Protein s-glutathionylation, the reversible formation of a mixed.d.sulfid. between glutathione and.protein thiols, is involved.in protection of protein cysteines from irreversible oxid.tion, but also in protein red.x regulation. Recent stud.es have implicated.s-glutathionylation as a cellular response to oxid.tive/nitrosative stress, likely playing an important role in signaling. Consid.ring the potential importance of glutathionylation, a number of method. have been d.veloped.for id.ntifying proteins und.rgoing glutathionylation. These method., ranging from analysis of purified.proteins in vitro to large-scale proteomic analyses in vivo, allowed.id.ntification of nearly 200 targets in mammals. By contrast, the number of known glutathionylated.proteins is more limited.in photosynthetic organisms, although they are severely exposed.to oxid.tive stress. The aim of this review is to d.tail the method. available for id.ntification and.analysis of glutathionylated.proteins in vivo and.in vitro. The ad.antages and.d.awbacks of each technique will be d.scussed.as well as their application to photosynthetic organisms. The importance of sorting proteins and.wall materials to their d.stination is critical for plant growth and.d.velopment, though the machinery orchestrating membrane trafficking is poorly und.rstood. Transporters that alter the environment across end.membrane compartments are thought to be important players. Using Escherichia coli and.yeast, we previously showed.that several Arabid.psis Cation/H＋ eXchanger （AtCHX） members were K＋ transporters with a role in pH homeostasis, though their subcellular location and.biological roles in plants are unclear. Co-expression of markers with CHX16, CHX17, CHX18, or CHX19 tagged.with a fluorescent protein ind.cated.these transporters associated.with plasma membrane （PM） and.post-Golgi compartments. Und.r its native promoter, AtCHX17（l_820）-GFP localized.to prevacuolar compartment （PVC） and.to PM in roots. Brefeld.n A d.minished.AtCHX17- GFP fluorescence at PM, whereas wortmannin caused.formation of GFP-labeled.ring-like structures, suggesting AtCHX17 trafficked.among PVC, vacuole and.PM. AtCHX17（1-472） lacking its carboxylic tail d.d.not associate with PVC or PM in plant cells. single chx17 mutant or higher-ord.r mutants showed.normal root growth and.vegetative d.vel- opment. However, quad.uple （chx16chx17chxlschx19） mutants were red.ced.in frequency and.prod.ced.50%-70% fewer seed., ind.cating overlapping roles of several AtCHX17-related.transporters in reprod.ction and.or seed.d.vel- opment. A method.for id.ntification of pulsations in time series of magnetic field.d.ta which are simultaneously present in multiple channels of d.ta at one or more sensor locations is d.scribed. Cand.d.te pulsations of interest are first id.ntified.in geomagnetic time series by inspection. Time series of these ＂training events＂ are represented.in matrix form and.transpose-multiplied.to generate time- d.main covariance matrices. The ranked.eigenvectors of this matrix are stored.as a feature of the pulsation. In the second.stage of the algorithm, a slid.ng wind.w （approxi- mately the wid.h of the training event） is moved.across the vector-valued.time-series comprising the channels on which the training event was observed. At each wind.w position, the d.ta covariance matrix and.associated.eigen- vectors are calculated. We compare the orientation of the d.minant eigenvectors of the training d.ta to those from the wind.wed.d.ta and.flag wind.ws where the d.minant eigenvectors d.rections are similar. This was successful in automatically id.ntifying pulses which share polarization and.appear to be from the same source process. We apply the method.to a case stud. of continuously sampled.（50 Hz） d.ta from six observatories, each equipped.with three- component ind.ction coil magnetometers. We examine a 90-d.y interval of d.ta associated.with a cluster of four observatories located.within 50 km of Napa, California, together with two remote reference stations-one 100 km to the north of the cluster and.the other 350 km south. When the training d.ta contains signals present in the remote reference observatories, we are reliably able to id.ntify and.extract global geomagnetic signals such as solar-generated.noise. When training d.ta contains pulsations only observed.in the cluster of local observatories, we id.ntify several types of non-plane wave signals having similar polarization.

Process control parameters influencing microbial perchlorate red.ction via a flow-through zero-valent iron （ZVI） column reactor were investigated.in ord.r to optimize perchlorate removal from water. Mixed.perchlorate red.cers were obtained.from a wastewater treatment plant and.inoculated.into the reactor without further acclimation. Examined.parameters includ.d.hyd.aulic resid.nce time （HRT）, pH, nutrients requirement, and.perchlorate red.ction kinetics. The minimum HRT for the system was conclud.d.to be 8 hr. The removal efficiency of 10 mg. L-1 influent perchlorate concentration was red.ced.by 20%-80% without control to the neutral pH （HRT = 8 hr）. Therefore pH was d.termined.to be an important parameter for microbial perchlorate red.ction. Furthermore, a viable alternative to pH buffer was d.scussed. The microbial perchlorate red.ction followed.the first ord.r kinetics, with a rate constant （K） of 0.761 hr-1. The results from this stud. will contribute to the implementation of a safe, cost effective, and.efficient system for perchlorate red.ction to below regulated.levels.

Yellow maize contains high levels of β-carotene(βC), making it an important crop for combating vitamin A d.ficiency through biofortification. In this stud., nine maize inbred.lines were selected.at rand.m from 31 provitamin A(PVA) maize inbred.lines and.crossed.in a partial d.allel mating d.sign to d.velop 36 crosses. The crosses were evaluated.in the field.in two locations(samaru and.Kerawa) and.their seed.carotenoid.content were d.termined.by high-performance liquid.chromatography. The mod.s of gene action, heritability, and.correlations between agronomic traits and.carotenoid.content were estimated. Ad.itive genetic variances(σ~2a) were lower than non-ad.itive genetic variances(σ~2d. for all the carotenoid., plant height(PH), and.grain yield.GY), suggesting a prepond.rance of non-ad.itive gene action. Broad.sense heritability(H^2) was high(H^2> 60%) for zeaxanthin,d.ys to anthesis, and.PH, mod.rate(30% d.GY, and.low(H^2d.PVA. Genetic ad.ance as a percentage of mean, consid.red.with H^2, also suggests a prepond.rance of non-ad.itive gene action for PVA carotenoid.. Hybrid.variety d.velopment is thus an appropriate approach to improving grain yield.and.PVA. GY showed.no significant genotypic correlations with carotenoid.content, suggesting that these traits can be improved.concurrently. Thus, there is ample scope for improvement of PVA and.GY in the sample of tropical-ad.pted.maize.
