Primary lung graft dysfunction could significantly attribute to ischemia-reperfusion lung injury(IRLI)during transplantation surgery.β2-adrenergic agonists were one of the bronchodilators that had been well-established in the management of asthma and chronic obstructive pulmonary disease(COPD)with anti-inflammatory *** applying the model of isolated rat lung,we evaluated the efficacy of short-actingβ2-agonist inhalation to ameliorate ischemia-reperfusion *** experiment protocol was 180 min of global ischemia and then reperfusion for 60 *** theβ2-agonist inhalation group,aerosolized albuterol was administrated prior ischemia *** weight ratios of wet to dry lung and microvascular permeability were characterized in the IRLI *** contrast,pre-inhaledβ2-agonist significantly mitigated the severity of pulmonary *** lavage from theβ2-agonist group presented decreased leukocyte counts and cytokines production,including interleukin-1β(IL-1β),tumor necrosis factor-α(TNF-α),and macrophage inflammatory protein 2(MIP-2).Devastating oxidative stress was widely recognized during the ischemia-reperfusion process,whileβ2-agonist pretreatment revealed subsided H2O2,myeloperoxidase(MPO),and the cleavage of *** blotting from lung homogenates identified the blockade of NF-κB and MAPK activation in theβ2-agonist inhalation ***,there was no specific pharmacotherapy in IRLI *** results elucidated the protective effect ofβ2-agonist bronchodilator against ischemia-reperfusion induced oxidative stress,inflammation reaction,and pulmonary edema.

Testosterone replacement therapy has benefits for aging men and those with hypogonadism. However, the effects of exogenous testosterone on Leydig cells are still unclear and need to be clarified. In this report, we demonstrate that testosterone supplementation can reduce oxidative damage in Leydig cells. The TM3 Leydig cell line was used as an in vitrocell model in this study. Cytoprotective effects were identified with 100-nmol l-1 testosterone treatment, but cytotoxic effects were found with ≥ 500-nmol l-1 testosterone supplementation. Significantly reduced reactive oxygen species （ROS） generation, lipid peroxide contents and hypoxia induction factor （HIF）-lα stabilization and activation were found with lO0-nmol 1-1 testosterone treatment. There was a 1.72-fold increase in ROS generation in the 500-nmol l-1 compared to the lO0-nmol l-1 testosterone treatment. A 1.58-fold increase in steroidogenic acute regulatory protein （STAR） expression was found in 50-nmol l-1 testosterone-treated cells （P〈0.01）. Chemically induced hypoxia was attenuated by testosterone supplementation. Leydig cells treated with low-dose testosterone supplementation showed cytoprotection by decreasing ROS and lipid peroxides, increasing StAR expression and relieving hypoxia stress as demonstrated by HIF-1α stabilization. Increased oxidative damage was found with ≥ 500-nmol l-1 testosterone manipulation. The mechanism governing the differential dose effects of testosterone on Leydig cells needs further investigation in order to shed light on testosterone replacement therapy.

Platelet-rich plasma(PRP)can stimulate the proliferation of stem cells and have a positive effect on tissue *** many commercialized PRP preparation kits are already on the market,first-line clinical workers are still not satisfied with most of the PRP *** work of commercial PRP kits is based on the density of blood ***,the blood elements are very close in density which makes the separation ***,the mentioned commercialized kits are generally contaminated by leucocytes and *** this study,a home-designed PRP device was developed to use a separation membrane with adequate cut-off pore size of 5μm,3μm and 2μm for the groups of H5M,H3M,and H2M,respectively,to be placed in the middle of the centrifuge *** home-designed H2M showed a very promising results regardless of the final volume(1.82±0.09 ml),platelet yield(8.39±0.44%),Red Blood Cells(0%),White Blood Cells(0%),and Relative Concentration of Platelet Increment value(225.09%).Further,it showed a good result in cell viability and cytotoxicity and confirmed a good multilineage *** concentration in PRP prepared by group H2M was relatively stable and far above *** the fibrin fibers were linked together as bridging strands or strings and turned into an inter-connected porous structure for nutrients transportation and regenerative cell *** believe that the home-designed group H2M should have a great potential to develop into the final product to meet the requirements of first-line clinical workers.

The high incidence of gastric cancer (GC) and its consequent mortality rate severely threaten human health. GC is frequently not diagnosed until a relatively advanced stage. Surgery is the only potentially curative treatment. Thus, early screening and diagnosis are critical for improving prognoses in patients with GC. Gastroscopy with biopsy is an appropriate method capable of aiding the diagnosis of specific early GC tumor types; however, the stress caused by this method together with it being excessively expensive makes it difficult to use it as a routine method for screening for GC on a population basis. The currently used tumor marker assays for detecting GC are simple and rapid, but their use is limited by their low sensitivity and specificity. In recent years, several markers have been identiﬁed and tested for their clinical relevance in the management of GC. Here, we review the serum-based tumor markers for GC and their clinical significance, focusing on discoveries from microarray/proteomics research. We also review tissue-based GC tumor markers and their clinical application, focusing on discoveries from immunohistochemical research. This review provides a brief description of various tumor markers for the purposes of diagnosis, prognosis and therapeutics, and we include markers already in clinical practice and various forthcoming biomarkers.

Many technologies have been developed for breast reconstruction after *** the technologies achieved promising success in clinical,there are still many shortages hanging over and trouble the *** engineering technology was introduced to plastic surgery that gave a light to lumpectomy patients in breast *** unexpected absorption rate,resulting from limited vascularization and low cell survival rate,is a major factor that leads to unsatisfactory results for the previous studies in our *** the study,the laminin-modified alginate synthesized by a new method of low concertation of sodium periodate would be mixed with ADSCs and Rg1 in the medium;and then sprayed into a calcium chloride(CaCl2)solution to prepare into microsphere(abbreviated as ADSC-G-LAMS)by bio-electrospray with a power syringe for the mass production and smaller bead *** developed ADSC-G-LAMS microspheres had the diameter of 232±42μ***-release of the Rg1 retained its biological ***-1,live/dead staining,and chromosome aberration assay were evaluated to confirm the safety of the *** in vivo study,ADSC-G-LAMS microspheres combined with autologous adipocytes were transplanted into the dorsum of rats by subcutaneous *** efficacy was investigated by H&E and immunofluorescence *** results showed that the bioactive ADSC-G-LAMS microspheres could integrate well into the host adipose tissue with an adequate rate of angiogenesis by constantly releasing Rg1 to enhance the ADSC or adipocyte survival rate to join tissue growth and repair with adipogenesis for breast reconstruction after lumpectomy.

AIM: To investigate the differentiation status and key factors to facilitate hepatic differentiation of human bone-marrow-derived mesenchymal stem cells (MSCs). METHODS: Human MSCs derived from bone marrow were induced into hepatocyte-like cells following a previously published protocol. The differentiation status of the hepatocyte-like cells was compared with various human hepatoma cell lines. Overexpression of hepatocyte nuclear factor (HNF)-4α was mediated by adenovirus infection of these hepatocyte-like cells. The expression of interesting genes was then examined by either re-verse transcription-polymerase chain reaction (RT-PCR) or real-time RT-PCR methods. RESULTS: Our results demonstrated that the differentiation status of hepatocyte-like cells induced from human MSCs was relatively similar to poorly differentiated human hepatoma cell lines. Interestingly, the HNF-4 isoform in induced MSCs and poorly differentiated human hepatoma cell lines was identified as HNF4γ instead of HNF-4α. Overexpression of HNF-4α in induced MSCs significantly enhanced the expression level of hepatic-specific genes, liver-enriched transcription factors, and cytochrome P450 (P450) genes. CONCLUSION: Overexpression of HNF-4α improves the hepatic differentiation of human MSCs from bone marrow and is a simple way of providing better cell sources for clinical applications.

Cluster of differentiation 74 （CD74） performs multiple roles in B cells, T cells, and antigen-presenting cells within the immune system; it also participates in ma-jor histocompatibility complex class Ⅱ-restricted an-tigen presentation and inflammation. Recently, a role for CD74 in carcinogenesis has been described. CD74 promotes cell proliferation and motility and prevents cell death in a macrophage migration inhibitory factor-dependent manner. Its roles as an accessory signal receptor on the cell surface and the ability to interact with other signaling molecules make CD74 an attrac-tive therapeutic target for the treatment of cancer. This review focuses on the original role of CD74 in the immune system and its emerging tumor-related func-tions. First, the structure of CD74 will be summarized. Second, the current understandings about the expres-sion, cellular localization, molecular mechanisms and signaling pathways of CD74 in immunity and cancer will be reviewed. Third, the examples that suggest CD74 is a promising molecular therapeutic target are reviewed and discussed. Although the safety and ef-fcacy of CD74-targeted strategies are under develop-ment, deeply understanding of the regulation of CD74 will hold promise for the use of CD74 as a therapeutic target and may develop the CD74-targeted therapeutic agents such as neutralized antibody and compounds.
