Anti-inflammatory effects of alkaloid enriched extract from roots of Eurycoma longifolia Jack

Objective: To examine the in vitro and in vivo anti-inflammatory effects of the alkaloid enriched extract(ELA) from the roots of Eurycoma longifolia. Methods: The in vitro antiinflammatory effects of ELA were evaluated by examining its inhibitory activities against nitric oxide(NO) production and inducible nitric oxide synthase(iN OS) and cyclooxygenase2(COX-2) expressions in lipopolysaccharide(LPS)-stimulated RAW264.7 cells. The level of NO produced in the culture media was determined by Griess method. The i NOS and COX-2 protein expressions were analyzed by Western blot. The in vivo effect of ELA was evaluated on LPS-induced septic shock in mice model. Mice mortality was monitored for5 days after injection of LPS. The chemical contents of the ELA were determined by using various chromatographic and spectroscopic techniques. Results: The ELA was found to exhibit a significant anti-inflammatory effect in both in vitro and in vivo models. The results demonstrated that ELA dose-dependently inhibited LPS-induced NO production as well as the protein iN OS and COX-2 expressions. In the septic shock model, ELA dose-dependently protected mice from LPS-induced mortality. Further study on the isolated components of ELA indicated that 9,10-dimethoxycanthin-6-one may contribute significantly to the antiinflammatory effects of the extract. Conclusions: These results suggest that ELA exhibits the anti-inflammatory activity via suppression of pro-inflammatory mediators such as NO, iN OS,and COX-2 and protects mice from LPS-induced mortality in septic shock model.

Land use type affects the physicochemical properties of *** value of OM and TN is high in acacia *** acidity of soil is considered a serious constraint for *** cash crops are acacia,cassava,banana,rice and *** fertility is affected by land-use types and land management,which exacerbates soil erosion and various other forms of soil degradation in the mountainous regions of *** study was conducted in A Luoi District,Thua Thien Hue,Vietnam to identify the effects of land-use types on specific soil physicochemical characteristics related to soil *** physicochemical properties,such as organic matter(OM),total nitrogen(TN),total phosphorous(TP),and K+were significantly affected by land-use *** results showed that the soils were sandy in rice but clay loam for acacia and *** mean bulk density value of acacia soil was significantly greater than that of other *** were higher in the acacia soils than those in the rice,maize,and banana *** OM content was significantly higher in the acacia,cassava,and banana soils than those in the rice and maize *** mean of exchangeable K+in the rice soil was higher compared to those in other soils and was affected by land-use *** high exchangeable acidity content in the soils was probably due to intensive ***,both land use type and management did not affect the CEC ***,the inappropriate land use caused the disturbance of soil physicochemical properties,indicating that the conditions of rice and maize soils are becoming worse than acacia ***,lowering the intensity of cultivation,adopting incorporated soil fertility management,and applying organic fertilizer should preserve the existing conditions and enhance soil properties.

Predicting 3D Radiotherapy Dose-Volume Based on Deep Learning

Cancer is one of the most dangerous diseaseswith *** of the principal treatments is radiotherapy by using radiation beams to destroy cancer cells and this workflow requires a lot of experience and skill from doctors and *** our study,we focused on the 3D dose prediction problem in radiotherapy by applying the deeplearning approach to computed tomography(CT)images of cancer *** image data has more complex characteristics than normal image data,and this research aims to explore the effectiveness of data preprocessing and augmentation in the context of the 3D dose prediction *** proposed four strategies to clarify our hypothesis in different aspects of applying data preprocessing and *** strategies,we trained our custom convolutional neural network model which has a structure inspired by the U-net,and residual blocks were also applied to the *** output of the network is added with a rectified linear unit(Re-Lu)function for each pixel to ensure there are no negative values,which are absurd with radiation *** experiments were conducted on the dataset of the Open Knowledge-Based Planning Challenge which was collected from head and neck cancer patients treatedwith radiation *** results of four strategies showthat our hypothesis is rational by evaluating metrics in terms of the Dose-score and the Dose-volume histogram score(DVH-score).In the best training cases,the Dose-score is 3.08 and the DVH-score is *** addition,we also conducted a comparison with the results of another study in the same context of using the loss function.

Effect of Freeze-Thaw and Urea in Solubility of GPC3-Csub Protein Expressed in Escherichia coli

Glypican-3 is a protein encoded by the Glypican-3 gene located on human X chromosome (Xq26), composed of two subunits, a 40 kDa N-terminal subunit, and a 30 kDa C-terminal subunit. Glypican-3 is a currently potential target molecule for liver cancer treatments because of its over-expression and growth effects on hepatocellular carcinoma (HCC). this study examined the expression and purification of a C-terminal subunit of Glypican-3 protein (GPC3-Csub) due to its application in both diagnosis and therapy for hepatocellular carcinoma. The gene encoding for GPC3-Csub was successfully cloned into plasmid pET28a fused with an affinity tag composed of six consecutive histidine residues (His-tag). Recombinant protein GPC3-Csub was expressed in Escherichia coli BL21 (DE3) in the condition of adding 3% ethanol with IPTG induction. GPC3-Csub was extracted using repeated freeze-thaw cycles with lysozyme, and inclusion bodies were solubilized by 8M Urea, SDS 10% in pH 12. His-tag fused GPC3-Csub proteins allowed it to be purified by affinity chromatography method using the Nickel-nitrilotriacetic acid (Ni-NTA) column. High expression of GPC3-Csub was confirmed by Coomassie staining and western-blot. GPC3-Csub could be isolated with a Ni-NTA column and have a purity of about 90%.

Recently, air quality has significantly improved in developed country, but that issue is of concern in emerging megacity in developing *** this study, aerosols and their precursor gas were collected by NILU filter pack at two distinct urban sites during the winter and summer in Osaka, Japan and dry and rainy seasons in Ho Chi Minh City(HCMC),*** aims are to investigate the contribution of water-soluble inorganic ions(WSIIs) to PM2.5, thermodynamic characterization and possible formation pathway of secondary inorganic aerosol(SIA).The PM2.5 concentration in Osaka(15.8 μg/m^3) is lower than that in HCMC(23.0 μg/m^3), but the concentration of WSIIs in Osaka(9.0 μg/m^3) is two times higher than that in HCMC(4.1 μg/m^3).Moreover, SIA including NH4^+, NO3^-and SO4^2-are major components in WSIIs accounting for 90% and 76%(in molar) in Osaka and HCMC,*** models were used to understand the thermodynamic characterization of urban ***, statistical analysis results indicate that very good agreement(R2> 0.8) was found for all species, except for nitrate aerosol in *** found that when the crustal species present at high amount, those compositions should be included in model calculation(*** the HCMC situation).Finally, we analyzed the characteristics of NH4^+– NO3^-– SO4^2-system.A possible pathway to produce fine nitrate aerosol in Osaka is via the homogeneous reaction between NH3 and HNO3, while nonvolatile nitrate aerosols can be formed by the heterogeneous reactions in HCMC.

A porous crosslinked organic polymer based on N-acryloxysuccinimide （NAS） and ethylene dimethacrylate （EDMA） was prepared inside 75 μm i.d. fused silica capillary as functionalizable monolithic stationary phase for electrochromatographic applications. Succinimide groups on the monolith surface provide reactive sites able to re- act readily through standard electrophile-nucleophile chemistry. Propargylamine was used to prepare alkyne func- tionalized poly（NAS-co-EDMA）. Onto this thiol-reactive polymer surface was grafted adamantane units via a photochemically-driven addition reaction. Chemical characterization was performed in situ after each synthetic step by means of Raman spectroscopy and grafting kinetics was investigated to ensure quantitative grafting of 1-adamantanethiol. The as-designed monolithic stationary phase exhibited typical reversed-phase separation mechanism as evidenced by the linear increase of the logarithm of retention factor of neutral aromatic solutes with the increase of the aqueous buffer content in the mobile phase.

Assessment of Livelihood Vulnerability to Drought: A Case Study in Dak Nong Province, Vietnam

In recent years,droughts have strongly affected the Central Highlands of Vietnam and have resulted in crop damage,yield decline,and serious water *** study investigated the livelihood vulnerability of five communities of farmers who are exposed to droughts in one of the more vulnerable regions of Vietnam—Dak Nong Province.A survey of 250 households was conducted in the five communities to collect data on the region’s sociodemographic profile,livelihood systems,social networks,health status,food and water security,drought conditions,and climate *** were aggregated using a livelihood vulnerability index and the IPCC vulnerability *** survey results indicate that Quang Phu community is the most vulnerable of the study’s communities,followed by Nam N’dir,Dak Nang,Duc Xuyen,and Dak D’ro in descending order of *** availability and livelihood strategies are the most important variables in determining the vulnerability of the five surveyed *** order to reduce vulnerability to droughts,water management practices and livelihood diversification in farming and nonfarming activities are recommended for the study area.

Evaluation of Weathering Performance of Rosin-Copper Based Treated Wood

this study aimed to evaluate the effect of natural weathering on some surface characteristics and mechanical properties of Styrax wood treated with mixtures of 1.0%,2.0%,or 4.0%rosin sizing agent and 3%copper *** samples after treatment were exposed to outdoor conditions for one year and changes in color,glossiness,weight loss,compression strength parallel to grain(CSPG),modulus of rupture(MOR),and modulus of elasticity(MOE)were investigated after 6 and 12-month of natural *** results showed that rosin-copper treatment could improve color stability and gloss of samples after *** losses of all rosin-copper treated samples after 12 months of exposure were negligible compared with the untreated control *** addition,rosin-copper treatment enhanced the CSPG compared to untreated controls,but slightly decreased the MOR and MOE of Styrax wood before *** weathering factors induced a reduction for all strength properties,however,the decrease rate for strength properties of rosin-copper treated samples was negligible compared with the untreated control samples after 12-month natural weathering *** FTIR and SEM-EDX confirmed that the use of the rosin-copper formulations to impregnate wood could decrease the hazard of the copper preservative leaching into the environment,while also enhancing more resistance against weathering factors and other biotic factors.
