A common mechanism in plant hormone signaling is hormonedependent degradation of downstream *** the case of strigolactone(SL)signaling,one group of SUPPReSSOR OF MORe AXILLARY BRANCHeS 2(MAX2)1-LIKe(SMXL)proteins undergoes hormone-dependent degradation,while another does not.A recent publication in Molecular Plant by Li et al.(2024)illustrates that mixing and matching a SMXL resistant to SL degradation with a SMXL sensitive to SL degradation finely tunes SL *** phenomenon could reflect a naturally occurring negative transdominance,which is a dominantnegative effect arising not from a mutant allele of the same gene but rather from a different gene altogether(Veitia,2007).This trans-acting protein attenuates SL-dependent degradation of the SMXL *** findings have exciting implications for how different SMXL combinations could result in a gradient of SL responses.

Next-generation sequencing(NGS),represented by Illumina platforms,has been an essential cornerstone of basic and applied ***,the sequencing error rate of 1 per 1000 bp(10^(−3))represents a serious hurdle for research areas focusing on rare mutations,such as somatic mosaicism or microbe *** examining the high-fidelity sequencing methods developed in the past decade,we summarized three major factors underlying errors and the corresponding 12 strategies mitigating these *** then proposed a novel framework to classify 11 preexisting representative methods according to the corresponding combinatory strategies and identified three trends that emerged during methodological *** further extended this analysis to eight long-read sequencing methods,emphasizing error reduction ***,we suggest two promising future directions that could achieve comparable or even higher accuracy with lower costs in both NGS and long-read sequencing.

Conventional numerical solutions developed to describe the geomechanical behavior of rock interfaces subjected to differential load emphasize peak and residual shear *** detailed analysis of preand post-peak shear stress-displacement behavior is central to various time-dependent and dynamic rock mechanic problems such as rockbursts and structural instabilities in highly stressed *** complete stress-displacement surface(CSdS)model was developed to describe analytically the pre-and post-peak behavior of rock interfaces under differential *** formulations of the CSdS model required extensive curve-fitting iterations which limited its practical applicability and transparent integration into engineering *** present work proposes modifications to the CSdS model aimed at developing a comprehensive and modern calibration protocol to describe the complete shear stressdisplacement behavior of rock interfaces under differential *** proposed update to the CSdS model incorporates the concept of mobilized shear strength to enhance the post-peak ***’s concepts of joint roughness coefficient(JRC)and joint compressive strength(JCS)are incorporated to facilitate empirical estimations for peak shear stress and normal closure ***/uniaxial compression test and direct shear test results are used to validate the updated model and exemplify the proposed calibration *** results illustrate that the revised model successfully predicts the post-peak and complete axial stressestrain and shear stressedisplacement curves for rock joints.

The joint european Space Agency and Chinese Academy of Sciences Solar wind Magnetosphere Ionosphere Link explorer(SMILe)mission will explore global dynamics of the magnetosphere under varying solar wind and interplanetary magnetic field conditions,and simultaneously monitor the auroral response of the Northern Hemisphere *** these large-scale responses with medium and fine-scale measurements at a variety of cadences by additional ground-based and space-based instruments will enable a much greater scientific impact beyond the original goals of the SMILe ***,we describe current community efforts to prepare for SMILe,and the benefits and context various experiments that have explicitly expressed support for SMILe can offer.A dedicated group of international scientists representing many different experiment types and geographical locations,the Ground-based and Additional Science Working Group,is facilitating these *** include constructing an online SMILe data Fusion Facility,the discussion of particular or special modes for experiments such as coherent and incoherent scatter radar,and the consideration of particular observing strategies and spacecraft *** anticipate growing interest and community engagement with the SMILe mission,and we welcome novel ideas and insights from the solar-terrestrial community.

The long-period-stacking-ordered(LPSO)structure affects the mechanical,corrosion and hydrolysis properties of Mg *** current work employs high angle annular dark field-scanning transmission electron microscopy(HAAdF-STeM)and atom probe tomography(APT)to investigate the structural and local chemical information of LPSO phases formed in Mg-Ni-Y/Sm ternary alloys after extended isothermal *** on the alloying elements and their concentrations,Mg-Ni-Y/Sm develops a two-phase LPSO+α-Mg structure in which the LPSO phase contains defects,hybrid LPSO structure,and Mg ***-STeM and APT indicate non-stoichiometric LPSO with incomplete Ni_(6)(Y/Sm)_(8) *** addition,the APT quantitatively determines the local composition of LPSO and confirms the presence of Ni within the Mg bonding *** results provide insight into a better understanding of the structure and hydrolysis properties of LPSO-Mg alloys.

Connecting cellular mechanisms and extracellular vesicle cargo in traumatic brain injury

Traumatic brain injury is followed by a cascade of dynamic and complex events occurring at the cellular level. These events include: diffuse axonal injury, neuronal cell death, blood-brain barrier break down, glial activation and neuroinflammation, edema, ischemia, vascular injury, energy failure, and peripheral immune cell infiltration. The timing of these events post injury has been linked to injury severity and functional outcome. extracellular vesicles are membrane bound secretory vesicles that contain markers and cargo pertaining to their cell of origin and can cross the blood-brain barrier. These qualities make extracellular vesicles intriguing candidates for a liquid biopsy into the pathophysiologic changes occurring at the cellular level post traumatic brain injury. Herein, we review the most commonly reported cargo changes in extracellular vesicles from clinical traumatic brain injury samples. We then use knowledge from animal and in vitro models to help infer what these changes may indicate regrading cellular responses post traumatic brain injury. Future research should prioritize labeling extracellular vesicles with markers for distinct cell types across a range of timepoints post traumatic brain injury.

Plant virus causes massive crop losses ***,there is currently no effective measure to control plant viral ***,we identify an antiviral protein Rhp-PSP,produced by the bacterial Rhodopseudomonas palustris strain *** this study,we discover that the antiviral activity of Rhp-PSP relies on its endoribonuclease *** the arginine(R)residue at position 129 onto alanine(A)abolishs its endoribonuclease activity on coat protein(CP)RNA of tobacco mosaic virus(TMV),consequentially,compromises the antiviral activity of *** investigation demonstrates that,the mutant Rhp-PSP^(R129A)is unable to form the homotrimer as the wild type,indicating the importance of quaternary junction for the endoribonuclease *** of Rhp-PSP in Nicotiana benthamiana significantly enhances the resistance against TMV of seedlings,while expression of Rhp-PSP^(R129A)did not,confirming that endoribonuclease activity is responsible for the antiviral activity of *** addition,foliar spray of Rhp-PSP solution on tomato and pepper plants significantly reduces the disease index of viral diseases,indicating that Rhp-PSP shows potential to develop antiviral agent in practice.
