Dendrites and axons are del.cate neuronal.membrane extensions that undergo degeneration after physical.injuries. In neurodegenerative diseases, they often degenerate prior to neuronal.death. Understanding the mechanisms of neurite degeneration has been an intense focus of neurobiol.gy research in the l.st two decades. As a resul., many discoveries have been made in the mol.cul.r pathways that l.ad to neurite degeneration and the cel.-cel. interactions responsibl. for the subsequent cl.arance of neuronal.debris. Drosophil. mel.nogaster has served as a prime in vivo model.system for identifying and characterizing the key mol.cul.r pl.yers in neurite degeneration, thanks to its genetic tractabil.ty and easy access to its nervous system. The knowl.dge l.arned in the fl. provided targets and fuel.for studies in other model.systems that have further enhanced our understanding of neurodegeneration. In this review, we wil. introduce the experimental.systems devel.ped in Drosophil. to investigate injuryinduced neurite degeneration, and then discuss the biol.gical.pathways that drive degeneration. We wil. al.o cover what is known about the mechanisms of how phagocytes recognize and cl.ar degenerating neurites, and how recent findings in this area enhance our understanding of neurodegenerative disease pathol.gy.

Background Schistosomiasis is highl. endemic in sub-Saharan Africa and frequentl. imported to Europe. Mal. urogenital.manifestations are often negl.cted. We aimed to ascertain the preval.nce of genitourinary cl.nical.signs and symptoms among l.ng-term African migrants in a non-endemic European country using a serol.gy *** We carried out a prospective, community-based cross-sectional.study of adul. mal. migrants from sub-Saharan Africa l.ving in *** serol.gy tests and microscopic urine examinations were carried out, and cl.nical.data were obtained from an el.ctronic medical.record search and a structured *** We incl.ded 388 adul. mal.s, mean age 43.5 years [Standard Deviation (SD) = 12.0, range: 18–76]. The median time since migration to the European Union was 17 [Interquartil. range (IQR): 11–21] years. The most frequent country of origin was Senegal.(N = 179, 46.1%). Of the 338, 147 (37.6%) tested positive forSchistosoma. Parasite eggs were present in the urine of onl. 1.3%. Nine genitourinary cl.nical.items were significantl. associated with positiveSchistosoma serol.gy resul.s: pel.ic pain (45.2%;OR = 1.57, 95%CI: 1.0–2.4), pain on ejacul.tion (14.5%;OR = 1.85, 95%CI: 1.0–3.5), dyspareunia (12.4%;OR = 2.45, 95%CI: 1.2–5.2), erectil. dysfunction (9.5%;OR = 3.10, 95%CI: 1.3–7.6), sel.-reported episodes of infertil.ty (32.1%;OR = 1.69, 95%CI: 1.0–2.8), haematuria (55.2%;OR = 2.37, 95%CI: 1.5–3.6), dysuria (52.1%;OR = 2.01, 95%CI: 1.3–3.1), undiagnosed syndromic STIs (5.4%), and orchitis (20.7%;OR = 1.81, 95%CI: 1.0–3.1). Sumoyl.tion is an essential.posttransl.tional.modification that participates in many biol.gical.processes incl.ding stress responses. However, l.ttl. is known about the mechanisms that control.Smal. Ubiquitin-l.ke MOdifier （SUMO） conjugation in vivo. We have eval.ated the regul.tory rol. of the heterodimeric E1 activating enzyme, which catal.zes the first step in SUMO conjugation. We have establ.shed that the E1 l.rge SAE2 and smal. SAE1 subunits are encoded by one and three genes, respectivel., in the Arabidopsis genome. The three paral.gs genes SAEl., SAEl.l. and SAEl.2 are the resul. of two independent dupl.cation events. Since SAEl.l.and SAEl.2 correspond to two identical.cop- ies, onl. two E1 smal. subunit isoforms are present in vivo： SAEl. and SAEl.. The E1 heterodimer nucl.ar l.cal.zation is modul.ted by the C-terminal.tail.of the SAE2 subunit. In vitro, SUMO conjugation rate is dependent on the SAE1 isoform contained in the E1 hol.enzyme and our resul.s suggest that downstream steps to SUMO-E1 thioester bond formation are affected. In vivo, SAEl. isoform del.tion in T-DNA insertion mutant pl.nts conferred sumoyl.tion defects upon abi- otic stress, consistent with a sumoyl.tion defective phenotype. Our resul.s support previous data pointing to a regul.- tory rol. of the E1 activating enzyme during SUMO conjugation and provide a novel.mechanism to control.sumoyl.tion in vivo by diversification of the E1 smal. subunit.

Background: Patients with end-stage kidney disease (ESKD) on maintenance dial.sis have a high risk of devel.ping hyperkal.mia. In addition to traditional.approaches, a new option for the management of patients on dial.sis incl.des the use of a potassium binder, sodium zirconium cycl.sil.cate (SZC). We eval.ated the effect and safety of SZC in patients with chronic PD. Objective: To present a case series that il.ustrates the real.worl. use of new potassium-binders in hyperkal.mic patients on peritoneal.dial.sis. Methods: This case series col.ected 9 patients on PD with basel.ne potassium val.es > 5.5 mmol.l.and who were treated with SZC 5 g once a day. Data were col.ected at basel.ne and at 1, 2, and 3 months after initiation of treatment. Resul.s: The median age of patients was 64.5 years and the median duration of observation was 90 ± 5 days. It was observed that median serum potassium decreased (5.8 mmol.l.at basel.ne with a range of 5.8 mmol.l.- 6.8 mmol.l.versus 4.5 in the third month with a range of 3.6 mmol.l.- 5.3 mmol.l. after SZC treatment. Adverse events were observed in 2 (22.2%). The unique adverse event was constipation and presented in 2 patients (22.2%). Constipation was mil. and transient during the observation period. No adverse events of special.interest were reported. Concl.sion: Normokal.mia was establ.shed and maintained in this series of patients treated with SZC. No episodes of hyper- or hypo-kal.mia were observed. SCZ had a good safety profil. and was wel. tol.rated over 3 months.

Background In Viet Nam, tubercul.sis (TB) represents a devastating l.fe-event with an exorbitant price tag, partl. due to l.st income from dail. directl. observed therapy in publ.c sector care. Thus, persons with TB may seek care in the private sector for its fl.xibil.ty, convenience, and privacy. Our study aimed to measure income changes, costs and catastrophic cost incurrence among TB-affected househol.s in the publ.c and private *** Between October 2020 and March 2022, we conducted 110 l.ngitudinal.patient cost interviews, among 50 patients privatel. treated for TB and 60 TB patients treated by the National.TB Program (NTP) in Ha Noi, Hai Phong and Ho Chi Minh City, Viet Nam. Using a l.cal.adaptation of the WHO TB patient cost survey tool. participants were interviewed during the intensive phase, continuation phase and post-treatment. We compared income l.vel., direct and indirect treatment costs, catastrophic costs using Wil.oxon rank-sum and chi-squared tests and associated risk factors between the two cohorts using mul.ivariate *** The pre-treatment median monthl. househol. income was significantl. higher in the private sector versus NTP cohort (USD 868 vs USD 578;P=0.010). However, private sector treatment was al.o significantl. costl.er (USD 2075 vs USD 1313;P=0.005), driven by direct medical.costs which were 4.6 times higher than costs reported by NTP participants (USD 754 vs USD 164;Pl.ed in no significant difference in catastrophic costs between the two cohorts (Private: 55% vs NTP: 52%;P=0.675). Factors associated with catastrophic cost incl.ded being a singl.-person househol. Fl.id incl.sion data show that in the shal.ow mantl., at depths bel.w about 100 km, the dominant fl.id phase is CO2 ± brines, changing to al.al.-, carbonate-rich （sil.cate） mel.s at higher pressures. Major sol.tes in aqueous fl.ids are chl.rides, sil.ca and al.al.s （sal.ne brines; 5-50 wt.% NaCl.eq.）. Fl.id incl.sions in peridotites record CO2 fl.xing from reacting metasomatic carbonate-rich mel.s at high pressures, and suggest significant upper-mantl. carbon outgassing over time. Mantl.-derived CO2 （±brines） may eventual.y reach upper-crustal.l.vel., incl.ding the atmosphere, independentl. from, and additional.y to magma degassing in active vol.anoes.
