Intestine Enzyme-responsive Polysaccharide-based Hydrogel to Open Epithelial Tight Junctions for Oral Delivery of Imatinib against Colon Cancer

Imatinib has been widely used as a selective kinase inhibitor for treating a variety of cancers,and this molecule is very hydrophobic so it is usually modified with mesylate salt in clinic to increase bioavailability.However,pH-dependent aqueous solubility and relatively high dosage of imatinib mesylate greatly reduce the clinical outcomes.To solve this problem,we developed an intestine enzyme-responsive hydrogel to efficiently encapsulate hydrophobic imatinib with long-term controlled release and enhanced intestinal permeability through oral administration.Methacrylic anhydride-modified carboxymethyl chitosan(MA-CMCS)was synthesized via amidation reaction and then MA-CMCS was crosslinked with photoinitator under UV-irradation to form a three-dimensional hydrophilic polymer network.The intestine enzyme responsiveness was endowed with imatinib-loaded hydrogel through hydrolyzation of glucosidic bond,which could achieve enzyme-triggered long-term drug release of up to 2 days.Furthermore,sodium deoxycholate was embedded into the hydrogel to synchronously open epithelial tight junctions with improved intestinal permeability.In vitro studies revealed similar lethality against colon cancer cell for both imatinib mesylate and imatinib-loaded hydrogels.Moreover,significantly enhanced in vivo tumor inhibition(6-fold higher compared to imatinib mesylate)was achieved after oral administration with imatinib-loaded hydrogels.Overall,this enzyme-responsive hydrogel could achieve long-term synchronous release of kinase inhibitor(imatinib)and tight junction permeation enhancer(sodium deoxycholate)at intestine with enhanced therapeutic efficiency,which could provide an effective approach to improve the bioavailability of hydrophobic anticancer chemodrugs with oral administration.

BACKGROUND Extreme heat exposure is a growing health problem,and the effects of heat on the gastrointestinal(GI)tract is unknown.This study aimed to assess the incidence of GI symptoms associated with heatstroke and its impact on outcomes.AIM To assess the incidence of GI symptoms associated with heatstroke and its impact on outcomes.METHODS Patients admitted to the intensive care unit(ICU)due to heatstroke were included from 83 centres.Patient history,laboratory results,and clinically relevant outcomes were recorded at ICU admission and daily until up to day 15,ICU discharge,or death.GI symptoms,including nausea/vomiting,diarrhoea,flatulence,and bloody stools,were recorded.The characteristics of patients with heatstroke concomitant with GI symptoms were described.Multivariable regression analyses were performed to determine significant predictors of GI symptoms.RESULTS A total of 713 patients were included in the final analysis,of whom 132(18.5%)patients had at least one GI symptom during their ICU stay,while 26(3.6%)suffered from more than one symptom.Patients with GI symptoms had a significantly higher ICU stay compared with those without.The mortality of patients who had two or more GI symptoms simultaneously was significantly higher than that in those with one GI symptom.Multivariable logistic regression analysis revealed that older patients with a lower GCS score on admission were more likely to experience GI symptoms.CONCLUSION The GI manifestations of heatstroke are common and appear to impact clinically relevant hospitalization outcomes.

Biodegradable Polypeptide-based Vesicles with Intrinsic Blue Fluorescence for Antibacterial Visualization

Fluorescence imaging has been an indispensable tool to provide dynamic information about the localization and quantity of organisms.Meanwhile,due to the intrinsic hollow structure and modularized biofunctionalities,polymer vesicles have been widely applied in biomedical field.However,most polymer vesicles are embedded with organic fluorophores for fluorescence imaging,which have certain drawbacks such as leakage and possible cytotoxicity.Here,we present a biodegradable polypeptide-based vesicle with intrinsic blue fluorescence without introducing any fluorophore for real-time visualization of antibacterial process.Through modular design to integrate multiple functional fragments,poly(ε-caprolactone)-block-poly(tryptophan)-block-poly(lysine-stat-phenylalanine)[PCL25-b-PTrp2-b-P(Lys13-statPhe4)]was synthesized,where PCL chains form the hydrophobic membrane,P(Lys-stat-Phe)and PTrp provide intrinsic fluorescence and broadspectrum antibacterial activity.It is noteworthy that the fluorescence emission was shifted from invisible ultraviolet range of amino acids to visible range(emission maximum at 436 nm),which makes it possible to visualize the antibacterial process.In addition,through utilizing the intrinsic fluorescence of vesicles,confocal fluorescent imaging of vesicles with bacteria validated the specific adhesion of vesicle towards bacteria,and the bacterial death through membrane disruption.Overall,we provided a novel approach to developing biodegradable fluorescent polypeptide-based vesicles for real-time visualization of antibacterial process.

Coherent link between a Ti:sapphire comb and a 1.5μm laser via nonlinear interaction in photonic crystal fiber

Optical clock networks have distinct advantages for the dissemination of time/frequency,geodesy,and fundamental research.To realize such a network,the telecom band and optical atomic clocks have to be coherently bridged.Since the telecom band and optical atomic clocks reside in a distinct spectral region,second-harmonic generation is usually introduced to bridge the large frequency gap.In this paper,we introduce a new method to coherently link a 1550 nm continuous wave laser with a Ti:sapphire mode-locked laser-based optical frequency comb.By coupling the 1550 nm continuous wave laser light and the Ti:sapphire comb light together into a photonic crystal fiber,nonlinear interaction takes place,and new comblike frequency components related to the 1550 nm laser frequency are generated in the visible region.Consequently,we can detect beat notes between two combs in the visible region with a signal-to-noise ratio of more than 40 dB in a resolution bandwidth of300 kHz.With this signal,we realize an optical frequency divider for converting the frequency of optical clocks in the visible region to the telecom band at 1.55μm.An out-of-loop measurement shows that the additional noise and uncertainty induced in optical frequency conversion are 5×10^(-18)at 1 s averaging time and 2.2×10^(-19),respectively,which are limited by the uncompensated light path fluctuation but fulfill precision measurement using state-of-the-art optical clocks.

Risk pre-assessment method for regional drilling engineering based on deep learning and multi-source data

Accurately predicting downhole risk before drilling in new exploration areas is one of the difficulties.Using intelligent algorithms to explore the complex relationship between multi-source data and downhole risk is a hot research topic and frontier in this field.However,due to the small number and uneven distribution of drilled wells in new exploration areas and the lack of sample data related to risk,the training model has insufficient generalization ability,and thus the prediction is not effective.In this paper,a drilling risk profile(depth domain)rich in geological and engineering information is constructed by introducing a quantitative evaluation method for drilling risk of drilled wells,which can provide sufficient risk sample data for model training and thus solve the small sample problem.For the problem of uneven distribution of drilling wells in new exploration areas,the concept of virtual wells and their deployment methods were proposed.Besides,two methods for calculating rock mechanical parameters of virtual wells were proposed,and the accuracy and applicability of the two methods are analyzed.The LSTM deep learning model was optimized to tap the quantitative relationship between drilling risk profiles and multi-source data(e.g.,seismic,logging,and rock mechanical parameters).The model was validated to have an average relative error of 9.19%.The quantitative prediction of the drilling risk profile of the virtual well was achieved using the trained LSTM model and the calculation of the relevant parameters of the virtual well.Finally,based on the sequential Gaussian simulation method and the risk distribution of drilled and virtual wells,a regional 3D drilling risk model was constructed.The analysis of real cases shows that the addition of virtual wells can significantly improve the identification of regional drilling risks and the prediction accuracy of pre-drill drilling risks in unexplored areas can be improved by up to 21%compared with the 3D risk model constructed based on drilled wells only.

Graphdiyne oxide substrate-enhanced peroxidase-mimicking performance of Ru nanoparticles with physiological pH preference

Modulating electronic structure of metal nanoparticles via metal–support interaction has attracted intense interest in the field of catalytic science.However,the roles of supporting substrates in regulating catalytic properties of nanozymes remain elusive.In this study,we find that the use of graphdiyne oxide(GDYO)as the substrate for self-terminating growth of Ru nanoparticles(Ru@GDYO)endows the peroxidase-like activity of Ru nanoparticles with intrinsic physiological pH preference and natural horseradish peroxidase(HRP)comparable performance.Ru nanoparticles electrolessly deposited onto GDYO possess a partially oxidized electronic structure owing to limited charge transfer between Ru and GDYO,contributing to the intrinsic physiological pH preference of the peroxidase-mimicking nanozyme.More importantly,the substrate GDYO plays an influential factor in enhancing catalytic activity,that is,the activity of Ru@GDYO is much higher than that of Ru nanoparticles deposited on other carbon substrates including graphene oxides and graphdiyne.To demonstrate the application of Ru@GDYO nanozyme in neutral solutions,we employ Ru@GDYO with nicotinamide adenine dinucleotide(NAD+)-dependent dehydrogenases in physiological conditions to realize a sustainable cascade reaction by means of forming continuous NAD^(+)/dihydronicotiamide adenine dinucleotide(NADH)recycling.Our finding represents a promising perspective on designing high-performance peroxidase-mimicking nanozymes with broader applicability,raising fundamental understanding of structure–activity relationship,and investigating new applications of nanozymes in biological systems.

Deterministic Learning-Based Neural PID Control for Nonlinear Robotic Systems

Traditional proportional-integral-derivative(PID)controllers have achieved widespread success in industrial applications.However,the nonlinearity and uncertainty of practical systems cannot be ignored,even though most of the existing research on PID controllers is focused on linear systems.Therefore,developing a PID controller with learning ability is of great significance for complex nonlinear systems.This article proposes a deterministic learning-based advanced PID controller for robot manipulator systems with uncertainties.The introduction of neural networks(NNs)overcomes the upper limit of the traditional PID feedback mechanism’s capability.The proposed control scheme not only guarantees system stability and tracking error convergence but also provides a simple way to choose the three parameters of PID by setting the proportional coefficients.Under the partial persistent excitation(PE)condition,the closed-loop system unknown dynamics of robot manipulator systems are accurately approximated by NNs.Based on the acquired knowledge from the stable control process,a learning PID controller is developed to further improve overall control performance,while overcoming the problem of repeated online weight updates.Simulation studies and physical experiments demonstrate the validity and practicality of the proposed strategy discussed in this article.

A practical guide for fast implementation of SNARE-mediated liposome fusion

Soluble N-ethylmaleimide-sensitive factor attachment protein receptor(SNAER) family proteins are the engines of most intra-cellular and exocytotic membrane fusion pathways(Jahn and Scheller 2006).Over the past two decades, in-vitro liposome fusion has been proven to be a powerful tool to reconstruct physiological SNARE-mediated membrane fusion processes(Liu et al. 2017). The reconstitution of the membrane fusion process not only provides direct evidence of the capability of the cognate SNARE complex in driving membrane fusion but also allows researchers to study the functional mechanisms of regulatory proteins in related pathways(Wickner and Rizo 2017). Heretofore, a variety of delicate methods for in-vitro SNARE-mediated liposome fusion have been established(Bao et al. 2018;Diao et al. 2012; Duzgunes 2003; Gong et al. 2015; Heo et al. 2021; Kiessling et al. 2015; Kreye et al.2008; Kyoung et al. 2013; Liu et al. 2017; Scott et al. 2003). Although technological advances have made reconstitution more physiologically relevant, increasingly elaborate experimental procedures, instruments, and data processing algorithms nevertheless hinder the non-experts from setting up basic SNARE-mediated liposome fusion assays. Here, we describe a low-cost, timesaving, and easy-to-handle protocol to set up a foundational in-vitro SNARE-mediated liposome fusion assay based on our previous publications(Liu et al. 2023; wang and Ma 2022). The protocol can be readily adapted to assess various types of SNARE-mediated membrane fusion and the actions of fusion regulators by using appropriate alternative additives(e.g., proteins, macromolecules, chemicals, etc.). The total time required for one round of the assay is typically two days and could be extremely compressed into one day.

Nanomedicine delivery technology plays an important role in modern medicine and has shown good therapeutic effects in scientific research.Polysaccharides have the characteristics of wide sources,excellent biocompatibility,and non-toxicity.In addition,there are multifunctional groups on the main chain of polysaccharides,which can be surface-modified or functionalized to have targeting ability through specific sugar parts.Amphiphilic polysaccharide micelles with good biocompatibility,degradability,high safety,easy structural modification,and special core-shell structure are regarded as ideal carriers for nanomedicines.Therefore,this review is focused on the hydrophobic modification designs of polysaccharides,the preparation methods and characteristics of micelles,and the applications of amphiphilic polysaccharide micelles in the field of biomedicine.It is expected to provide some ideas and inspiration for the design of polysaccharide drug carriers.
