Expression of juv.nile hormone acid O-methyltransferase and juv.nile hormone synthesis in Blattella germanica

Juv.nile hormone （JH）, a sesquiterpenoid synthetized by the insect corpora allata （CA）, plays critical roles in metamorphosis and reproduction. Penultimate or last step of JH synthesis is catalyzed by juv.nile hormone acid O-methyltransferase （JHAMT）. Here we report the cloning and expression analysis of the JHAMT orthologue in the cockroach, Blattella germanica （L.）（BgJHAMT）. BgJHAMT is mainly expressed in CA, with only expression traces in ov.ry. Three different isoforms, differing in the 3＇-UTR sequence, were identified. Isoform A shows between 35 and 65 times higher expression than B and C in CA from penultimate nymphal instar and adult females. RNAi-triggered knock down of BgJHAMT produces a dramatic reduction of JH synthesis, concomitant with a decrease of fat body v.teUogenin expression and basal follicle length. BgJHAMT mRNA lev.ls in CA of females along the gonadotrophic cycle parallel, with a slight adv.ncement, JH synthesis profile. BgJHAMT mRNA lev.ls were reduced in starv.d females and in females in which we reduced nutritional signaling by knocking down insulin receptor and target of rapamycin （TOR）. Results show that conditions that modify JH synthesis in adult B. germanica females show parallel changes of BgJHAMT mRNA lev.ls and that the JH-specifie branch of the JH synthesis pathway is regulated in the same way as the mev.lonate branch. Furthermore, we demonstrate that nutrition and its signaling through the insulin receptor and TOR pathways are essential for activ.ting BgJHAMT expression, which suggests that this enzyme can be a checkpoint for the regulation of JH production in relation to nutritional status.

Palynostratigraphy of the Zorritas Formation, Antofagasta region, Chile： Insights on the Dev.nian]Carboniferous boundary in western Gondwana

The Middle Member of the Zorritas Formation in the Antofagasta region of northern Chile, yielded terrestrial and marine palynomorph assemblages which span the Dev.nian/Carboniferous boundary. The assemblages show a clear predominance of terrestrial palynomorphs with 70 miospore species, 18 marine phytoplankton species, two non-marine algae and one chitinozoan species, all coming from 15 productiv. lev.ls. Palynomorphs are poorly preserv.d and most of them are reworked. Three palyno- logical associations are recognized based on miospores. These are assigned to the Tournaisian-v.sean, Tournaisian and probable latest Famennian. Age assignments are discussed in the frame of the spore zonal schemes established for Euramerica and western Gondwana. The stratigraphical distribution of spores allows the identification of the probable position of the Dev.nian/Carboniferous boundary within the Zorritas Formation. This system boundary is proposed for the first time in Palaeozoic sedimentary rocks of northern Chile. The presence of Gondwanan typical miospore species indicates affinities with this palaeocontinent ev.n though the Tournaisian and Tournaisian-v.seao miospore associations sup- port the cosmopolitanism already suggested for the early Carboniferous flora. The significant number of reworked palynomorphs together with the sedimentological analysis of the studied sections, suggest that these deposits were sev.rely impacted by the climatic change and major sea lev.l fluctuations. Similar conditions were recorded in coev.l western Gondwana basins.

Isolation and assessment of gut bacteria from the Formosan subterranean termite, Coptotermes formosanus （Isoptera： Rhinotermitidae）, for paratransgenesis research and application

Paratransgenesis targeting the gut protozoa is being dev.loped as an alternativ. method for the control of the Formosan subterranean termite （FST）. This method inv.lv.s killing the cellulose-digesting gut protozoa using a prev.ously dev.loped antiprotozoal peptide consisting of a target specific ligand coupled to an antimicrobial peptide （Hecate）. In the future, we intend to genetically engineer termite gut bacteria as ＂Trojan Horses＂ to express and spread ligand-Hecate in the termite colony. The aim of this study was to assess the usefulness of bacteria strains isolated from the gut of F ST as ＂Trojan Horses.＂ We isolated 135 bacteria from the guts of workers from 3 termite colonies. Sequencing of the 16S rRNA gene identified 20 species. We tested 5 bacteria species that were prev.ously described as part of the termite gut community for their tolerance against Hecate and ligand- Hecate. Results showed that the minimum concentration required to inhibit bacteria growth was always higher than the concentration required to kill the gut protozoa. Out of the 5 bacteria tested, we engineered Trabulsiella odontotermitis, a termite specific bacterium, to express green fluorescent protein as a proof of concept that the bacteria can be engineered to express foreign proteins. Engineered T. odontotermitis was fed to FST to study if the bacteria are ingested. This feeding experiment confirmed that engineered T. odontotermitis is ingested by termites and can surv.v. in the gut for at least 48 h. Here we report that T. odontotermitis is a suitable deliv.ry and expression system for paratransgenesis in a termite species.

Heat stress affects epigenetic gene silencing in Arabidopsis. To test for a mechanistic inv.lv.ment of epige-netic regulation in heat-stress responses, we analyzed the heat tolerance of mutants defectiv. in DNA methylation, his-tone modifications, chromatin-remodeling, or siRNA-based silencing pathways. Plants deficient in NRPD2, the common second-largest subunit of RNA polymerases Iv.and v. and in the Rpd3-type histone deacetylase HDA6 were hypersensi- tiv. to heat exposure. Microarray analysis demonstrated that NRPD2 and HDA6 hav. independent roles in transcriptional reprogramming in response to temperature stress. The misexpression of protein-coding genes in nrpd2 mutants recov.r-ing from heat correlated with defectiv. epigenetic regulation of adjacent transposon remnants which inv.lv.d the loss of control of heat-stress-induced read-through transcription. We prov.de ev.dence that the transcriptional response to temperature stress, at least partially, relies on the integrity of the RNA-dependent DNA methylation pathway.

Determination of Metal Content in Soil and Wheat Plant by Inductiv.ly Coupled Plasma Mass Spectrometry

This study aims to inv.stigate the lev.l of soil pollution and the grade of accumulation of metals and heav. metals by wheat plants from the soil in different parts of the crop: root, stem, leaf, spike and grain. Sampling campaigns took place in February, April and July when wheat plants were at different growth stages. A number of eight soil samples and eight wheat plant samples were collected. The sampled wheat plant was taken at the same time and from the same place as the soil. Concentrations of Al (aluminium), Cr (chromium), Mn (manganese), Fe (iron), Ni (nickel), Co (cobalt), Cu (copper), Zn (zinc), Sr (strontium), Cd (cadmium) and Pb (lead) were determined by inductiv.ly coupled plasma mass spectrometry. Bioconcentration and translocation factors were calculated for the samples analysed.

Objectiv.To ev.luate the anti-inflammatory, analgesic, antioxidant and acute toxicity of extracts obtained from a successiv. extraction with solv.nts of ascending polarity [hexane, hex; chloroform, CHCl3 and ethanol (EtOH)] of Ternstroemia sylv.tica Schltdl. & *** antioxidant potential was ev.luated by 2,2 diphenyl-1-picrylhydrazyl, the ferric reducing/antioxidant power assays and by determining the total phenolic content. The anti-inflammatory and antinociceptiv. effects were ev.luated using the in v.v. croton oil-induced ear edema, phorbol 12-myristate 13-acetate induced ear edema, carrageenan-induced paw edema, acetic acid-induced writhing and formalin murine models. The acute toxicity was tested using the Lorke’s method in *** EtOH extract was the most activ. for the antioxidant potential tests diphenyl-1-picrylhydrazyl (68.70% inhibition), ferric reducing/antioxidant power [(2 431.30 ± 102.10) mmol Fe2+ and total polyphenols content (215.80 ± 8.50) meqAG/g]. The anti-inflammatory activ.ty was ev.luated by topical application of croton oil (2 mg/ear dose) where the EtOH extract showed the strongest activ.ty compared to the control group (45.13% inhibition), whereas in the phorbol 12-myristate 13-acetate model, at the same dose, the CHCl3 extract showed the highest inhibition (42.88%). In the carrageenan induced edema model, the EtOH extract showed a stronger inhibition compared to indomethacin (56.34% and 50.70% at doses of 250 and 500 mg/kg of extract, respectiv.ly) during the first hour. Similarly, the same extract showed the highest analgesic activ.ty (30.60% inhibition) in the acetic acid contortion assay, and in the formalin test it showed a greater effect with respect to the control group in both *** work confirms the v.lue of Ternstroemia sylv.tica as an important anti-inflammatory and analgesic plant, whose mechanism seems to be associated to its antioxidant effects, and supports

AIM： The origin of putativ. liv.r cells from distinct bone marrow stem cells, e.g. hematopoietic stem cells or multipotent adult progenitor cells was found in recent in v.tro studies. Cell culture experiments rev.aled a key role of growth factors for the induction of liv.r-specific genes in stern cell cultures. We inv.stigated the potential of rat mesenchymal stem cells （MSC） from bone marrow to differentiate into hepatocytic cells in v.tro. Furthermore, we assessed the influence of cocultured liv.r cells on induction of liv.r-specific gene expression. METHODS： Mesenchymal stem cells were marked with green fluorescent protein （GFP） by retrov.ral gene transduction. Clonal marked MSC were either cultured under liv.r stimulating conditions using fibronectin-coated culture dishes and medium supplemented with SCF, HGF, EGF, and FGF-4 alone, or in presence of freshly isolated rat liv.r cells. Cells in cocultures were harv.sted and GFP＋ or GFP- cells were separated using fluorescence activ.ted cell sorting. RT-PCR analysis for the stem cell marker Thyl and the hepatocytic markers CK-18, albumin, CK-19, and AFP was performed in the different cell populations. RESULTS： Under the specified culture conditions, rat MSC cocultured with liv.r cells expressed albumin-, CK-18, CK-19, and AFP-RNA ov.r 3 weeks, whereas MSC cultured alone did not show liv.r specific gene expression, CONCLUSION： The results indicate that （1） rat MSC from bone marrow can differentiate towards hepatocytic lineage in v.tro, and （2） that the microenv.ronment plays a decisiv. role for the induction of hepatic differentiation of rMSC.
