Using multimodal MRI to investigate alterations in brain structure and function in the BBZDR/Wor rat model of type 2 diabetes

Background:This is an exploratory study using multimodal magnetic resonance imaging(MRI)to interrogate the brain of rats with type 2 diabetes(T2DM)as compared to *** was hypothesized there would be changes in brain structure and function that reflected the human disorder,thus providing a model system by which to follow disease progression with noninvasive ***:The transgenic BBZDR/Wor rat,an animal model of T2MD,and agematched controls were studied for changes in brain structure using voxel-based morphometry,alteration in white and gray matter microarchitecture using diffusion weighted imaging with indices of anisotropy,and functional coupling using restingstate BOLD functional *** from each modality were registered to,and analyzed,using a 3D MRI rat atlas providing site-specific data on over 168 different brain ***:There was an overall reduction in brain volume focused primarily on the somatosensory cortex,cerebellum,and white matter *** putative changes in white and gray matter microarchitecture were pervasive affecting much of the brain and not localized to any *** was a general increase in connectivity in T2DM rats as compared to *** cerebellum presented with strong functional coupling to pons and brainstem in T2DM rats but negative connectivity to ***:The neuroradiological measures collected in BBBKZ/Wor rats using multimodal imaging methods did not reflect those reported for T2DB patients in the *** data would suggest the BBBKZ/Wor rat is not an appropriate imaging model for T2DM.

Development of soft kernel durum wheat

Kernel texture(grain hardness) is a fundamental and determining factor related to wheat(Triticum spp.) milling, baking and flour *** are three kernel texture classes in wheat: soft and hard hexaploid(***), and very hard durum(*** ***).The genetic basis for these three classes lies with the Puroindoline ***, the easiest means of quantifying kernel texture is with the Single Kernel Characterization System(SKCS), although other means are valid and can provide fundamental material *** SKCS values for soft wheat would be around 25 and for durum wheat≥*** kernel durum wheat was created via homeologous recombination using the ph1b mutation, which facilitated the transfer of ca.28 Mbp of5 DS that replaced ca.21 Mbp of 5 *** 5 DS translocation contained a complete and intact Hardness locus and both Puroindoline *** of the Puroindoline genes in durum grain resulted in kernel texture and flour milling characteristics nearly identical to that of soft wheat, with high yields of break and straightgrade flours, which had small particle size and low starch *** water absorption was markedly reduced compared to durum flour and *** strength was essentially unchanged and reflected the inherent gluten properties of the durum *** quality was essentially equal-to-or-better than pasta made from ***, soft durum germplasm showed good potential with moderate grain yield and resistance to a number of fungal pathogens and *** breeding efforts will no doubt further improve the quality and competitiveness of soft durum cultivars.

Long-term fertilization and crop rotation can influence both organic C sequestration as well as the C composition of soils and the more resistant organic C compounds contained in humic acid(HA). This study examined the effects of fertilization and cropping type(monoculture corn(MC) and Kentucky bluegrass sod(KBS) and corn-oat-alfalfa-alfalfa rotation(RC)) on the HA composition of soil from a 52-year field study in southern Ontario, Canada. Humic acid samples were extracted from soil, and elemental analysis, infrared spectroscopy, solid state 13C nuclear magnetic resonance spectra, and electron paramagnetic resonance methods were used to determine the influence of the cropping type on the characteristics of HA. Both fertilization and cropping type affected the chemical characteristics of HA. fertilization led to a 5.9% increase in C, a 7.6% decrease in O, and lower O/C and(N + O)/C ratios in HA as compared to the corresponding non-fertilized treatments. Rotation resulted in a lower proportion of C(48.1%) and a greater(N + O)/C ratio(0.7) relative to monoculture cropping. Infrared spectroscopy analysis showed that HA contained more C-O groups in fertilized soil than in non-fertilized soil under MC and KBS. fertilization increased the O-alkyl-C, phenolic-C, and free radical contents of HA relative to non-fertilization treatments. Rotation decreased the aliphatic and carboxyl groups and increased the O-alkyl, carbohydrate, aryl, and phenolic groups and free radicals, relative to MC and KBS. Both long-term crop rotation and fertilization dramatically modified the soil HA composition. Significant relationships were observed between the molecular composition of HA and soil organic C. Hence, humic acid characterization could be used as an indicator of the long-term sustainability of crop management practices.

Single-lens dynamic z-scanning for simultaneous in situ position detection and laser processing focus control

Existing auto-focusing methods in laser processing typically include two independent modules,one for surface detection and another for z-axis *** latter is mostly implemented by mechanical z stage motion,which is up to three orders of magnitude slower than the lateral processing *** alleviate this processing bottleneck,we developed a single-lens approach,using only one high-speed z-scanning optical element,to accomplish both in situ surface detection and focus control quasi-simultaneously in a dual-beam *** probing beam scans the surface along the z-axis continuously,and its reflection is detected by a set of confocal *** on the temporal response of the detected signal,we have developed and experimentally demonstrated a dynamic surface detection method at 140-350 kHz,with a controlled detection range,high repeatability,and minimum linearity error of 1.10%.Sequentially,by synchronizing at a corresponding oscillation phase of the z-scanning lens,the fabrication beam is directed to the probed z position for precise focus ***,our approach provides instantaneous surface tracking by collecting position information and executing focal control both at 140-350 kHz,which significantly accelerates the axial alignment process and offers great potential for enhancing the speed of advanced manufacturing processes in three-dimensional space.

As the sole obligate symbiotic birds in Africa,oxpeckers offer a unique model for studying symbiotic *** to the multitrophic level they occupy and the context dependent foraging behavior they exhibit,the type of symbiotic relationship can be *** addition to providing a cleaning service to the host by removing ticks,oxpeckers frequently feed on blood,mucus,and saliva,inflicting potential damage on the ***,we used DNA metabarcoding on faecal samples to analyze the taxonomic composition of the trophic interactions of the Yellow-billed Oxpecker(Buphagus africanus)and Red-billed Oxpecker(***)in northeastern *** contrast to conventional methods,DNA metabarcoding allows for a detailed identification of dietary resources encompassing both mammal hosts and consumed arthropods within the same *** this information,we examined differences in the diet composition between oxpecker species and localities,as well as the co-occurrence between host and arthropod *** findings revealed that oxpeckers predominantly source their diet from mammals,ticks,and flies;however,ticks and flies rarely co-occur in the diet of an *** observed variability among individuals in their feeding ecology,which is strongly correlated with locality and,to a lesser extent,with the mammal *** noted a high degree of mobility between hosts within relatively short periods,with 32%of the samples showing traces of at least two mammal *** study illustrates the dynamic foraging behavior of these specialized symbiotic birds,shedding light on their potential role in pest control services and disease transmission.

Detection and quantitation of host cell proteins in monoclonal antibody drug products using automated sample preparation and data-independent acquisition LC-MS/MS

Desmopression is an effective adjunct treatment for reversing excessive hyponatremia overcorrection

We report a case of a 50-year-old malnourished African American male with hiccups, nausea and vomiting who was brought to the Emergency Department after repeated seizures at home. Laboratory evaluations revealed sodium(Na+) 107 mmol/L, unmeasurably low potassium, chloride f 38 mmol/L and serum osmolality 217 m Osm/kg. Seizures were controlled with 3% saline Ⅳ. Once nausea was controlled with iv antiemetics, he developed large volume free water diuresis with 6 L of dilute urine in 8 h(urine osmolality 40-60 m Osm/kg) and serum sodium rapidly rose to 126 mmol/L in 12 h. Both intravenous desmopressin and 5% dextrose in water was given to achieve a concentrated urine and to temporarily reverse theacute rise of sodium, respectively. Serum Na+ was gradually re-corrected in 2-3 mmol/L daily increments from 118 mmol/L until 130 mmol/L. Hypokalemia was slowly corrected with resultant auto-correction of metabolic alkalosis. The patient discharged home with no neurologic sequaele on the 11 th hospital day. In euvolemic hyponatremic patient, controlling nausea may contribute to unpredictable free water diuresis. The addition of an antidiuretic hormone analog, such as desmopressin can limit urine output and prevent an unpredictable rise of the serum sodium.

Using a procedure based on interpolation via continued fractions supplemented by statistical sampling,we analyze proton magnetic form factor data obtained via electron+proton scattering on Q^(2)∈[0.027,0.55]GeV^(2)with the goal of determining the proton magnetic *** approach avoids assumptions about the function form used for data interpolation and ensuing extrapolation onto Q^(2)■0 for extraction of the form factor *** this way,we find r_(M)=0.817(27)*** the difference between proton electric and magnetic radii calculated in this way,extant data are seen to be compatible with the possibility that the slopes of the proton Dirac and Pauli form factors,f1,2(Q^(2)),are not truly independent observables;to wit,the difference f_(1)’(0)-f_(2)’(0)/κp=[1+Kp]/[4 m_(p)^(2)],viz.,the proton foldy term.
