In the classical microbial isolation technique,the isolation process inevitably destroys all microbial interactions and thus makes it difficult to culture the many microorganisms that rely on these interactions for *** this study,we designed a simple coculture technique named the“sandwich agar plate method,”which maintains microbial interactions throughout the isolation and pure culture *** total yield of uncultured species in sandwich agar plates based on eight helper strains was almost 10-fold that of the control *** uncultured species displayed commensal *** study found that heme was the growth-promoting factor of some marine commensal *** genomic analysis revealed that heme auxotrophies were common in various biotopes and prevalent in many uncultured microbial ***,our study supported that the survival strategies of heme auxotrophy in different habitats varied *** findings highlight that cocultivation based on the“sandwich agar plate method”could be developed and used to isolate more uncultured bacteria.

Many marine bacteria are difcult to culture because they are dormant,rare or found in *** culturing has been widely tested as an important strategy to isolate rare or dormant ***,many more mechanisms remain ***,based on 16S rRNA gene high-throughput sequencing and metabolomics technology,it was found that the short-chain fatty acids(SCFAs)in metabolites were signifcantly correlated with uncultured bacterial groups during enrichment cultures.A pure culture analysis showed that the addition of SCFAs to media also resulted in high efciency for the isolation of uncultured strains from marine *** a result,238 strains belonging to 10 phyla,26 families and 82 species were successfully *** uncultured rare taxa within Chlorobi and Kiritimatiellaeota were successfully *** the newly isolated uncultured microbes,most genomes,***,possess SCFA oxidative degradation genes,and these features might aid these microbes in better adapting to the culture media.A further resuscitation analysis of a viable but non-culturable(VBNC)Marinilabiliales strain verifed that the addition of SCFAs could break the dormancy of Marinilabiliales in 5 days,and the growth curve test showed that the SCFAs could shorten the lag phase and increase the growth ***,this study provides new insights into SCFAs,which were frst studied as resuscitation factors in uncultured marine ***,this study can help improve the utilisation and excavation of marine microbial resources,especially for the most-wanted or key players.

BACKGROUND Gallbladder neuroendocrine carcinoma(NEC)represents a subtype of gallbladder malignancies characterized by a low incidence,aggressive nature,and poor *** its clinical severity,the genetic alterations,mechanisms,and signaling pathways underlying gallbladder NEC remain *** SUMMARY This case study presents a rare instance of primary gallbladder NEC in a 73-year-old female patient,who underwent a radical cholecystectomy with hepatic hilar lymphadenectomy and resection of liver segments IV-B and *** gene sequencing and bioinformatics analysis tools,including STRING,GeneMANIA,Metascape,TRRUST,Sangerbox,cBioPortal and GSCA,were used to analyze the biological functions and features of mutated genes in gallbladder *** mutations(APC,ARID2,IFNA6,KEAP1,RB1,SMAD4,TP53,BTK,GATA1,GNAS,and PRDM3)were identified,and the tumor mutation burden was determined to be 9.52 muts/Mb via targeted gene sequencing.A protein-protein interaction network showed significant interactions among the twelve mutated *** Ontology and Kyoto Encyclopedia of Genes and Genomes analyses were used to assess mutation functions and *** results revealed 40 tumor-related pathways.A key regulatory factor for gallbladder NEC-related genes was identified,and its biological functions and features were compared with those of gallbladder *** Gallbladder NEC requires standardized *** with other gallbladder carcinomas revealed clinical phenotypes,molecular alterations,functional characteristics,and enriched pathways.

服务器无感知计算是新兴的云计算模式,它基于“函数即服务(FaaS)”的范式,以函数为部署和调度的基本单位,为用户提供大规模并行和自动伸缩的函数执行服务,且无需用户管理底层资源.对于用户,服务器无感知计算能够帮助他们摆脱集群底层基础设施管理的负担,专注于业务层的开发和创新;对于服务提供商,服务器无感知计算将应用分解为细粒度的函数,极大地提高了调度效率和资源利用率.显著的优势让服务器无感知计算迅速吸引了业界的注意,然而,服务器无感知计算与传统云计算迥然不同的计算模式以及对任务各方面的严格限制给应用的迁移带来了诸多障碍,各种越来越复杂的任务也对服务器无感知计算的性能提出了越来越高的要求,服务器无感知计算的性能优化成为一个重要的研究课题.从4个方面对服务器无感知计算系统性能优化技术的相关研究工作进行梳理和综述,并介绍现有的系统实现.(1)介绍面向典型任务的优化技术,包括任务适配和针对特定任务的系统优化;(2)综述沙箱环境的优化工作,包括沙箱方案和冷启动优化技术,它们是决定函数运行速度的核心;(3)概括I/O和通信技术的优化,它们是服务器无感知计算应用程序的主要性能瓶颈;(4)简述相关的资源调度技术,包括面向平台和面向用户的调度策略,它们决定着系统的资源利用率和任务的执行效率.最后,总结当前服务器无感知计算性能优化技术所面临的问题和挑战,并展望未来可能的发展方向.

Diabetes,a prevalent chronic metabolic disorder,often leads to severe ***,existing treatment methods may pose life-threatening risks due to poor patient compliance and inaccurate dosing of subcutaneous insulin ***,a biomimetic red blood cell(RBC)membrane-coated glucose-responsive nanoplatform is developed for controlling insulin *** nanoplatforms with RBC membrane can prolong the half-life of nano-formulation in vivo mediated by the biomimetic immune ***,the cascade catalytic effect of glucose oxidase(GOx)encapsulated in metal-organic frameworks(MOFs)and hemoglobin(Hb)in the RBC membrane are able to not only facilitate glucose-responsive insulin release,but also eliminate the detrimental by-product hydrogen peroxide(H_(2)O_(2))resulting from the Hb mediated H_(2)O_(2)*** in vitro and in vivo studies have demonstrated the favorable glucose-responsive performances of this advanced nano-platform with a single intravenous injection maintaining blood glucose balance in Type 1 Diabetes(T1D)mice for an extended duration without the hypoglycemia ***,this biomimetic insulin delivery system is poised to function as a strategy for the intravenous insulin administration,offering a promising drug candidate for the self-adaptive long-term T1D treatment.

Series of novel derivatives of desloratadine designed as arginine vasopressin receptor antagonists were synthesized and structurally characterized by melting points,^1H NMR and *** in vivo diuretic activities were evaluated on rats,and several target compounds showed promising diuretic results, especially compounds 8,18,27 and *** in vitro bonding assay and cAMP assay showed that these compounds had a higher affinity to vasopressin V2 receptor than VI a *** studies indicated that desloratadine may be an active substructure for novel arginine vasopressin receptor antagonist development.

随着交通日益发达和机动车保有量跃升,公路沿线农田土壤多环芳烃(PAHs)污染问题不容忽视.通过搜集近16 a来我国各地路域农田土壤污染的研究数据,对PAHs的整体污染水平、组成特征、主要来源、地域分布以及生态健康风险进行了系统分析.结果表明,我国路域农田土壤ω(∑_(16)PAHs)在86.40~3 568.00μg·kg^(-1)范围内,中位值为490.15μg·kg^(-1);PAHs各单体含量(以中位值计)在5.55~47.39μg·kg^(-1)范围内,其中菲的含量最高;PAHs组成以中高环(4~6环)为主,占总PAHs的56.81%,多数省份处于“轻度污染”甚至更高的污染程度.全国范围内PAHs的空间分布差异明显,污染程度从高到低依次为东北和西北地区、中部和东南沿海地区、西南地区.源解析结果表明,路域农田土壤中PAHs污染主要来自石油和天然气等燃料的混合燃烧源,污染程度与当地的石油、煤炭储量之间存在较强的正相关性.通过风险商值法、毒性当量法和健康风险评估模型这3种不同的评价方法对PAHs污染情况进行了生态和健康风险综合分析,结果显示出我国整体路域农田土壤的PAHs生态风险处于中度污染水平,而健康风险较为乐观.

Ghost Module Based Residual Mixture of Self-Attention and Convolution for Online Signature Verification

Online Signature Verification (OSV), as a personal identification technology, is widely used in various ***, it faces challenges, such as incomplete feature extraction, low accuracy, and computational heaviness. Toaddress these issues, we propose a novel approach for online signature verification, using a one-dimensionalGhost-ACmix Residual Network (1D-ACGRNet), which is a Ghost-ACmix Residual Network that combines convolutionwith a self-attention mechanism and performs improvement by using Ghost method. The Ghost-ACmix Residualstructure is introduced to leverage both self-attention and convolution mechanisms for capturing global featureinformation and extracting local information, effectively complementing whole and local signature features andmitigating the problem of insufficient feature extraction. Then, the Ghost-based Convolution and Self-Attention(ACG) block is proposed to simplify the common parts between convolution and self-attention using the Ghostmodule and employ feature transformation to obtain intermediate features, thus reducing computational ***, feature selection is performed using the random forestmethod, and the data is dimensionally reducedusing Principal Component Analysis (PCA). Finally, tests are implemented on the MCYT-100 datasets and theSVC-2004 Task2 datasets, and the equal error rates (EERs) for small-sample training using five genuine andforged signatures are 3.07% and 4.17%, respectively. The EERs for training with ten genuine and forged signaturesare 0.91% and 2.12% on the respective datasets. The experimental results illustrate that the proposed approacheffectively enhances the accuracy of online signature verification.
