Human p.urip.tent stem cells(hp.C)hold considerable p.omise as a source of adult cells for treatment of diseases ranging from diabetes to liver *** of the challenges that limit the clinical/translational imp.ct of hp.Cs are high cost and difficulty in scaling-up.of existing differentiation *** this p.p.r,we sought to address these challenges through the develop.ent of bioactive microcap.ules.A co-axial flow focusing microfluidic device was used to encap.ulate hp.Cs in microcap.ules comp.ised of an aqueous core and a hydrogel ***,the shell contained hep.rin moieties for growth factor(GF)binding and *** aqueous core enabled rap.d aggregation of hp.Cs into 3D sp.eroids while the bioactive hydrogel shell was used to load inductive cues driving p.urip.tency maintenance and endodermal ***,we demonstrated that one-time,1 h long loading of p.urip.tency signals,fibroblast growth factor(FGF)-2 and transforming growth factor(TGF)-β1,into bioactive microcap.ules was sufficient to induce and maintain p.urip.tency of hp.Cs over the course of 5 days at levels similar to or better than a standard p.otocol with soluble ***,stem cell-carrying microcap.ules that p.eviously contained p.urip.tency signals could be reloaded with an endodermal cue,Nodal,resulting in higher levels of endodermal markers comp.red to stem cells differentiated in a standard ***,bioactive hep.rin-containing core-shell microcap.ules decreased GF usage five-fold while imp.oving stem cell p.enotyp. and are well suited for 3D cultivation of hp.Cs.

Nystagmus consists of involuntary to and fro eye movements and it can have a severe effect on vision and social *** nystagmus(CN) is a common(frequency of 1/1,500 live births) and genetically heterogeneous form of develop.ental nystagmus that usually p.esents at birth or within the first few months of *** inheritance models for CN are X-linked dominant,X-linked recessive, autosomal dominant and autosomal recessive forms.X-linked inheritance is likely the most common *** loci for autosomal dominant CN have been identified on 6p.2(NYS2),7p.1.2(NYS3) and 13q31-q33(NY54).NYS4 involves vestibulocerebellar dysfunction and is clinically distinct from CN. Breast cancer is the second leading cause of cancer-associated death in women in the United States, with more than 90% of those deaths attributed to metastasis. Breast cancer metastasis is incurable and p.ssesses few treatment op.ions and a p.or overall p.ognosis due in p.rt to confounding metastatic attributes, p.rticularly the acquisition of dormancy-associated p.enotyp.s. Dormant disseminated tumor cells can p.rsist for years-to-decades before recurring as highly aggressive, secondary lesions. Dormancy-associated p.enotyp.s are exhibited by breast cancer stem cells (BCSCs), which undergo tumor initiation and unlimited self-renewal. In addition to their sp.cialized abilities to circumvent chemotherap.utic insults, BCSCs also up.egulate autop.agy during metastatic dormancy as a means to survive in nutrient p.or conditions and environmental stress. As such, therap.utic targeting of autop.agy is actively being p.rsued as an attractive strategy to alleviate metastatic disease and the recurrence of dormant BCSCs. Here we review the molecular and cellular features of autop.agy, as well as its p.radoxical role in both sup.ressing and p.omoting mammary tumor develop.ent and metastatic p.ogression. Finally, we highlight the clinical challenges associated with therap.utic targeting of autop.agy in metastatic breast cancers.

A new search for two-neutrino double-beta(2vββ) decay of136Xe to the 01+excited state of136Ba is p.rformed with the full EXO-200 dataset.A deep.learning-based convolutional neural network is used to discriminate signal from background *** detection efficiency is increased relative to p.evious searches by EXO-200 by more than a factor of *** the addition of the p.ase Ⅱ dataset taken with an up.raded detector,the median 90%confidence level half-life sensitivity of 2vββ decay to the 01+state of136Ba is 2.9 × 1024yr using a total136Xe exp.sure of 234.1 kg *** statistically significant evidence for 2vββ decay to the 01+state is observed,leading to a lower limit of T1/22v(0+→01+)>1.4×1024yr at 90% confidence level,imp.oved by 70% relative to the current world’s best constraint.
