python was chosen as the programming language because it has advanced libraries for numerical modeling, automation work and other useful tools for pre-and post-processing. By means of adapters, an important number of atmospheric, hydrodynamic and wave models have been considered to create the scenarios efficiently. The modular distribution in which the code has been created facilitates its use for other dispersion analysis and biophysical applications. Finally, a set of simple images are presented, aimed at informing decision-makers in order to mitigate the effects of the spill on the environment.

Background: electrocardiographic abnormalities may be associated with acute pancreatitis (AP). Goals: To describe the electrocardiographic disturbances present in patients with AP and to assess differences in electrolyte and pancreatic enzyme levels among patients with and without these abnormalities. Study: Fifty-one consecutive patients with AP and without pre-existing heart disease underwent a standard 12-lead electrocardiogram (eKG) and a serum electrolyte profile. eKG abnormalities were summarized in terms of frequencies,means, and standard deviations. electrolyte and enzyme levels were summarized as medians. Differences were analyzed using the Mann-Whitney U test. Results: Twenty-eight patients (55% ) had an abnormal eKG. Non specific changes of repolarization (20% ), sinus tachycardia (12% ), and left anterior hemiblock (10% ) were the most frequent disturbances. Patients with sinus tachycardia had lower levels of phosphorus (2.3 vs. 3.4 meq/L, P endency to higher levels of potassium and lower levels of phosphorus was found in patients with sinus tachycardia and nonspecific changes of repolarization, respectively. No differences were found in amylase, pancreatic amylase, or lipase among patients with normal and abnormal eKG. Conclusions: More than 50% of the patients with AP had eKG abnormalities, and these changes could be related to electrolyte alterations.

Application of the Distance to Natural (D2N) Index and Statistical Analysis of Different Anthropic Uses of the Río Grande De Comitán Watershed, Chiapas, México

The intensification of anthropic uses (i.e., increase of the hemerobic condition) threatens the remnants of native vegetation due to the reduction of its self-regulation capacity. In this research, the Distance to Nature (D2N) index for land use and land cover was applied in the Río Grande de Comitán watershed (Southern Mexico) to answer the following questions: 1) What were the land use dynamics observed in the Rio Grande de Comitán watershed in the trajectory through 1999, 2009 and 2019? 2) Does the subcategorization of the D2N allow one to identify which anthropic uses influence more the territorial expression of the watershed? To answer these questions, we performed a supervised classification of land use and land cover was performed in this watershed, and for the D2N index, the classification was simplified to three-category scale for the subcategorization of the anthropic component. Through Principal Component Analysis (PCA), we identified that agricultural anthropogenic use had the greatest influence on territorial expression. The reported scenario indicates a trend of gradual and continuous reduction of naturalness over the last 20 years. Additionally, the D2N index proved to be a useful tool to demonstrate both the anthropic impact, with the simplified scale, and the component that most influences the territory, by subcategorizing the anthropic scale.
