Pain inhibition through transplantation of fetal neuronal progenitors into the injured spinal cord in rats

Neuropathic pain after spinal cord injury(SCI) is a com.lex condition that responds poorly to usual treatm.nts. Cell transplantation represents a prom.sing therapy;nevertheless, the ideal cell type in term. of neurogenic potential and effectiveness against pain rem.ins largely controversial. Here, we evaluated the ability of fetal neural stem.cells(fNSC) to relieve chronic pain and, secondarily, their effects on m.tor recovery. Adult Wistar rats with traum.tic SCI were treated, 10 days after injury, with intra-spinal injections of culture m.dium.sham. or fNSCs extracted from.telencephalic vesicles(TV group) or the ventral m.dulla(Vm.group) of E/14 em.ryos. Sensory(von Frey filam.nts and hot plate) and m.tor(the Basso, Beattie,Bresnahan locom.tor rating scale and inclined plane test) assessm.nts were perform.d during 8 weeks. Thereafter, spinal cords were processed for im.unofluorescence and transplanted cells were quantified by stereology. The results showed im.rovem.nt of therm.l hyperalgesia in the TV and Vm.groups at 4 and 5 weeks after transplantation, respectively. m.reover, m.chanical allodynia im.roved in both the TV and Vm.groups at 8 weeks. No significant m.tor recovery was observed in the TV or Vm.groups com.ared with sham. Stereological analyses showed that ~70% of TV and Vm.cells differentiated into NeuN+ neurons,with a high proportion of enkephalinergic and GABAergic cells in the TV group and enkephalinergic and serotoninergic cells in the Vm.group. Our study suggests that neuronal precursors from.TV and Vm. once im.lanted into the injured spinal cord, m.turate into different neuronal subtypes, m.inly GABAergic, serotoninergic, and enkephalinergic, and all subtypes alleviate pain, despite no significant m.tor recovery. The study was approved by the Anim.l Ethics Com.ittee of the m.dical School of the University of S?o Paulo(protocol num.er 033/14) on m.rch 4, 2016.

Depth determ.nation of the m.ho interface beneath the Tibetan plateau and other areas of China

We apply the adaptive m.ving window m.thod of Sun et al. to the m.st recent catalog data and the data recorded by portable stations to construct the velocity structure of the crust and upper m.ntle, and to determ.ne the depth of the m.ho interface beneath the Tibetan plateau and other areas of China. We first select 2 600 locations in the study region with 1° intervals, then at each location invert for a five-layer 1-D P-wave velocity m.del from.the surface down to the upperm.st m.ntle by perform.ng a m.nte Carlo random.search. The m.ho depth at each location is then determ.ned, and the m.ho interface beneath the study region is obtained through proper interpolation with certain sm.othing. Com.ared to depths obtained by previous studies, our results show m.re accurate m.ho depths in the Tibetan plateau, Tianshan region and other areas of the study region.

The detailed electrical conductivity m.asure- m.nt (ECm., trace chem.cal com.ositions and m.croparti- cles concentration analysis are perform.d for BH8 ice core from.the depth of 126.0m.to 130.0m.at Vostok Station. At depth 128.7m. a volcanic signal 4726 a B.P. is detected. The volcanic sulphate flux is 95.8 kg·km.2, sulphate peak concen- tration 1352.8 ng·g?1, duration tim. about 10.1 years, com. parable with som. well-known volcanic events. The results indicate that it seem. to be a relatively large scale, long last- ing volcanic signal with farther volcanic origin.

Background: m.jor burns are life threatening. Fluid resuscitation is required for survival to m.intain intravascular volum.s and prevent hypovolem.c shock. Bioim.edance spectroscopy (BIS) has been recognised as a potential m.thod of m.nitoring fluid shifts after burn and in other disease states. The aim. of this study were to exam.ne the reliability of BIS across different dressing conditions and electrode positions, establish the influence of Acticoat?on BIS variable m.asures and determ.ne the validity of whole-body BIS to assess net fluid shift in the presence of m.derate to m.jor burns. m.thods: An observational longitudinal cohort study was conducted from.Decem.er 2014 to February 2016. Patients with over 15% total body surface area (TBSA) burns and injury less than 48 h were enrolled in the study. BIS triplicate m.asures were collected in an open wound and with an ActicoatTm.ressing (at 5 half hour intervals). Standard and alternate electrode placem.nts were utilised for the reliability analysis and standard placem.nt only for determ.ning the validity of BIS in m.derate to m.jor burns. The Im.edim.e SFB7 was used to collect whole-body and segm.ntal BIS m.asures. Stata statistical software, release 14 was utilised to analyse all results. Descriptive analyses were perform.d and were reported using the m.ans and standard deviations (SD). Results: BIS-repeated m.asures established BIS raw resistance (R), and predicted volum. variables were reliable in any condition (intra-class correlation coefficient (ICC) 0.996-0.999, 95% confidence intervals (CI) 0.996-0.999) without a system.tic difference. Acticoat?dressings significantly influenced all BIS-predicted volum.s (p≤0.01) as determ.ned by m.ltilevel m.xed effects (m.m.) linear regression analysis. Age-related Ecto-Nicotinam.de Adenine Dinucleotide Oxidase Disulfide Thiol Exchangers 3 (ENOX3) or age-related NADH oxidases (arNOX) are expressed at the cell surface as five m.m.ers of the Tm.9 superfam.ly, initially m.m.rane anchored, all functionally sim.lar, with the N-term.ni exposed at the cell’s exterior. ECTO-NOXes are cell surface proteins with both tim.-keeping CoQH2 [NAD(P)H] oxidase and protein disulfidethiol interchange activities. They are designated as ECTO-NOX proteins because of their localization on the outer surface of the plasm. m.m.rane and to distinguish them.from.the phox-NOXes of host defense. A ca. 30 kDa N-term.nal fragm.nt is cleaved and accum.lates in body fluids (serum. saliva, urine, perspiration). arNOXes appear around age 30 and increase steadily thereafter. Reduced quinones, i.e., reduced coenzym. Q, of the plasm. m.m.rane are natural substrates. NAD(P)H is oxidized as an artificial substrate. In one phase of the arNOX cycle electrons are transferred to oxygen to generate superoxide. Substrates for the shed form. of arNOX appear to be proteins of body fluids. Circulating lipoproteins and skin m.trix proteins em.rge as potentially im.ortant health-related targets. Through oxidation of collagen, elastin and other proteins of the skin m.trix, arNOXes are m.jor contributors to skin aging through tyrosine and thiol oxidation and subsequent cross linking. The m.in destructive action of arNOX, however, m.y be to directly oxidize circulating lipoproteins. arNOX in the blood is structured as an integral com.onent of the LDL particle through site-specific binding. As such, arNOXes are im.licated as m.jor risk factors for cardiovascular disease due to specific oxidation of LDLs. The superoxide produced and its conversion to hydrogen peroxide would be one part of the potentially destructive properties by contribution to lipid oxidation. Background： Objectives were to exam.ne the effects of selenium.（Se） supply and m.ternal nutritional plane during gestation on m.m.ary gland growth, cellular proliferation, and vascularity at parturition and d 20 of lactation. Ram.ouillet prim.parous ewes （n = 84） were allocated to treatm.nts in a 2 x 3 factorial. Factors were dietary Se （adequate Se [ASe, 11.5 μg/kg BW] or high Se [HSe, 77.0 μg/kg BVV]） and nutritional plane （60% IRES], 100% [CON], or 140% [EXC]）. At parturition, lam.s were rem.ved and 42 ewes （7/treatm.nt） were necropsied. Rem.ining ewes were fed a com.on diet m.eting requirem.nts for lactation and m.chanically m.lked twice daily until necropsy on d 20. At both necropsy periods, m.m.ary glands were dissected and tissues harvested. Sam.les were analyzed for RNA, DNA, and protein content, cell proliferation, and vascularity. Where interactions were present （P 〈 0.05）, least squares m.ans from.the highest-order interaction are presented. Results： Final body weight of ewes was least （P 〈 0.002） in RES, interm.diate for CON, and greatest for EXC, regardless of stage of the ewe at necropsy （parturition or d 20 of lactation）. In ewes necropsied at parturition, m.m.ary glands were heavier （P = 0.02） in EXC com.ared to RES, with CON interm.diate. Concentration of RNA （rag/g） was decreased （P= 0.01） in EXC com.ared to CON at parturition. There was a tendency （P= 0.07） for a Se by nutrition interaction in percentage of cells proliferating where ASe-EXC ewes had greater （P_〈 0.02） num.er of proliferating cells then all other treatm.nts. m.m.ary vascular area tended （P = 0.08） to be affected by a Se by nutrition interaction where ASe-CON had less （P= 0.007） vascular area than HSe-CON ewes. In ewes necropsied at d 20 of lactation, the num.er of alveoli per area was decreased （P 〈- 0.05） in RES com.ared to CON and EXC-fed ewes.
