A phase Ⅰ study of Hemay022, an irreversible dual EGFR/HER2 tyrosine kinase inhibitor in Chinese patients with HER2-positive advanced breast cancer

Objective: Hemay022 is a novel small-molecule and an irreversible tyrosine kinase inhibitor with the target of epidermal growth factor receptor(EGFR)/human epidermal growth factor receptor 2(HER2), which demonstrated anti-tumor activity in preclinical studies. This first-in-human study evaluated the safety, pharmacokinetics,tolerability and preliminary anti-tumor activity of Hemay022 in HER2-positive advanced breast cancer ***: Heavily pretreated patients with HER2-positive advanced breast cancer were assigned to eight dose cohorts in a 3+3 dose-escalation pattern at doses of 50-600 mg QD and 300 mg BID. eligible patients were given a single dose of Hemay022 on d 1 in week 0, followed by once daily continuous doses for four weeks in 28-day *** samples were obtained on d 1 and d 28. Clinical responses were assessed every eight ***: Twenty-eight patients with advanced breast cancer were treated with Hemay022. The most frequently reported drug-related adverse events were diarrhoea(85.7%), vomiting(28.6%), nausea(25.0%) and decreased appetite(17.9%). No grade 4 drug-related adverse events were reported. At 50-600 mg doses, steady state areas under the concentration-time curve and peak concentrations increased with doses. One patient achieved complete response(CR), and three achieved partial response(pR). The objective response rate(ORR) and disease control rate(DCR) were 14.3% and 46.4% in 28 patients, respectively. The median progression-free survival(pFS) was3.98 ***: Hemay022 at the dose of 500 mg once daily was well tolerated. The pharmacokinetic properties and encouraging anti-tumor activities of Hemay022 in advanced breast cancer patients warranted further evaluation of Hemay022 for treating breast cancer patients in the current phase Ⅲ trial(No. NCT05122494).

The assembly of exogenous artificial architectures inside cells can regulate a series of biological events,which heavily relies on the development of spatiotemporally controlled molecular assembly *** herein report a designer deoxyribonucleic acid(DNA)nanostructure that enables light-mediated spatiotemporally dynamic assembly in living cells and consequently achieves efficient regulation of cell *** DNA nanostructure was constructed from i-motif moiety-containing branched DNA,photocleavable bond-containing linker,and tumor cell-targeting *** cellular uptake mediated by aptamers,under the spatiotemporal control of both UV light and late endosomal/lysosomal acidic environments,disassembly/reassembly of DNA nanostructure occurred via two rationally designed routes,generating microsized DNA *** a result,autophagy was significantly enhanced with the increase of DNA assembly *** enhanced autophagy showed an impact on related biological *** system is expected to be a powerful tool for the regulation of intracellular events and cellular behaviors.

The loss of hydrocarbon production caused by the dynamic behavior of the inner boundary and propped fractures under long-term production conditions has been widely reported in recent studies. However,the quantitative relationships for the variations of the inner boundary and propped fractures have not been determined and incorporated in the semi-analytical models for the pressure and rate transient analysis. This work focuses on describing the variations of the inner boundary and propped fractures and capturing the typical characteristics from the pressure transient curves.A generalized semi-analytical model was developed to characterize the dynamic behavior of the inner boundary and propped fractures under long-term production conditions. The pressure-dependent length shrinkage coefficients, which quantify the length changes of the inner zone and propped fractures, are modified and incorporated into this multi-zone semi-analytical model. With simultaneous numerical iterations and numerical inversions in Laplace and real-time space, the transient solutions to pressure and rate behavior are determined in just a few seconds. The dynamic behavior of the inner boundary and propped fractures on transient pressure curves is divided into five periods: fracture bilinear flow(FR1),dynamic pFs flow(FR2), inner-area linear flow(FR3), dynamic inner boundary flow(FR4), and outer-area dominated linear flow(FR5). The early hump during FR2 period and a positive upward shift during FR4period are captured on the log-log pressure transient curves, reflecting the dynamic behavior of the inner boundary and propped fractures during the long-term production *** transient pressure behavior will exhibit greater positive upward trend and the flow rate will be lower with the shrinkage of the inner boundary. plant virus causes massive crop losses ***,there is currently no effective measure to control plant viral ***,we identify an antiviral protein Rhp-pSp,produced by the bacterial Rhodopseudomonas palustris strain *** this study,we discover that the antiviral activity of Rhp-pSp relies on its endoribonuclease *** the arginine(R)residue at position 129 onto alanine(A)abolishs its endoribonuclease activity on coat protein(Cp)RNA of tobacco mosaic virus(TMV),consequentially,compromises the antiviral activity of *** investigation demonstrates that,the mutant Rhp-pSp^(R129A)is unable to form the homotrimer as the wild type,indicating the importance of quaternary junction for the endoribonuclease *** of Rhp-pSp in Nicotiana benthamiana significantly enhances the resistance against TMV of seedlings,while expression of Rhp-pSp^(R129A)did not,confirming that endoribonuclease activity is responsible for the antiviral activity of *** addition,foliar spray of Rhp-pSp solution on tomato and pepper plants significantly reduces the disease index of viral diseases,indicating that Rhp-pSp shows potential to develop antiviral agent in practice.
