Antioxidants Enzymes Activities and nO Levels in Hemodialysis and Continuous Ambulatory Peritoneal Dialysis—A Comparative Study

It consists of a retrospective study with twenty-two individuals diagnosed with DRC: fourteen HD and eight CAPD, followed up in the dialysis center of Antonio Pedro University Hospital (HUAP) of fluminense federal University (Uff), between 2015 and 2017. fifteen healthy control (HC) subjects were included, without diagnosed CKD. Patients with HIV positive, hepatitis A, B, and C, pregnant, cancer, smokers, alcoholics and those exposed to X-rays in the last 3 months, were excluded. Objectives: As oxidative stress and endothelial dysfunction may be linked to the higher prevalence of CVD in CKD patients, we measured the activities of the antioxidants enzymes: SOD and GPx and total nO levels in the plasma and serum of end-stage CKD patients undergoing dialysis therapy, comparing with the HC group. Methods: Quantification of nO levels was performed by fluorometric kit, while activities of SOD and GPx were determined using kinetic methods. Results: We found higher plasma SOD activity in HD (8.58 U/ml) and CAPD (10.14 U/ml), compared to C (3.73 U/ml) group, while GPx activity was decreased in HD (115.38 nmol/h/ml) and CAPD (122.76 nmol/h/ml) groups compared to HC group (275.83 nmol/h/ml). Total serum nO concentration was decreased in HD (14.09 pmol/μl) and CAPD (10.26 pmol/μl), compared to non-CKD patients (49.65 pmol/μl). Conclusion: Decreased total serum nO and GPx activities may lead to endothelial dysfunction and consequently a higher prevalence of CVD in CKD patients.

AIM： To study the association between Interleukin-1 （IL-1） and tumor necrosis factor （Tnf）-α polymorphisms, infection by Helicobacter pylori （H pylori） and the development of gastrointestinal diseases.METHODS： Genomic DnA was extracted from the peripheral blood of 177 patients with various gastrointestinal diseases and from 100 healthy volunteers. The polymorphisms in IL-1β and Tnf-α genes were analyzed using the polymerase chain reactionrestriction fragment length polymorphism method （PCRRfLP） and those from IL-1Rn with PCR. The presence of infection due to H pylori and the presence of the CagA toxin were detected by serology. The histopathological parameters in the gastric biopsies of the patients were according to the Sydney classification.RESULTS： A comparison of the frequencies of the different polymorphisms studied among the patients and the control group demonstrated that the allele IL- 1Rn＊2 was more frequent among patients with gastric ulcers and adenocarcinoma. Carriers of the allele IL- Rn＊2 and those with reactive serology for anti-CagA IgG had a greater risk of developing peptic ulcer and gastric adenocarcinoma, as well as a higher degree of inflammation and neutrophilic activity in the gastricCOnCLUSIOn： Our results indicate a positive association between IL-1Rn gene polymorphism and infection by positive H pylori CagA strains and the development of gastric ulcers and adenocarcinoma.

Pancreatic ductal adenocarcinoma(PDAC)has one of the highest mortality rates among all cancer *** delayed diagnosis precludes curative resection,thus most of the current therapies against PDAC are based on chemo-and ***,these strategies are insufficient to improve its poor *** the advances made in chemotherapy(***-paclitaxel and gemcitabine),many patients with PDAC are unable to benefit from them due to the rapid development of drug ***,more than 165 genes have been found to be implicated in drug resistance of pancreatic tumors,including different integrins,mucins,nf-κB,RAS and ***,drug resistance in PDAC is thought to be mediated by the modulation of miRnAs(***-21,miRnA-145 and miRnA-155),which regulate genes that participate in cell proliferation,invasion and ***,cancer stem cells are intimately related to drug resistance in PDAC due to their ability to overexpress ABC genes-involved in drug transport-,and enzymes such as aldehyde dehydrogenases-implicated in cellular drug metabolism-and poly(ADP-ribose)polymerases-involved in drug-induced DnA damage *** the mechanisms involved in drug resistance will contribute to the development of efficient therapeutic strategies and to improve the prognosis of patients with PDAC.

Allogeneic hematopoietic stem cell transplantation(alloHSCT)is a life-saving procedure used for the treatment of selected hematological malignancies,inborn errors of metabolism,and bone marrow *** role of neutrophils in alloHSCT has been traditionally evaluated only in the context of their ability to act as a first line of defense against ***,recent evidence has highlighted neutrophils as key effectors of innate and adaptive immune responses through a wide array of newly discovered ***,neutrophils are emerging as highly versatile cells that are able to acquire different,often opposite,functional capacities depending on the microenvironment and their differentiation ***,we review the current knowledge on the multiple functions that neutrophils exhibit through the different stages of alloHSCT,from the hematopoietic stem cell(HSC)mobilization in the donor to the immunological reconstitution that occurs in the recipient following HSC *** also discuss the influence exerted on neutrophils by the immunosuppressive drugs delivered in the course of alloHSCT as part of graft-versushost disease(GVHD)***,the potential involvement of neutrophils in alloHSCT-related complications,such as transplant-associated thrombotic microangiopathy(TA-TMA),acute and chronic GVHD,and cytomegalovirus(CMV)reactivation,is also *** on the data reviewed herein,the role played by neutrophils in alloHSCT is far greater than a simple antimicrobial ***,much remains to be investigated in terms of the potential functions that neutrophils might exert during a highly complex procedure such as alloHSCT.

Dual role of fGf in proliferation and endoreplication of Drosophila tracheal adult progenitor cells

Adult progenitor cells activation is a key event in the formation of adult *** Drosophilat formation of abdominal adult trachea depends on the specific activation of tracheal adult progenitors(tracheoblasts)at the Tr4 and Tr5 spiracular *** of these tracheoblasts generates a pool of tracheal cells that migrate toward the posterior part of the trachea by the activation of the branchless/fibroblast growth factor(Bnl/fGf)signaling to form the abdominal adult ***,we show that,in addition to migration,Bnl/fGf signaling,mediated by the transcription factor Pointed,is also required for tracheoblast *** tracheoblast activation relies on the expression of the fGf ligand bnl in their nearby ***,we show that,in the absence of the transcription factor Cut(Ct),Bnl/fGf signaling induces endoreplication of tracheoblasts partially by promoting fizzy-related ***,our results suggest a dual role of Bnl/fGf signaling in tracheoblasts,inducing both proliferation and endoreplication,depending on the presence or absence of the transcription factor Ct,respectively.
