Classification and authentication of spices and aromatic herbs by means of HPLC-UV and chemometrics

Recent increase in the adulteration of spices and aromatic herbs in food industry constitutes a problem that requires exhaustive quality *** every spice has a different composition with characteristic biomarkers,chromatographic profiles are especially valuable to authenticate these ***,in this work a new high performance liquid chromatography(HPLC)method with UV-vis detection was developed for the characterization,identification and authentication of cinnamon,oregano,thyme,sesame,bay leaf,clove,cumin,and *** separation was optimized based on the separation of six characteristic biomarkers(sesamol,eugenol,thymol,carvacrol,salicylaldehyde and vainillin)and was performed using a C18 reversed-phase column under a 35 min gradient elution based on 0.1%(v/v)formic aqueous solution and methanol by means of UV-Vis detection at 280 nm.87 samples,purchased in local supermarkets,were analyzed and the obtained profiles were processed by chemometric ***,data treatment was evaluated by principal component analysis(PCA);next soft independent modelling by class analogy(SIMCA)and partial least squares discriminant analysis(PLS-DA)were carried out in order to verify if classification according to their biomarkers was *** study concluded that PLS-DA(0.14-0.75%global error)classifies better the types of spice or aromatic herb than SIMCA(0.82-3.67%global error).

Development of a vehicle-track interaction model to predict the vibratory benefits of rail grinding in the time domain

Imperfections in the wheel-rail contact are one of the main sources of generation of railway vibrations. Consequently, it is essential to take expensive corrective maintenance measures, the results of which may be unknown. In order to assess the effectiveness of these measures, this paper develops a vehicle-track interaction model in the time domain of a curved track with presence of rail corrugation on the inner rail. To characterize the behavior of the track, a numerical finite element model is developed using ANSYS software, while the behavior of the vehicle is characterized by a unidirectional model of two masses developed with VAMPIRE PRO software. The overloads obtained with the dynamic model are applied to the numerical model and then, the vibrational response of the track is obtained. Results are validated with real data and used to assess the effectiveness of rail grinding in the reduction of wheel-rail forces and the vibration generation phenomenon.

Immune response-related genes associated to blocking midgut dengue virus infection in Aedes aegypti strains that differ in susceptibility

Aedes (Stegomyia) aegypti, the principal global vector of dengue viruses, has difTerences in its susceptibility to dengue virus infection. We compared the global expres? sion of genes in the midguts of Colombian Ae. aegypti dengue-susceptible (Cali-S) and dengue-refractory (Cali-MIB) field derived strains after ingesting either a sugarmeal, a bloodmeal, or a bloodmeal containing dengue virus serotype 2 (DENV-2). Microarraybased transcriptome analysis among treatments indicated a total of 4725 transcripts with differential expression between the two strains. Eleven genes were selected from different functional groups based on their significant up or down expression levels as well as reports in the literature suggesting they are associated with dengue virus elimination. We measured mRNA abundance of these 11 genes at 0, 8, 24, and 36 h postinfection using quantitative real time PCR (qPCR) to confirm the microarray results and assess any temporal patterns. Four genes were selected (Gram-negative binding protein—GNBP [AAEL009176], Niemann Pick Type-C2一NPC2 [AAEL015136], Keratinocyte lectin [AAEL009842], and Cathepsin-b [AAEL007585]) for knockdown experiments using RNA interference (RNAi) methodology to determine the phenotype (DENV-2 susceptible or refractory). Silencing GNBP、Cathepsin-b and Keratinocyte lectin reduced the percentage of mosquitoes with disseminated virus in the Cali-S strain to 8%, 20%, and 12% respectively compared with 96% in the controls. Sile ncing of NPC2 increased the percentage of mosquitos with disseminated virus infections in Cali-MIB to 66% compared with 35% in the controls. This study provides in sight into genes that may contribute to the Cali-S susceptible and Cali-MIB refractory phenotypes in 昇匕 aegypti.

Alpha-1 antitrypsin deficiency and the risk of hepatocellular carcinoma in end-stage liver disease

AIM:To evaluate the association between alpha-1 antitrypsin deficiency(A1ATD) and hepatocellular carcinoma(HCC) in patients with end-stage liver disease(ESLD).METHODS:Patients with cirrhosis and ESLD referred to the Cleveland Clinic Foundation for liver transplantation between 2003 and 2014 were included in the study(N = 675). ESLD was defined as having histological features of cirrhosis and/or radiological evidence of cirrhosis in the context of portal hypertension(ascites,variceal bleeding,thrombocytopenia,or hepatic encephalopathy). A1 ATD was diagnosed using phenotype characterization(MZ or ZZ),liver biopsy detection of PAS-positive diastaseresistant(PAS+) globules,or both. Patients with other causes of liver diseases such as hepatitis C virus(HCV),alcoholic liver disease and non-alcoholic steatohepatitis(NASH) or NASH were also included in the study. HCC was diagnosed by using imaging modalities,biopsy findings,or explanted liver inspection. Follow-up time was defined as the number of years from the diagnosis of cirrhosis to the diagnosis of hepatocellular carcinoma,or from the diagnosis of cirrhosis to the last follow up visit. The rate of HCC was assessed using time-tointerval analysis for interval censored ***:This study included 675 patients. 7% of subjects had A1ATD(n = 47). Out of all subjects who did not have A1 ATD,46% had HCV,17% had alcoholic liver disease,19% had NASH and 18% had another primary diagnosis. Of the 47 subjects with A1 ATD,15 had a primary diagnosis of A1ATD(PI*ZZ phenotype and PAS+ globules),8 had a PI*MZ phenotype alone,14 had PAS+ alone,and 10 had both the PI*MZ phenotype and PAS+. Median follow-up time was 3.4(25th,75 th percentiles:1,5.2) years. The overall rate of hepatocellular carcinoma in all subjects was 29%(n = 199). In the A1 ATD group,the incidence rate of HCC was 8.5% compared to 31% in the group of patients with other causes of cirrhosis(P = 0.001). Vehicle Dynamics with Additional Entry Lane of a Roundabout

This research is dedicated to the study of dynamics of vehicles moving through the additional entry lanes of a roundabout. Using the data created on the base of that model the constant coefficients of the sixth-degree polynomial function, previously proposed by us, have been found. In result of this there were obtained two analytic equations of vehicles’ movement, where one of them presented with the positive sign of the leading coefficient and the other one negative. These two equations allowed to make comprehensive investigation of the delays and the kinematics of the vehicles moving by such kinds of lanes. Based on the appropriate calculations it is determined vehicles’ consequent delays, velocities, accelerations and decelerations. Analyzing the delays, the dynamics of accelerations and decelerations, it is done an assumption that if the leading coefficient of algorithm of movement of vehicles is negative than it will lead to larger deceleration and consequently higher environmental pollutions and higher noises.

Wind effects on the migration routes of trans-Saharan soaring raptors： geographical, seasonal, and interspecific variation

Wind is among the most important environmental factors shaping birds＇ migration patterns. Birds must deal with the displacement caused by crosswinds and their behavior can vary according to different factors such as flight mode, migratory season, experience, and distance to goal areas. Here we analyze the relationship between wind and migratory movements of three raptor species which migrate by soaring-gliding flight： Egyptian vulture Neophron percnopterus, booted eagle Aquila pennata, and short-toed snake eagle Circaetus gallicus. We analyzed daily migratory segments （i.e., the path joining consecutive roosting locations） using data recorded by GPS satellite telemetry. Daily movements of Egyptian vultures and booted eagles were significantly affected by tailwinds during both autumn and spring migrations. In contrast, daily movements of short-toed eagles were only significantly affected by tailwinds during autumn migration. The effect of crosswinds was signifi- cant in all cases. Interestingly, Egyptian vultures and booted eagles showed latitudinal differences in their behavior： both species compensated more frequently at the onset of autumn migration and, at the end of the season when reaching their wintering areas, the proportion of drift segments was higher. In contrast, there was a higher drift at the onset of spring migration and a higher compensation at the end. Our results highlight the effect of wind patterns on the migratory routes of soaring raptors, with different outcomes in relation to species, season, and latitude, ultimately shaping the loop migration patterns that current tracking techniques are showing to be widespread in many long distance migrants.

Can microcalcifications' characteristics predict the risk of breast cancer metastasis to bone?

Aim: To correlate the microcalcifications' characteristics, such as morphology and elemental compositions, with the occurrence of bone metastatic lesions at 5 years from diagnosis. Methods: In this retrospective study, we enrolled 70 patients from which we collected one breast biopsy each. From each biopsy, paraffin serial sections were obtained to perform histological classifications, immunohistochemical analyses and Energy Dispersive X-ray evaluation. Results: Microcalcifications analysis showed a significant association between the presence of calcium crystals made of magnesium substituted hydroxyapatite and the development of bone metastasis from 5 years from diagnosis. No significant association was observed by evaluation the morphological appearance of microcalcifications. Immunohistochemical analysis displayed a significant association between the expression of bone morphogenetic proteins 2 and pentraxin-3, two osteoblast induction factors, and the formation of bone metastatic lesions. Conclusion: Results here reported highlighted the possible use of breast microcalcifications as a negative prognostic marker of bone metastatic diseases. In particular, the association between elemental composition of breast microcalcifications and the formation of bone lesions can lay the foundation for the development of new in vivo diagnostic tools based on the analysis of microcalcifications and capable to predict the formation of bone metastasis.
