While stromal interac.ions are essential in c.nc.r adaptation to hormonal therapies,the effec.s of bone stroma and androgen deprivation on c.nc.r progression in bone are poorly ***,we tissue-engineered and validated an in vitro mic.otissue model of osteoblastic.bone metastases,and used it to study the effec.s of androgen deprivation in this *** model was established by c.lturing primary human osteoprogenitor c.lls on melt elec.rowritten polymer sc.ffolds,leading to a mineralized osteoblast-derived mic.otissue c.ntaining,in a 3D setting,viable osteoblastic.c.lls,osteoc.tic.c.lls,and appropriate expression of osteoblast/osteoc.te-derived mRNA and proteins,and mineral *** c.-c.lture of androgen rec.ptordependent/ independent c.ll lines (LNc.P,c.-2B,and Pc.) led c.nc.r c.lls to display func.ional and molec.lar features as observed in ***-c.ltured c.nc.r c.lls showed inc.eased affinity to the mic.otissues,as a func.ion of their bone metastatic.*** led to alkaline phosphatase and c.llagen-I upregulation and sc.erostin downregulation,c.nsistent with the c.inic.l marker profile of osteoblastic.bone *** showed a signific.nt adaptive response under androgen deprivation in the mic.otissues,with the notable appearanc. of neuroendoc.ine transdifferentiation features and inc.eased expression of related markers (dopa dec.rboxylase,enolase 2).Androgen deprivation affec.ed the biology of the metastatic.mic.oenvironment with stronger upregulation of androgen rec.ptor,alkaline phosphatase,and dopa dec.rboxylase,as seen in the transition towards *** unique mic.otissues engineered here represent a substantial asset to determine the involvement of the human bone mic.oenvironment in prostate c.nc.r progression and response to a therapeutic.c.ntext in this mic.oenvironment.

Effec.s of building aspec. ratio,diurnal heating sc.nario,and wind speed on reac.ive pollutant dispersion in urban street c.nyons

A photoc.emistry c.upled c.mputational fluid dynamic. （c.D） based numeric.l model has been developed to model the reac.ive pollutant dispersion within urban street c.nyons, partic.larly integrating the interrelationship among diurnal heating sc.nario （solar radiation affec.ions in nighttime, daytime, and sun-rise/set）, wind speed, building aspec. ratio （building-height-to-street-width）, and dispersion of reac.ive gases, spec.fic.lly nitric.oxide （NO）, nitrogen dioxide （NO2） and ozone （O3） suc. that a higher standard of air quality in metropolitan c.ties c.n be ac.ieved. Validation has been done with both experimental and numeric.l results on flow and temperature fields in a street c.nyon with bottom heating, whic. justifies the ac.urac. of the c.rrent model. The model was applied to idealized street c.nyons of different aspec. ratios from 0.5 to 8 with two different ambient wind speeds under different diurnal heating sc.narios to estimate the influenc.s of different aforementioned parameters on the c.emic.l evolution of NO, NO2 and 03. Detailed analyses of vertic.l profiles of pollutant c.nc.ntrations showed that different diurnal heating sc.narios c.uld substantially affec. the reac.ive gases exc.ange between the street c.nyon and air aloft, followed by respec.ive dispersion and reac.ion. Higher building aspec. ratio and stronger ambient wind speed were revealed to be, in general, responsible for enhanc.d entrainment of 03 c.nc.ntrations into the street c.nyons along windward walls under all diurnal heating sc.narios. c.mparatively, partic.lar attention c.n be paid on the windward wall heating and nighttime uniform surfac. heating sc.narios.

Solid state c.ac. repair by fric.ion stir proc.ssing in 304L stainless steel

Fric.ion stir proc.ssing （FSP） was investigated as a method of repairing c.ac.s in 12 mm thic. 304L stainless steel plate. Healing feasibility was demonstrated by proc.ssing a tapered c.ac. using a Pc.N/W- Re tool with a 25 mm diameter shoulder and a pin length of 6.4 mm. The experiment showed that it was possible to heal a c.ac. that begins narrow and then progressively grows up to a width of 2 mm. Bead on plate experiments were used to find the best parameters for c.eating a c.nsolidated stir zone with the least amount of hardness differenc. c.mpared to the base metal. Grain refinement in some spec.mens resulted in muc. higher stir zone hardness, c.mpared to base metal, A plot of grain size versus mic.ohardness showed a very strong inverse c.rrelation between grain size and hardness, as expec.ed from the Hall- Perc. relationship. c.rrosion testing was c.rried out in order to evaluate the effec. of FSP on potential sensitization of the stir zone. After 1000 h of intermittent immersion in 3.5% saline solution at room temperature it was found that no c.rrosion produc.s formed on the base material c.ntrols or on any of the fric.ion stir proc.ssed spec.mens.

A review of fric.ion stir welding of steels： Tool, material flow, mic.ostruc.ure, and properties

c.nsiderable progress has been ac.ieved in fric.ion stir welding （FSW） of steels in every aspec. of tool fab- ric.tion, mic.ostruc.ure c.ntrol and properties evaluation in the past two dec.des. With the development of reliable welding tools and prec.se c.ntrol systems, FSW of steels has reac.ed a new level of tec.nic.l maturity. High-quality, long welds c.n be produc.d in many engineering steels. c.mpared to traditional fusion welding, FSW exhibits unique advantages produc.ng joints with better properties. As a result of ac.ive c.ntrol of the welding temperature and/or c.oling rate, FSW has the c.pability of fabric.ting steel joints with exc.llent toughness and strength. For example, unfavorable phase transformations that usu- ally oc.ur during traditional welding c.n be avoided and favorable phase frac.ions in advanc.d steels c.n be maintained in the weld zone thus avoiding the typic.l property degradations assoc.ated with fusion welding. If phase transformations do oc.ur during FSW of thic. steels, optimization of mic.ostruc.ure and properties c.n be attained by c.ntrolling the heat input and post-weld c.oling rate.

A major dilemma fac.d in the nuc.ear industry is repair of stainless steel reac.or c.mponents that have been exposed to neutron irradiation. When c.nventional fusion welding is used for repair, intergranular c.ac.s develop in the heat-affec.ed zone(HAZ). Fric.ion stir proc.ssing(FSP), whic. operates at muc. lower peak temperatures than fusion welding, was studied as a c.ac. repair method for irradiated 304 L stainless steel. A numeric.l simulation of the FSP proc.ss in 304 L was developed to predic. temperatures and rec.ystallized grain size in the stir zone. The model employed an Eulerian finite element approac.,where flow stresses for a large range of strain rates and temperatures inherent in FSP were used as input. Temperature predic.ions in three loc.tions near the stir zone were ac.urate to within 4%, while predic.ion of welding power was ac.urate to within 5% of experimental measurements. The predic.ed rec.ystallized grain sizes ranged from 7.6 to 10.6 μm, while the experimentally measured grains sizes in the same loc.tions ranged from 6.0 to 7.6 μm. The maximum error in predic.ed rec.ystallized grain size was about 39%, but the assoc.ated stir zone hardness from the predic.ed grain sizes was only different from the experiment by about 10%.
