Immunotherapy of rat glioma without accumulation of CD4^+CD25^+FOXP3^+ regulatory T cells

Immunotherapy may be used for the treatment of glioblastoma multiforme; however, the induced immune response is inadequate when either T cells or dendritic cells are used alone. In this study we established a novel vaccine procedure in rats, using dendritic cells pulsed with C6 tumor cell lysates in combination with adoptive transfer of T lymphocytes from syngenic donors. On day 21 after tumor inoculation, all the rats were sacrificed, the brains were harvested for calculation of glioma volume, cytolytic T lymphocyte responses were measured by cytotoxic assay, and the frequency of regulatory T lymphocytes （CD4＋CD25~FOXP3~） in the peripheral blood was investigated by flow cytometric analysis. The survival rate of rats bearing C6 glioma was observed. Results showed that the co-immunization strategy had significant anti-tumor potential against the pre-established C6 glioma, and induced a strong cytolytic T lymphocyte response in rats. The frequency of peripheral blood CD4＊CD25＊FOXP3＊ regulatory T lymphocytes was significantly decreased following the combination therapy, and the rats survived for a longer period. Experimental findings indicate that the combined immunotherapy of glioma cell lysate-pulsed dendritic cell vaccination following adoptive transfer of T cells can effectively inhibit the growth of gliomas in rats, boost anti-tumor immunity and produce a sustained immune response while avoiding the accumulation of CD4＋CD25＋FOXP3＋ regulatory r lymphocytes.

AIM： To retrospectively analyze the pathogenesis, clinical characteristics, treatment and prognostic characteristics in patients with traumatic basal ganglia hematomas （TBGH）. METHODS： A retrospective analysis of the clinical data was performed in 40 patients with TBGH who were selected from 1 250 patients with closed brain injury, who admitted to the Department of Neurosurgery of Shangqiu First People＇s Hospital from January 1990 to January 2004. The pathogenesis, clinical characteristics and signs, results of radiological examination, treatment and prognostic characteristics were analyzed. The patients all had definite history of brain injury, manifested by neurological functional disturbance to different extent after brain injury, and basal ganglia hemorrhage was identified by CT after brain injury, and hemorrhagic volume were more than or equal to 2 mL. Totally 34 males and 6 females were enrolled, aged 16-72 years and 28 cases of them were younger than 40 years old. The prognosis of the patients was evaluated with Glasgow outcome scale （GOS） at 6 months after injury, and GOS scoring standard was 1-5 points （1 for dead; 2 for vegetative survival, long-term coma, manifestations of decorticate rigidity or decerebrate rigidity; 3 for severely disabled, should be look after by others; 4 for moderately disabled, be able in self-care; 5 for good recovery, adults can work and study）. RESULTS： The enrolled cases accounted for 3.20% of the 1250 patients with closed brain injury admitted at the same period. ① The causes of injury included traffic accident in 36 cases, fall in 2 cases, and assault in 2 cases. ②At admission, the Glasgow coma scale （GCS） scores were as follow： 13-15 scores （mild） in 10 cases, 9-12 scores （moderate） in 20 cases, and 3-8 scores （severe） in 10 cases. Hemiplegia presented in 37 cases, aphasia in 20 cases, conscious disturbance in 10 cases, unilateral mydriasis in 6 cases, and decerebrate rigidity in 2 cases. ③ TBGH was detected by CT within 30 m

In this work,the forms of potassium and chlorine from oxy-fuel co-combustion of lignite coal and corn stover under atmospheric pressure were *** order to check transitional stage,the feedstocks were combusted stepwise,first by pyrolysis to form coke under N2 environment and later by coke combustion into the ash at 850℃ in O_(2)/CO_(2) *** results show that an increase in blend ratio from 15%to 40%results in an increase in water-soluble potassium in the feedstock and the ashes from 0.15%to 0.4%and 0.015%to 0.038%in weight *** water-soluble potassium is present mainly as KCl and *** ammonium acetate soluble potassium,a similar trend to water-soluble potassium is presented but with a much lower content of *** bound potassium in the fuel matrix exists,likely in the form of AlKSi_(2)O_(6).Chlorides are present mainly in the form of KCl which is the dominant water-soluble compound.

OBJECTIVE： To explain the mechanisms of tuhe synthesis, secretion and regulation of brain natriuretic peptide （BNP）, and analyze its role in central nervous system diseases. DATA SOURCES： An online search of Pubmed was undertaken to identify articles related to BNP published in English from January 1990 to February 2007 by using the key words of ＂brain natriuretic pepfide （BNP）, central nervous system, subarachnoid hemorrhage （SAH）, brain edema, epilepsy＂. Other articles were searched in China Hospital Knowledge Database （CHKD） by concrete name of journals and title of articles. STUDY SELECTION： The collected articles were primarily screened, those about BNP and its association with central nervous system diseases were selected, whereas the obviously irrelative ones excluded, and the full-texts of the other literatures were searched manually. DATA EXTRACTION： Totally 96 articles were collected, 40 of them were enrolled, and the other 56 were excluded due to repetitive studies or reviews. DATA SYNTHESIS： At present, there are penetrating studies on BNP in the preclinical medicine and clinical medicine of cerebrovascular and cardiovascular diseases, and the investigative outcomes have been gradually applied in clinical practice, and satisfactory results have been obtained. However, the application of BNP in diagnosing and treating central nervous system diseases is still at the experimental phase without - outstanding outcomes, thus the preclinical and clinical studies should be enhanced. CONCLUSION： As a kind of central medium or modulator, BNP plays a certain role in the occurrence, development and termination of central nervous system diseases, the BNP level in serum has certain changing law in SAH, brain edema, epilepsy, etc., but the specific mechanisms are unclear.

Anti-wear beams installed on water walls of circulating fluidized bed （CFB） boilers are one of the most effective ways to protect against water-wall erosion. Beam effects from, for example, beam size and superficial gas velocity were investigated on gas-solid hydrodynamics in a CFB test rig using CFD simulations and experimental methods. The downward flow of the wall layer solids is observed to be disrupted by the beam but is then restored some distance further downstream. When falling solids from the wall layer hit the anti-wear beam, the velocity of the falling solids decreases rapidly. A fraction of the solids accumulates on the beam. Below the beams, the falling solids have reduced velocities but upward-moving solids were observed on the wall. The effect of the beam increases with width and superficial gas velocity. Wear occurs mainly above the beam and its variation with width is different above to below the beam. There is an optimum width that, when combined with beam height, results in less erosion.

Pyrolysis Mechanism of Hemicellulose Monosaccharides in Different Catalytic Processes

The pyrolysis behaviors of four different hemicellulose monosaccharides, namely, two pentoses（xylose and arabinose） and two hexoses（mannose and galactose） catalyzed by HZSM-5 were investigated. The effects of two different processes by which the catalyst comes into contact with the substrate, namely, mixed with monosaccha- ride（in-bed） or layered above monosaccharide（in situ）, were compared. Evolution characteristics of typical pyrolytic products（H20, CO2, acids, furans and aromatics） were achieved by thermogravimetry-Fourier transform infrared spectroscopy. The in-bed catalytic process significantly lowered the pyrolytic temperature and increased the produc- tion of furans and acids at a low temperature by enhancing dehydration, retro-aldol fragmentation and Grob fragmen- tation. During the in situ catalytic process, volatiles generated from monosaccharides passed through a catalyst bed and underwent further dehydration, decarboxylation, and decarbonylation, significantly lowering the yields of acids and furans. The yield of aromatics was enhanced, and the corresponding volatilization temperature was lowered, es- pecially under the in-bed catalytic conditions. Pentoses entered into the zeolite pores more easily than hexoses did because of their smaller molecular size; thus, the in-bed catalytic process drastically affected pentose pyrolysis.

Therapeutic potential of bone marrow-derived mononuclear cells for experimental cerebral ischemia in mice

Bone marrow mononuclear cells （BMMCs） can be directly harvested from the donor, allowing for easier application compared with bone marrow mesenchymal stem cells. The present study hypothesized that BMMC transplantation could ameliorate cerebral ischemia in a mouse model. BMMCs were double-labeled with PKH26 and 4＇, 6-diamidino-2-phenylindole （DAPI）, followed by infusion into mice via the tail vein to induce focal cerebral ischemia. At 14 days after transplantation, morphological and neurofunctional recovery were analyzed. PKH26 and DAPI double-positive BMMCs were detected in the cerebral hemisphere of all transplantation mice. Following BMMC administration, there was significant difference in neurofunctional recovery, but no significant difference in survival rates between BMMC-treated mice and other mice. These results demonstrate that transplanted BMMCs migrate to brain tissue and promote neurological function recovery in a mouse model of cerebral ischemia.
