Macroalgae are multicellular,aquatic autotrophs that play vital roles in global climate maintenance and have diverse applications in biotechnology and eco-engineering,which are directly linked to their multicellularity ***,their genomic diversity and the evolutionary mechanisms underlying multicellularity in these organisms remain *** this study,we sequenced 110 macroalgal genomes from diverse climates and phyla,and identified key genomic features that distinguish them from their microalgal *** for cell adhesion,extracellular matrix formation,cell polarity,transport,and cell differentiation distinguish macroalgae from microalgae across all three major phyla,constituting conserved and unique gene sets supporting multicellular *** genes show phylum-and climate-specific expansions that may facilitate niche ***,our study reveals genetic determinants of convergent and divergent evolutionary trajectories that have shaped morphological diversity in macroalgae and provides genome-wide frameworks to understand photosynthetic multicellular evolution in aquatic environments.

Conceptualization and development of the Household/Neighborhood Model for skilled nursing facilities： A case study

The conceptualization of the HousehoLd/Neighborhood Model for skilled nursing facilities began in 1987 at Evergreen Retirement Community in Oshkosh, WI, USA in the search for the ＂perfect＂ nursing home. Being able to see perfection required being freed from the mental constraints of regulations, reimbursement systems, and existing staff training programs so that visionary thinking was possible. ConceptuaLization was followed by a pilot project to test various aspects of the vision. The result was （1） a management philosophy built on Continuous QuaLity Improvement, （2） a team-based organization structure, （3） cross-functionaL staff roles, （4） sociaL-modeL activity programming, and （5） a residential style physical setting that won strong support from the State of Wisconsin. With this support, we had the opportunity to open in 1997 the first fuLL-scaLe model of a HousehoLd/Neighborhood in the United States based upon providing ＂resident and relationship centered services and care＂. The mode[ was validated through a year-tong evaluative research study. In 2004 we opened a second full-scale model incorporating significant improvements identified through experience. The opportunity to further develop the model has been provided through its adoption in China by China Senior Care, which wiLL open their first facility in 2014.

Mantle-derived magmas:intraplate,hot-spots and mid-ocean ridges

Primary or parental magmas act as probes to infer eruption and source temperatures for both mid-ocean ridge （MOR） and‘hot-spot＇ magmas （tholeiitic picrites）. The experimental petrogenetic constraints （＇inverse＇ experiments） argue for no significant temperature differences between them. However, there are differences in major, minor and trace elements which characterise geochemical, not thermal, anomalies beneath ‘hot-spots＇. We suggest that diapiric upwelling from interfaces （redox contrasts） between old subducted slab and normal MOR basalt source mantle is the major reason for the observed characteristics of island chain or ‘hot-spot＇ volcanism. Intraplate basalts also include widely distributed volcanic centres containing lherzolite xenoliths, i.e. mantle-derived magmas. Inverse experiments on olivine basalt, alkali oli- vine basalt, olivine basanite, olivine nephelinite, olivine melilitite and olivine leucitite （lamproite） determined liquidus phases as a function of pressure, initially under anhydrous and CO2-absent conditions. Under C- and H-absent conditions, only tholeiites to alkali olivine basalts had O1 ＋ Opx 4-Cpx as high-pressure liquidus phases. Addition of H20 accessed olivine basanites at 2.5-3 GPa, ,- 1,200 ℃, but both CO2 and H20 were necessary to obtain saturation with O1, Opx, Cpx and Ga at 2.5-3.5 GPa for olivine nephelinite and olivine melilitite. The forward and inverse experimental studies are combined to formulate a petrogenetic grid for intraplate, ‘hot-spot＇ and MOR magmatism within the plate tectonics paradigm. The asthenosphere is geochemically zoned by slow upward migration of incipient melt. The solidus and phasestabilities of lherzolite with very small water contents （〈3,000 ppm） determine the thin plate behaviour of the oceanic lithosphere and thus the Earth＇s convection in the form of plate tectonics. Although islet transplantation for individuals with type I diabetes has been shown to yield superior blood glucose control, it remains inadequate for long-term control. This is partly due to islet injuries and stresses that can lead to beta cell loss. Inhibition of excess IL-1β activity might minimize islet injuries, thus preserving function. The IL-1 receptor antagonist （IL-1Ra）, an endogenous inhibitor of IL-1β, protects islets from cytokine-induced necrosis and apoptosis. Therefore, an imbalance between IL-1β and IL-1Ra might influence the courses of allogeneic and autoimmune responses to islets. Our group previously demonstrated that the circulating serine-protease inhibitor human alpha-1-antitrypsin （hAAT）, the levels of which increase in circulation during acute-phase immune responses, exhibits anti-inflammatory and islet-protective properties, as well as immunomodulatory activity. In the present study, we sought to determine whether the pancreatic islet allograft-protective activity of hAAT was mediated by IL- 1Ra induction. Our results demonstrated that hAAT led to a 2.04-fold increase in IL-1Ra expression in stimulated macrophages and that hAAT-pre-treated islet grafts exhibited a 4.851-fold increase in IL-1Ra transcript levels, which were associated with a moderate inflammatory profile. Unexpectedly, islets that were isolated from IL-1Ra-knockout mice and pre-treated with hAAT before grafting into wild-type mice yielded an increase in intragraft IL-1Ra expression that was presumably derived from infiltrating host cells, albeit in the absence of hAAT treatment of the host. Indeed, hAAT-pre-treated islets generated hAAT-free conditioned medium that could induce IL-1Ra production in cultured macrophages. Finally, we demonstrated that hAAT promoted a distinct phosphorylation and nuclear translocation pattern for p65, a key transcription factor required for IL- 1Ra expression.

DNA and silica-coated magnetic particles entangle and form visible aggregates under chaotropic conditions with a rotating magnetic field, in a manner that enables quantification of DNA by image analysis. As a means of exploring the mechanism of this DNA quantitation assay, nanoscale SiO2-coated Fe304 （Fe3O4@SiO2） particles are synthesized via a solvothermal method. Characterization of the particles defines them to be -200 nm in diameter with a large surface area （141.89 m2/g）, possessing superparamagnetic properties and exhibiting high saturation magnetization （38 emu/g）. The synthesized Fe3O4@SiO2 nanoparticles are exploited in the DNA quantification assay and, as predicted, the nanoparticles provide better sensitivity than commercial microscale Dynabeads for quantifying DNA, with a detection limit of 4 kilobase-pair fragments of human DNA. Their utility is proven using nanoparticle DNA quantification to guide efficient polymerase chain reaction （PCR） amplification of short tandem repeat loci for human identification.

Diarrhea and Hypokalemic Rhabdomyolysis Due to Apoptotic Colitis as the Initial Manifestation of Common-Variable Immunodeficiency in an Adolescent

We present the case of an adolescent who presented with rhabdomyolysis due to severe hypokalemia arising from chronic diarrhea. Initial evaluation for celiac disease, known to present in this manner, was negative. Further evaluation with colonoscopy showed a normal appearing colon but biopsies showed a significant number of apoptotic cells in the mucosal crypts supporting a diagnosis of apoptotic colitis. Investigation into the cause of apoptotic colitis resulted in a diagnosis of common variable immune deficiency due to a defect in the inducible T-cell costimulator (ICOS) gene. Physicians should be aware of this uncommon condition and the importance of mucosal biopsy despite the presence of normal appearing mucosa.

Predicting an Athlete’s Physiological and Haematological Response to Live High-Train High Altitude Training Using a Hypoxic Sensitivity Test

Purpose Elite endurance runners frequently utilise live high-train high(LHTH)altitude training to improve endurance per-formance at sea level(SL).Individual variability in response to the hypoxic exposure have resulted in contradictory *** the present case study,changes in total haemoglobin mass(tHbmass)and physiological capacity,in response to 4-weeks of LHTH were *** tested if a hypoxic sensitivity test(HST)could predict altitude-induced adaptations to *** Fifteen elite athletes were selected to complete 4-weeks of LHTH(~2400 m).Athletes visited the laboratory for preliminary testing(PRE),to determine lactate threshold(LT),lactate turn point(LTP),maximal oxygen uptake VO_(2max)and *** LHTH,athletes completed daily physiological measures[arterial oxygen saturation(SpO2)and body mass]and subjective wellbeing *** was repeated,for those who completed the full camp,post-LHTH(POST).Additionally,athletes completed the HST prior to *** A difference(P<0.05)was found from PRE to POST in average tHbmass(1.8%±3.4%),VO_(2max)(2.7%±3.4%),LT(6.1%±4.6%)and LTP(5.4%±3.8%),after 4-weeks *** revealed a decrease in oxygen saturation at rest(ΔSp_(r))and higher hypoxic ventilatory response at rest(HVRr)predicted individual changes *** hypoxic cardiac response at rest(HCRr)and higher HVRr predicted individual changes VO_(2max).Conclusion Four weeks of LHTH at~2400 m increased tHbmass and enhanced physiological capacity in elite endurance *** was no observed relationship between these changes and baseline characteristics,pre-LHTH serum ferritin levels,or reported incidents of musculoskeletal injury or *** HST did however,estimate changes in tHbmass and VO_(2max).HST prior to LHTH could allow coaches and practitioners to better inform the acclimatisation strategies and train-ing load application of endurance runners at altitude.
