Impact of source tissue and ex vivo expansion on the characterization of goat mesenchymal stem cells

Background： There is considerable interest in using goats as models for genetically engineering dairy animals and also for using stem cells as therapeutics for bone and cartilage repair. Mesenchymal stem cells （MSCs） have been isolated and characterized from various species, but are poorly characterized in goats. Results： Goat MSCs isolated from bone marrow （BM-MSCs） and adipose tissue （ASCs） have the ability to undergo osteogenic, adipogenic and chondrogenic differentiation. Cytochemical staining and gene expression analysis show that ASCs have a greater capacity for adipogenic differentiation compared to BM-MSCs and fibroblasts. Different methods of inducing adipogenesis also affect the extent and profile of adipogenic differentiation in MSCs. Goat fibroblasts were not capable of osteogenesis, hence distinguishing them from the MSCs. Goat MSCs and fibroblasts express CD90, CD105, CD73 but not CD45, and exhibit cytoplasmic localization of OCT4 protein. Goat MSCs can be stably transfected by Nucleofection, but, as evidenced by colony-forming efficiency （CFE）, yield significantly different levels of progenitor cells that are robust enough to proliferate into colonies of integrants following G418 selection. BM-MSCs expanded over increasing passages in vitro maintained karyotypic stability up to 20 passages in culture, exhibited an increase in adipogenic differentiation and CFE, but showed altered morphology and amenability to genetic modification by selection. Conclusions： Our findings provide characterization information on goat MSCs, and show that there can be significant differences between MSCs isolated from different tissues and from within the same tissue. Fibroblasts do not exhibit trilineage differentiation potential at the same capacity as MSCs, making it a more reliable method for distinguishing MSCs from fibroblasts, compared to cell surface marker expression.

Purpose: To report sclerochoroidal calcification in a patient with classic Bartter’ s syndrome. Design: Observational case report. Methods: A 42- year- old woman with a 26- year history of classic Bartter’ s syndrome was found to have bilateral fundus tumors. The patient presented initially with quivering lips and hand stiffness at age 6 years but was not diagnosed until age 16 years. Treatment included magnesium and potassium supplementation and Amiloride therapy. Results: On ocular examination, there were multifocal, yellow- white, geographic, solid choroidal lesions along the superior and inferior retinal vascular arcades in both eyes. Ultrasonography showed echogenic, placoid calcified lesions at the level of the sclera and choroid, consistent with bilateral sclerochoroidal calcification. Conclusions: Sclerochoroidal calcification can be associated with classic Bartter’ s syndrome.

Permanent magnet rings are presented, which exploit the image effect in the surrounding circular iron shields. The theory is given for a general permanent ring when the magnetization orientation at each coordinate angle ψ changes by =(n+1)ψ, where n is a positive or negative integer. For the uniformly magnetized case n=-1, the permanent ring produces no field in its bore, and the field is that of a dipole outside. When the ring is surrounded by a soft iron shield, its field becomes uniform in the bore, and zero outside the ring. The field can be varied continuously by moving the iron shield along the magnet axis. A small variable field device was constructed by using NdFeB permanent rings, which produced a field flux density of 0-0.5 T in the central region.

In contrast to the cries of human infants, sounds made by non-human infants in different stressful behavioral contexts （hunger or physical discomfort, isolation, capture by humans or predators） are usually treated as distinct types of vocalizations. However, if distress vocalizations produced by different species and in different contexts share a common motivational state and associated neurochemical pathways, we can expect them to share a common acoustic structure and adaptive function, showing only limited variation that corresponds to the infant＇s level of arousal. Based on this premise, we review the acoustic structure and adaptive function of two types of distress calls, those given when infants were isolated from their mothers （isolation caUs） or captured by humans （capture calls）. We conducted a within-context comparison examining the two call types across a diverse se- lection of mammalian species and other vertebrate groups, followed by a comparison of how acoustic structure and function dif- fers between these contexts. In addition, we assessed acoustic traits that are critical to the response of caregivers. Across verte- brate species, distress vocalizations produced in these two behavioral contexts tend to be tonal with a simple chevron, flat or de- scending pattern of frequency modulation. Reports that both isolation and capture calls of vertebrate infants serve to attract care- givers are universal, and the fundamental frequency of infant vocalizations is often critical to this response. The results of our re- view are consistent with the hypothesis that differences in the acoustic structure of isolation and capture distress vocalizations re- flect differences in arousal, and not discrete functions. The similarity in acoustic structure and caregiver response observed across vertebrates adds support to the hypothesis that the production and processing of distress vocalizations are part of a highly-conserved system of social vocal behaviour in vertebrate

In this paper we describe the scanning electron microscopy techniques of electron backscatter diffraction, electron channeling contrast imaging, wavelength dispersive X-ray spectroscopy, and cathodoluminescence hyperspectral imaging. We present our recent results on the use of these non-destructive techniques to obtain information on the topography, crystal misorientation, defect distributions, composition, doping, and light emission from a range of UV-emitting nitride semiconductor structures. We aim to illustrate the developing capability of each of these techniques for understanding the properties of UV-emitting nitride semiconductors, and the benefits were appropriate, in combining the techniques.

Results of laser photocoagulation of a growing pigmented choroidal lesion for which the differential diagnosis included enlarging nevus and small melanoma and the rationale for such treatment are described. A37-year-old woman underwent ablative laser photocoagulation for a small juxtapapillary pigmented choroidal lesion that was documented photographically to enlarge in diameter from 300 to 1,750 microns after 8 years. Sixteen years after laser photocoagulation,the visual acuity remains 20/20 with no further growth or recurrence of the lesion. Local treatment may be justified for a small pigmented choroidal lesion that is documented to grow.

AIM:To investigate the progression of hepatic histopathology in serial liver biopsies from Wilson disease(WD)***:We report a group of 12 WD patients treated with zinc and/or penicillamine who underwent multiple follow-up liver ***,clinical and laboratory data were gathered and all patients underwent an initial biopsy and at least one repeat ***:Time to repeat biopsy ranged from 2 to 12 *** patients(non-progressors)showed stable hepatic histology or *** one case,we observed improvement of fibrosis from stage 2 to *** patients(progressors)had worsening of *** was no significant correlation between the histological findings and serum aminotransferases or copper me-tabolism *** hepatic copper concentration reached normal levels in only two patients:one from the non-progressors and one from the progressors *** estimated rate of progression of hepatic fibrosis in the entire group was 0 units per year in the time frame between the first and the second liver biopsy(4 years),and 0.25 between the second and the third(3 years).In the progressors group,the rate of progression of liver fibrosis was estimated at 0.11 fibrosis units per year between the first and second biopsy and,0.6 fibrosis units between the second and third ***:The inability of clinical tools to detect fibrosis progression in WD suggests that a liver biopsy with hepatic copper quantification every 3 years should be considered.
