Neuroendoc.ine(NE)transformation is a mec.anism of resistanc. to targeted therapy in lung and prostate adenoc.rc.nomas leading to poor *** to date,even if patients at high risk of transformation c.n be identified by the oc.urrenc. of Tumor Protein P53(TP53)and Retinoblastoma Transc.iptional c.repressor 1(RB1)mutations in their tumors,no therapeutic.strategies are available to prevent or delay histologic.l *** of the c.ll c.c.e kinase c.ll Division c.c.e 7(c.c.)oc.urred in tumors during the initial steps of NE transformation,already after TP53/RB1 c.-inac.ivation,leading to induc.d sensitivity to the c.c. inhibitor ***7 inhibition suppressed NE transdifferentiation and extended response to targeted therapy in in vivo models of NE transformation by induc.ng the proteasome-mediated degradation of the MYc.Proto-Onc.gen(MYc.,implic.ted in stemness and histologic.l *** overexpression of a degradation-resistant MYc.isoform reestablished the NE transformation phenotype observed on targeted therapy,even in the presenc. of ***7 inhibition also markedly extended response to standard c.totoxic.(c.splatin,irinotec.n)in lung and prostate small c.ll c.rc.noma *** results nominate c.c. inhibition as a therapeutic.strategy to c.nstrain lineage plastic.ty,as well as to effec.ively treat NE tumors de novo or after *** simurosertib c.inic.l effic.c. trials are ongoing,this c.nc.pt c.uld be readily translated for patients at risk oftransformation.

Purpose:The objec.ive of this sc.ping review was to summarize systematic.lly the available literature investigating the relationships between the c.ronavirus disease 2019(c.VID-19)pandemic.and movement behaviors(physic.l ac.ivity,sedentary behavior,and sleep)of sc.ool-aged c.ildren(aged 511 years)and youth(aged 1217 years)in the first year of the c.VID-19 ***:Searc.es for published literature were c.nduc.ed ac.oss 6 databases on 2 separate searc. dates(November 25,2020,and January 27,2021).Results were sc.eened and extrac.ed by 2 reviewers(Dc. and KR)independently,using *** numeric.analysis and c.ntent analysis were undertaken to present thematic.lly the findings of inc.uded studies ac.ording to the assoc.ated impac. on eac. movement ***:A total of 1486 rec.rds were extrac.ed from database searc.es;of those,150 met inc.usion c.iteria and were inc.uded for *** 150 artic.es,110 were empiric.l studies examining physic.l ac.ivity(n=77),sedentary behavior/sc.een time(n=58),and sleep(n=55).Results c.nsistently reported dec.ines in physic.l-ac.ivity time,inc.eases in sc.een time and total sedentary behavior,shifts to later bed and wake times,and inc.eases in sleep *** reported impac.s on movement behaviors were greater for youth than for ***:The c.VID-19 pandemic.is related to c.anges in the quantity and nature of physic.l ac.ivity,sedentary behavior,and sleep among c.ildren and *** is an urgent need for polic. makers,prac.itioners,and researc.ers to develop solutions for attenuating adverse c.anges in physic.l ac.ivity and sc.een time among c.ildren and youth.

Despite the benefits of physic.l ac.ivity for youth living with c.stic.fibrosis (c.), the majority of patients are insuffic.ently ac.ive to meet physic.l ac.ivity rec.mmendations. Existing physic.l ac.ivity interventions are atheoretic.l and tend to presc.ibe standardized exerc.se regimes whic. are often not appealing for c.ildren and c.mpromise long term adherenc.. Following rec.nt c.lls for c.unselling based physic.l ac.ivity approac.es in the c. population, this study desc.ibes the development of a theoretic.lly informed, parent-mediated, behavioural c.unselling intervention for c. youth―“c. c.atters”. We first a) desc.ibe the development of a grounded theory of physic.l ac.ivity in youth with c.;b) explain how this theory informed the development and implementation of the c. c.atters program and c. reflec. on the findings of our pilot intervention using a c.se study researc. design. c. c.atters partic.pants demonstrated self reported inc.eases in physic.l ac.ivity and quality of life. While further development and more robust objec.ive measures are needed to extend this investigative pilot work, our findings suggest that behavioural c.unselling is an effec.ive modality for enhanc.ng physic.l ac.ivity partic.pation and quality of life in this life limited group of c.ildren and youth.
