Two-dimensional （2D） nanomaterials have gained tremendous attention in the field of biomedicine because of their high specific surface areas and fascinating physicochemical properties. Herein, 2D monolayered double hydroxide （MLDH） nanosheets were employed to localize doxorubicin （DOX）, an anticancer drug, with a loading capacity of as high as 3.6 ***-1 （w/w）. Structural characterizations and theoretical calculations indicate that the DOX molecule is uniformly arranged and oriented at the surface of the MLDHs with a binding energy of 15.90 eV, showing significant electrostatic interaction. With the assistance of the targeting agent folic acid （FA）, DOX-FA/MLDHs demonstrate targeted cellular uptake and superior anticancer behavior based on in vitro tests performed with cancer cells. In addition, this composite material exhibits a selective release toward cancer cells and good biocompatibility with normal cells, which would guarantee its practical applications in cancer therapy.

Background: Prediction of complications after pancreatoduodenectomy (PD) remains of interest. Blood parameters and biomarkers during rst and second postoperative days (POD1, POD2) may be early indi- cators of complications. Methods: This case-control study included 50 patients. Baseline, POD1 and POD2 values of leukocytes, neutrophils, lymphocytes, platelets, hemoglobin, C-reactive protein (CRP), procalcitonin and arterial lactate were compared between individuals presenting Clavien ≥ III morbidity, pancreatic stula (PF) or clinically relevant PF (CRPF) and those without these morbidities. Common variables reaching signi cance were further analyzed in order to calculate a predictive score. Results: Severe morbidity, PF and CRPF rates were 28.0%, 26.0% and 14.0%, respectively. Patients with severe morbidity had lower leukocytes on POD2 (P=0.04). Patients with PF presented higher CRP on POD2 (P=0.001), higher lactate on POD1 (P=0.007) and POD2 (P=0.008), and lower lymphocytes on POD1 (P=0.007) and POD2 (P=0.008). Patients with CRPF had lower leukocytes and neutrophils on POD1 (P =0.048, P =0.038), lower lymphocytes on POD1 (P =0.001) and POD2 (P =0.003), and higher CRP on POD2 (P =0.001). Baseline parameters and procalcitonin obtained no statistical associations. Score was de ned according to lymphocytes on POD1 good accuracy. Conclusions: Impaired immunological reaction during early postoperative period (lower leukocytes and, particularly, lymphocytes) in response to surgical aggression would favor complications after PD. Likewise, acidosis (higher arterial lactate) could behave as risk factor of PF. An elevated CRP on POD2 is also an early biomarker of PF. Our novel score based on postoperative lymphocyte count and CRP seems reliable

Dear Editor,Over the last 15 years endocytosis has moved from being a process of only minor importance to plant physiologists to being one of the most exciting research areas in plant cell biology. These days, nobody doubts the operation of clathrin-mediated endocytosis as a mechanism for the internalization of a range of physiologically important transmembrane protein complexes at the plasma membrane （PM） of plant cells. These include both receptors and transporters. As in animal cells, most of these proteins are constitutively recycled back to the PM from an early endosome （EE）. However, some are destined for degradation and proceed further downstream in the endocytic pathway to late endosomes （LE）, where they are internalized into the intraluminal vesicles of the LE. Fusion of the LE with the lysosome/vacuole releases the vesicles leading to their degradation. The signal that marks PM proteins for degradation has been known in mammalian and yeast cells for quite some time and is polyubiquitination （Mukhopadhyay and Riezman, 2007）. In their Spotlight article, Zelazny and Vert （2015） highlight recent publications from the plant field that also demonstrate a key role for multiple monoubiquitination in the endocytosis of the metal transporters IRT1 （Barberon et al., 2011）, BOR1 （Kasai et al., 2011）, as well as lysine63-1inked polyubiquitination in vacuolar sorting of the auxin transporter PIN2 （Leitner et al., 2012）. They also draw attention to the discovery of a ubiquitin-binding protein （FREE1 ; also termed FYVE1 by Barberon et al., 2014） which locates to the LE in plant cells and is part of the ESCRT-I （endosomal sorting complex for transport） complex （Gao et al., 2014）. This complex sequesters ubiquitinated membrane cargo proteins and internalizes them. Finally, Zelazny and Vert draw attention to the work of Ivanov et al. （2014） in showing an increase in IRT1 degradation and iron deficiency in SNX1 mutants. 利用硫磺素T（ ThT）荧光分析法和透射电子显微镜检测β-酪蛋白形成淀粉样纤维沉淀（ Fibril）的动力学过程,研究了磷脂和硫酸肝素对其 Fibril 形成的影响.实验结果表明,β-酪蛋白在65℃下, pH 值为5.4~9.0的范围内,加热252 h以上,并未形成Fibril,说明β-酪蛋白是一种很好的分子伴侣,在高温、弱酸和弱碱条件下均不形成淀粉样纤维沉淀.甘油磷酸胆碱D6PC和D9PC可以显著地促进β-酪蛋白的Fibril的形成,说明一定条件下蛋白质可能与细胞膜之间存在相互作用而导致其二级构象的转变.硫酸肝素对β-酪蛋白形成Fibril具有促进作用,在炎症组织中,硫酸肝素表达量的增加有可能促进β-酪蛋白形成Fibril,说明乳腺炎与乳腺中的Corpora Amylacea的形成存在一定的联系.

Risk factors for colonoscopic perforation: A population-based study of 80118 cases

AIM: To assess the incidence and risk factors associated with colonic perforation due to colonoscopy. METHODS: This was a retrospective cross-sectional study. Patients were retrospectively eligible for inclusion if they were 18 years and older and had an inpatient or outpatient colonoscopy procedure code in any facility within the Geisinger Health System during the period from January 1, 2002 to August 25, 2010. Data are presented as median and inter-quartile range, for continuous variables, and as frequency and percentage for categorical variables. Baseline comparisons across those with and without a perforation were made using the two-sample t -test and Pearson's χ2 test, as ***: A total of 50 perforations were diagnosed out of 80118 colonoscopies, which corresponded to an incidence of 0.06% (95%CI: 0.05-0.08) or a rate of 6.2 per 10000 colonoscopies. All possible risk factors associated with colonic perforation with a P -value < 0.1 were checked for inclusion in a multivariable logbinomial regression model predicting 7-d colonic perforation. The final model resulted in the following risk factors which were significantly associated with risk of colonic perforation: age, gender, body mass index, albumin level, intensive care unit (ICU) patients, inpatient setting, and abdominal pain and Crohn's disease as indications for colonoscopy. CONCLUSION: The cumulative 7 d incidence of colonic perforation in this cohort was 0.06%. Advanced age and female gender were significantly more likely to have perforation. Increasing albumin and BMI resulted in decreased risk of colonic perforation. Having a colonoscopy indication of abdominal pain or Crohn's disease resulted in a higher risk of colonic perforation. Colonoscopies performed in inpatients and particularly the ICU setting had substantially greater odds of perforation. Biopsy and polypectomy did not increase the risk of perforation and only three perforations occurred with screening colonoscopy.

In order to obtain better anti-cancer compounds, nine glycosylated derivatives were designed and synthe- sized using diosgenin as starting material. Their structures were confn-med by 1H NMR, 13C NMR and MS spectra. The anti-cancer activities of intermediate compounds 5a-5i and the target compounds 6a-6i were investigated against human leukemia HEL, K562, HL60 and melanoma WM9 cell lines via MTT method. The bioassay results show that these derivatives possess good inhibitory activities against the four cancer cell lines. Furthermore, these derivatives show better inhibitory activities against K562 than against other cell lines, and most derivatives show better inhibitory activities than the parental material. Moreover, compounds 6a--6i are more active than their inter- mediates 5a--5i when against these ceils. The above results demonstrate the effects of glycosylation on 3-OH of di- osgenin and acetylation of the sugar moiety on their antitumor activities.
