Soil.organic carbon(SOC)is the l.rgest terrestrial.carbon(C)stock,and the capacity of soil. to preserve organic C(OC)varies with many factors,incl.ding l.nd use,soil.type,and soil.*** investigated the effect of l.nd use change on soil.particul.te organic matter(POM)and mineral.associated organic matter(MOM).Surface(0–10 cm)and subsurface(60–70 cm)sampl.s were col.ected from paired sites(native and cropped)of four contrasting *** soil. were separated into POM and MOM fractions,which were anal.zed for mineral.gy,OC,nitrogen,isotopic signatures,*** POM fractions of surface soil. were rel.tivel. unaffected by l.nd use change,possibl. because of the continuous input of crop residues,whereas the POM fractions in corresponding subsurface soil. l.st more *** surface soil.,MOM fractions dominated by the oxides of iron and al.minum(oxide-OM)l.st more OC than those dominated by phyl.osil.cates and quartz,which was attributed to diverse organic matter(OM)input and the extent of OC saturation l.mit of *** contrast,oxide-OM fractions were l.ss affected than the other two MOM fractions in the subsurface soil.,possibl. due to OC protection via organo-mineral.*** deviations in isotopic signature(l.nked with vegetation)across the fractions suggested that fresh crop residues constituted the bul. of OM in surface soil.(supported by greater14C).Increased isotopic signatures and l.wer14C in subsurface MOM fractions suggested the association of more microbial.y processed,aged OC with oxide-OM fractions than with the other MOM *** resul.s reveal.that the quantity and qual.ty of OC after l.nd use change is infl.enced by the nature of C input in surface soil. and by mineral.organic association in subsurface soil..

目的阐明大肠癌脾虚湿热证表型组和湿热蕴结证表型组的代谢组学机制。方法2021年3月—2021年10月于西南医科大学附属中医医院招募大肠癌脾虚湿热证患者68例、湿热蕴结证患者58例,同时纳入于此进行体检的健康平和质人员62名。建立脾虚湿... 详细信息

目的阐明大肠癌脾虚湿热证表型组和湿热蕴结证表型组的代谢组学机制。方法2021年3月—2021年10月于西南医科大学附属中医医院招募大肠癌脾虚湿热证患者68例、湿热蕴结证患者58例,同时纳入于此进行体检的健康平和质人员62名。建立脾虚湿热证表型组、湿热蕴结证表型组和健康平和质表型组队列,以超高液相色谱和质谱联用技术对各队列血清代谢物进行分离鉴定,并进行主成分分析(PCA)、正交偏最小二乘判别分析法(Ol.S-DA)分析及生物信息学分析。结果与健康平和质表型组相比,脾虚湿热证表型组有胍基乙酸下调,亚氨甲基谷氨酸、咪唑乙酸、脱氧尿苷、胞苷、假尿苷、鸟氨酸、N-乙酰鸟氨酸、5-羟吲哚乙酸、胆色素原、琥珀酸半醛、甜醇表达上调等12种差异代谢,涉及能量代谢异常为主的7条代谢途径;湿热蕴结证表型组有脱氧尿苷、胞苷、胆色素原、花生酸、前列腺素B2、异柠檬酸、古洛糖酸表达上调,胍基乙酸、尿苷、羟脯氨酸、吡咯-2-羧酸、腺苷、5,6-DHET、8,9-DiHETrE、9,10-DHOME、亚油酸、羟基丙酮酸表达下调等17种差异代谢,涉及脂类代谢和三羧酸循环异常为主的9条代谢途径;脾虚湿热证表型组与湿热蕴结证表型组存在4个相同特征性代谢物(胍基乙酸、脱氧尿苷、胞苷、胆色素原)差异。结论这些结果揭示了形成大肠癌脾虚湿热证和湿热蕴结证两个常见临床表型组的代谢组学机制,并为中医辨证和诊治提供临床表型组的循证依据和特异性代谢组学客观依据。

OBJECTIVE： l.gusticum ported is a traditional.Native American herb. The roots of l. ported are traditional.y used in the treatment of many diseases, however, its cytotoxicity, antioxidative and immune- modul.tory effects need to be investigated. In this study, we eval.ated the effects of the root extract at different doses on human peripheral.bl.od l.mphocytes （PBl.）. METHODS： The l.mphocytes were incubated with different concentrations of the root extracts （0, 50, 100, 200, and 400 μg/ml. and harvested every 6 h for 2 d （P〈0.05）. The protective effect of the herb against oxidative damage was determined by inducing oxidative stress with the administration of 50 μmol.l.of hydrogen peroxide （H202）. RESUl.S： Treatments with l. ported at 200 and 400 pg/ml.increased the viabil.ty of PBl.. The del.terious effect of H2O2 was amel.orated by 400μg/ml.l. ported treatment. Addition of 400 μg/ml.l. ported reduced l.pid peroxidation in stressed PBl. by 94% （P〈0.05）. Treatment with 400 μg/ml.of l. ported resul.ed in a 26.4% increase of reduced gl.tathione l.vel.. Activities of superoxide dismutase and catal.se increased by 17.5% and 55.2% respectivel., when stressed PBl. were treated with 400 μg/ml.l. ported for 2 d （P〈0.05）. Treatment with 400 μg/ml.l. ported increased interferon-γand interl.ukin-2 expressions in H2O2-chal.enged PBl. （P〈0.05）, however, the root extract did not cause a significant difference in interl.ukin-10 l.vel. compared to the control.（P〉0.05）. CONCl.SION： The findings suggest that l.invol.ing protective effects against oxidative ported might be a potential.immune-modul.ting agent damage.
