Fecal marker levels as predictors of need for endoscopic balloon dilation in Crohn's disease patients with anastomotic strictures

AIM To evaluate the accuracy and best cut-off value of fecal calprotectin(FC) and fecal lactoferrin(FL) to predict disease recurrence in asymptomatic patients presenting with anastomotic strictures. METHOdS This was a longitudinal single tertiary center study based on prospectively collected data(recorded in a clinical database created for this purpose) performed between March 2010 and November 2014. Crohn's disease(Cd) patients with anastomotic stricture who submitted to postoperative endoscopic evaluation were included. Stools were collected on the day before bowel cleaning for FC and FL. Endoscopic balloon dilation(EBd) was performed if the patient presented an anastomotic stricture not traversed by the colonoscope, regardless of patients' symptoms. Successful dilation was defined as passage of the colonoscope through the dilated stricture into the neotermimal *** recurrence was defined as a modified Rutgeerts score of ≥ i2 b. RESULTS In a total of 178 patients who underwent colonoscopy, 58 presented an anastomotic stricture, 86% were asymptomatic, and 48(54% male; median age of 46.5 years) were successfully dilated. Immediate success rate was 92% and no complications were recorded. FC and FL levels correlated significantly with endoscopic recurrence(P dictive value(PPV) of 72.4%, negative predictive value(NPV) of 94.7% and accuracy of 81%] for FC and of 5.6 μg/g(sensitivity of 77.3%, specificity of 69.2%, PPV of 68%, NPV of 78.4% and accuracy of 72.9%) for *** Fecal markers are good predictors of Cd endoscopic recurrence in patients with asymptomatic anastomotic stricture. FC and FL may guide the need for EBd in this context.

Spectral reflectance indices as proxies for yield potential and heat stress tolerance in spring wheat: heritability estimates and marker-trait associations

The application of spectral reflectance indices(SRIs) as proxies to screen for yield potential(YP) and heat stress(HS) is emerging in crop breeding ***, a comparison of SRIs and their associations with grain yield(GY) under YP and HS conditions is *** this study, we assessed the usefulness of 27 SRIs for indirect selection for agronomic traits by evaluating an elite spring wheat association mapping initiative(WAMI) population comprising 287 elite lines under YP and HS *** and phenotypic analysis identified 11 and 9 SRIs in different developmental stages as efficient indirect selection indices for yield in YP and HS conditions,*** identified enhanced vegetation index(EVI) as the common SRI associated with GY under YP at booting, heading and late heading stages, whereas photochemical reflectance index(PRI) and normalized difference vegetation index(NdVI) were the common SRIs under booting and heading stages in *** association study(GWAS) using 18704 single nucleotide polymorphisms(SNPs) from Illumina i Select90 K identified 280 and 43 marker-trait associations for efficient SRIs at different developmental stages under YP and HS, *** genomic regions for multiple SRIs were identified in 14 regions in 9 chromosomes: 1 B(60–62 cM), 3 A(15, 85–90, 101–105 cM), 3 B(132–134 cM), 4 A(47–51 cM), 4 B(71–75 cM), 5 A(43–49, 56–60, 89–93 cM), 5 B(124–125 cM),6 A(80–85 cM), and 6 B(57–59, 71 cM).Among them,SNPs in chromosome 5 A(89–93 cM) and 6 A(80–85 cM)were co-located for yield and yield related ***,this study highlights the utility of SRIs as proxies for GY under YP and *** heritability estimates and identification of marker-trait associations indicate that SRIs are useful tools for understanding the genetic basis of agronomic and physiological traits.

Mg-Zn-Zr(ZK)alloys exhibit notably high mechanical strength amongst all magnesium alloy ***,due to the formation of low melting point Mg_3Zn_7-precipitates,these alloys are susceptible to hot cracking,thus compromising their metallurgical *** addition of rare earths to ZK alloys is an alternative to form higher melting point intermetallic compounds,speed up dynamic recrystallization,refine grain size,enhance corrosion resistance and extend the service temperature due to improved creep *** work deals with the effect of Ce-base mischmetal addition on the hot rolling behaviour of as-cast ZK60 *** microstructure investigation conducted using electron microscopy and X-Ray diffraction shows that precipitation of Mg_(7)Zn_(3) intermetallics occur during hot rolling,whereas no further precipitation is observed for the ZK60-Mm *** fragmentation of the intermetallic compounds occur during hot rolling and finer particles of Mg_(7)Zn_(3) are observed for the ZK60,whereas Mg_(7)Zn_(3) and Mg Zn_(2) Ce intermetallics are formed in the alloy modified with mischmetal addition.A higher fraction of dynamically recrystallized grains is observed for the ZK60-Mm in comparison to the *** recrystallization takes place in ZK60 with the formation of sub-grains near to the intermetallics and the addition of mischmetal promotes the occurrence of discontinuous recrystallization with the nucleation of new grains close to the *** mechanical strength and,in particular,the ductility of the hot-rolled alloys are notably improved when compared to the same alloys in the as-cast *** mechanical strength is,however,higher for the ZK60 *** solid solution strengthening,softer Mg Zn_(2) Ce intermetallics and more extensive recrystallization contribute to reduce the mechanical strength of *** in both alloys are initiated at coarse intermetallics and propagate through intermetallic-rich regions.

TGF-beta receptor mediated telomerase inhibition, telomere shortening and breast cancer cell senescence

Human telomerase reverse transcriptase （hTERT） plays a central role in telomere lengthening for continuous cell proliferation, but it remains unclear how extracellular cues regulate telomerase lengthening of telomeres. Here we report that the cytokine bone morphogenetic protein-7 （BMP7） induces the hTERT gene repression in a BMPRII receptor- and Smad3-dependent manner in human breast cancer cells. Chonic exposure of human breast cancer cells to BMP7 results in short telomeres, cell senescence and apoptosis. Mutation of the BMPRII receptor, but not TGFbRII, ACTRIIA or ACTRIIB receptor, inhibits BMP7-induced repression of the hTERT gene promoter activity, leading to increased telomerase activity, lengthened telomeres and continued cell proliferation. Expression of hTERT prevents BMP7-induced breast cancer cell senescence and apoptosis. Thus, our data suggest that BMP7 induces breast cancer cell aging by a mechanism involving BMPRII receptor- and Smad3-mediated repression of the hTERT gene.

Textile-based technologies are considered as potential routes for the production of 3d porous architectures for tissue engineering( TE) applications. We describe the use of two polymers,namely polybutylene succinate( PBS) and silk fibroin(SF) to produce fiber-based finely tuned porous architectures by weft and warp knittings. The obtained knitted constructs are described in terms of their morphology, mechanical properties,swelling ability,degradation behaviour,and cytotoxicity. Each type of polymer fibers allows for the processing of a very reproducible intra-architectural scaffold geometry,with distinct characteristics in terms of the surface physicochemistry,mechanical performance,and degradation capability,which has an impact on the resulting cell behaviour at the surface of the respective biotextiles. Preliminary cytotoxicity screening shows that both materials can support cell adhesion and proliferation. Furthermore, different surface modifications were performed( acid /alkaline treatment, UV radiation,and plasma) for modulating cell behavior. An increase of cell-material interactions were observed,indicating the important role of materials surface in the first hours of culturing. Human adipose-derived stem cells( hASCs) became an emerging possibility for regenerative medicine and tissue replacement therapies. The potential of the recently developed silk-based biotextile structures to promote hASCs adhesion,proliferation,and differentiation is also evaluated. The obtained results validate the developed constructs as viable matrices for TE applications. Given the processing efficacy and versatility of the knitting technology, and the interesting structural and surface properties of the proposed polymer fibers,it is foreseen that our developed systems can be attractive for the functional engineering of tissues such as bone,skin,ligaments or cartilage and also for develop more complex systems for further industrialization of TE products.

Quantification of theophylline or paracetamol in milk matrices by high-performance liquid chromatography

A simple, accurate and sensitive high-performance liquid chromatography （HPLC） method was developed, validated and applied to the determination of either theophylline or paracetamol in milk-based samples. The method allowed drug quantification in fresh and powdered milk with a relatively short run time of analysis and it was also successfully applied to the quantification of the drugs in solid dosage forms intended for pediatric use. Moreover, the main significant advantages over other published works are the simplicity of the sample preparation, reduced assay time and sample loss. The method meets the International Conference on Harmonization guideline for analytical methods validation regarding specificity, linearity, accuracy, precision, specificity and robustness as required by health authorities and applied by industry while designing and marketing new drug products. The technique encompasses the separation of the analytes with a reverse phase C18 column under isocrafic conditions and UV detection at 272 nm and 243 nm, respectively, for theophylline and paraeetamol. The lower limit of quantification for both drugs was determined as 0.2 μg/mL and the between-batch accuracy was 99.7%. This HPLC method allows quantification of theophylline and paracetamol in milk matrices and it can be applied in the design, development and production of milk-based pediatric dosage forms.

The complexity of sediment dynamics in aquatic systems can be better understood by applying numerical models. The development of a comprehensive morphological model is presented in this *** model aims to predict the sediment transport and bed evolution in natural systems composed of different sediment types. The morphological model was implemented in MOHId, a modelling system that solves the three-dimensional hydrodynamics and advection-diffusion transport of suspended sediments. Multiple sediment classes were taken into account(non-cohesive and cohesive) considering the effects of sediment mixtures and bed consolidation on resistance to erosion. To represent bottom stratigraphy, the bottom column can be divided into several layers. The key points of the simulated processes are discussed in this paper. Model results are assessed in six test cases through comparison with analytic solutions or experimental data. The outcomes demonstrate the model's capacity to simulate the transport dynamics of non-cohesive and cohesive sediments. The speed up of morphological changes by an acceleration factor permitted modelling bed evolution for long time ***, a test case for the Tagus Estuary demonstrated the model's capacity for generating realistic sediment distribution based on the local hydrodynamic conditions. Limitations in the availability of bed composition data can be overcome by considering a warm-up run to provide realistic initial conditions for further predictions of morphological developments.
